        Characterization of a novel variant HMW‐glutenin gene from Elymus canadensis

        High molecular weight (HMW) glutenin subunits (GS) play a key role in the determination of end‐use quality of wheat and other cereal crops. In this study, we report the isolation and characterization of both promoter region and ORF of novel HMW‐GS allele 1St1.3 from a perennial Triticeae species,Elymus canadensis. The amino acid (AA) sequences of E. canadensis 1St1.3 were deduced as 434 aa. Its protein primary structure comprises a signal peptide with a conserved N‐terminal domain, a central repetitive domain and a C‐terminal domain. E. canadensis 1St 1.3 possesses several distinct characteristics which are different from those of wheat HMW‐GSs. The N‐terminal domains of E. canadensis 1St 1.3 resemble that of y‐type subunits, while their C‐terminal domains are more similar to x‐type subunits. The deletion of 85 bp fragment has been observed in promoter region of 1St 1.3, however which has not interrupted the expression of this gene. Our results indicate that 1St 1.3 is novel HMW‐GS variants which will be valuable for enhancing our understanding of structural differentiation and the evolutionary relationship among HMW‐GSs in Triticeae species.

      • CARC: A Reliable Routing Mechanism for Cognitive Ad Hoc Networks

        In cognitive radio Ad Hoc networks, the events of node failure and link failure maybe happen frequently, especially the primary users will access the authorized channel whenever possible. In order to maintain the connection between cognitive users and resume their communication as soon as possible when the current route is invalid, a reliable routing mechanism CARC is proposed, which employs primary and candidate route to communicate under differential cases. Meanwhile, how to choose candidate route, maintain or rebuild route are also elaborated. Numerical simulations show that the proposed CARC protocol has a higher performance compared with other typical protocols.

        Characterization of a novel ribose-5-phosphate isomerase B from Curtobacterium flaccumfaciens ZXL1 for D-allose production

        Enzymatic preparation of rare sugars as an alternative to traditional sweeteners is an effective strategy to achieve a low-calorie healthy diet. Ribose-5-phosphate isomerase B (RpiB) is a key enzyme in the non-oxidative branch of the catalytic pentose phosphate pathway. Here, we investigated the potential of Curtobacterium flaccumfaciens ZXL1 (C. flaccumfaciens ZXL1) derived RpiB (CfRpiB) in D-allose preparation. The optimal reaction conditions for recombinant CfRpiB were found experimentally to be pH 7.0, 55 °C, and no metal ions. The kinetic parameters Km, kcat, and catalytic efficiency kcat/Km were 320 mM, 4769 s−1, and 14.9 mM−1 s−1 respectively. The conversion of D-allulose by purified enzyme (1 g L−1 ) to D-allose was 13% within 1 h. In addition, homology modeling and molecular docking were used to predict the active site residues: Asp13, Asp14, Cys72, Gly73, Thr74, Gly77, Asn106, and Lys144.

        Inactivation of epithelial sodium ion channel molecules serves as effective diagnostic biomarkers in clear cell renal cell carcinoma

        Background Non-voltage-gated sodium channel, also known as the epithelial sodium channel (ENaC), formed by heteromeric complexes consisting of SCNN1A, SCNN1B, and SCNN1G, is responsible for maintaining sodium ion and body fluid homeostasis in epithelial cells. However, no systematic study of SCNN1 family members has been conducted in renal clear cell carcinoma (ccRCC) to date. Objective To investigate the abnormal expression of SCNN1 family in ccRCC and its potential correlation with clinical parameters. Methods The transcription and protein expression levels of SCNN1 family members in ccRCC were analyzed based on the TCGA database, and were confirmed by quantitative RT-PCR and immunohistochemical staining assays, respectively. The area under curve (AUC) was used to evaluate the diagnostic value of SCNN1 family members for ccRCC patients. Results The mRNA and protein expression of SCNN1 family members was significantly downregulated in ccRCC compared with normal kidney tissues, which might be due to DNA hypermethylation in the promoter region. It is worth noting that the AUC of SCNN1A, SCNN1B, and SCNN1G were 0.965, 0.979, and 0.988 based on the TCGA database (p < 0.0001), respectively. The diagnostic value was even higher when combing these three members together (AUC = 0.997, p < 0.0001). Intriguingly, the mRNA level of SCNN1A was significantly lower in females compared with males, while SCNN1B and SCNN1G were increased with the progression of ccRCC and remarkably associated with a worse outcome for patients. Conclusion The aberrantly decrease of SCNN1 family members might serve as valuable biomarkers for the diagnosis of ccRCC.

      • SNS상의 온라인 구전정보 특성 및 브랜드 이미지가 구매의도에 미치는 영향 - 중국 SNS상의 한국 화장품 브랜드를 대상으로 -

        온라인 구전이 소비자의 구매의도에 영향을 미치는 것은 많은 선행연구에서 이미 검증되었다. 특히, 온라인 구전이 활발하게 일어나는 원인 중 하나인 “소셜 네트워크 서비스(social network service)”는 정보를 공유하고자 하는 동기가 많은 부분을 차지하고 있다. 소비자는 온라인상에서 정보를 획득하고, 정보를 확산시키며, 정보를 생산하는 범주에서 SNS를 활용한다. 이러한 이유에서 SNS와 온라인 구전은 밀접한 관련이 있어서 이용자들 간에 정보 공유는 구매 행동에 많은 영향을 미칠 것으로 예상한다. 본 연구는 브랜드 이미지가 매개 변수로서 SNS상에 온라인 구전 정보의 3가지 특성(방향성, 수량, 내용 품질)이 소비자 구매의도에 미치는 영향을 연구하였으며, SNS상에 온라인 구전 정보의 특성, 브랜드 이미지, 소비자 구매의도 간의 관계를 검증하고자 한다. 좀 더 구체적으로 살펴보면, 본 연구에서는 SNS상에 온라인 구전 정보의 특성, 브랜드 이미지, 구매의도 간에 관계의 모델을 수립하고 중국 SNS상에 한국 화장품 브랜드의 온라인 구전 정보가 연구대상으로 중국 SNS 이용자를 조사 대상으로 하였다. 그 결과 SNS상에 온라인 구전 정보의 특성이 소비자 구매의도에 대하여 유의미한 영향을 미쳤고, SNS상에 온라인 구전 정보의 특성은 브랜드 이미지에 대하여 부분적으로만 영향을 미쳤다. 마지막으로 브랜드 이미지는 SNS상에 온라인 구전 정보의 특성과 소비자 구매의도 사이의 매개효과가 있는 것으로 나타났다. The influence of Word of Mouth on consumer purchase Intention has been unanimously approved by preceding research. Depending on the development of online economy, communication of word of mouth developed from offline to the online has been important information for consumers to reference when shopping. At the same time, more and more brand products which before were sold in offline channels is to sell through online channel, the affect of brand image on purchase intention should not be underestimated. Especially, SNS(social network service) which the reasons for the rapid development of IWOM, the information dissemination and acceptance is the main reason for use. Consumers make use of SNS to obtain information, disseminate information and product information in online. For this reason, SNS and IWOM are closely linked. So we predict that the behavior of intelligence sharing have a significant impact on the purchasing behavior among users. The study uses brand image as mediator variables to the model of the effect that IWOM in SNS puts on consumer purchase intention, and explore the relationship among the three variables: IWOM in SNS, brand image and consumer purchase intention. The study construct a relationship model of IWOM in SNS, brand image and consumer purchase intention, choice commodity cosmetics IWOM in China`s SNS as the object of study. We choice the China`s SNS users as the investigation object. The study used SPSS and AMOS to make descriptive statistics analysis, evaluation of the measurement model, confirmatory factor analysis, path analysis, mediator effect analysis. The conclusions are as follows: Firstly, IWOM in SNS has a significant positive impact on consumer purchase intention. Secondly, IWOM in SNS has a partly positive impact on brand image. Thirdly, brand image has a mediator effect in the effect that the direction of IWOM and content quality of IWOM and put on consumer purchase intention. According to the conclusions, the study put forward corresponding suggesttons: positively analyse the reason of negative word of mouth appearance and to eliminate, Implement development strategies brand image, train brand image that suitable for online marketing.

This article mainly from the three aspects of the topic spreads out discuss, first studied the travel service quality management content, from the hardware management and software management combination of two levels is discussed, then on the basis of the travel agency establishing quality management system, from the four aspects of the process, the last around travel cooperative quality management approach to such problems in many aspects. In this paper, starting from theoretical analysis, through a systematic study and analysis for the travel service quality management to provide some reference to the study, at the same time as the consumers’ choice of a travel agency to provide efficient quality judgment.

      • Outcome after Simultaneous Resection of Gastric Primary Tumour and Synchronous Liver Metastases: Survival Analysis of a Single-center Experience in China

        Background: The optimal surgical strategy for the treatment of synchronous resectable gastric cancer liver metastases remains controversial. The aims of this study were to analyze the outcome and overall survival of patients presenting with gastric cancer and liver metastases treated by simultaneous resection. Materials and Methods: Between January 1990 and June 2009, 35 patients diagnosed with synchronous hepatic metastases from gastric carcinoma received simultaneous resection of both primary gastric cancer and synchronous hepatic metastases. The clinicopathologic features and the surgical results of the 35 patients were retrospectively analyzed. Results: The 5-year overall survival rate after surgery was 14.3%. Five patients survived for more than 5 years after surgery. No mortality has occurred within 30 days after resection, although two patients (5.7%) developed complications during the peri-operative course. Univariate analysis revealed that patients with the presence of lymphovascular invasion of the primary tumor, bilateral liver metastasis and multiple liver metastases suffered poor survival. Lymphovascular invasion by the primary lesion and multiple liver metastases were significant prognostic factors that influenced survival in the multivariate analysis (p=0.02, p=0.001, respectively). Conclusions: The presence of lymphovascular invasion of the primary tumor and multiple liver metastases are significant prognostic determinants of survival. Gastric cancer patients without lymphovascular invasion and with a solitary synchronous liver metastasis may be good candidates for hepatic resection. Simultaneous resection of both primary gastric cancer and synchronous hepatic metastases may effectively prolong survival in strictly selected patients.

        Research on influence of harmonic wear wheel on wheel/rail contact geometry of high-speed train

        Harmonic wear is one of the common wear phenomena of high-speed train wheels. The change of wheel/rail rolling contact geometry leads to unstable characteristics of wheel/rail contact, which directly affects the quality and safety of high-speed train operation. Because wheelset lateral displacement directly affected the wheel/rail contact geometry, in this paper, the UM software was used to establish a high-speed train vehicle-track coupled dynamics simulation model of the CRH2 (China Railway High-speed 2) head car. Lateral displacement of harmonic wear wheel was calculated under different wear condition to analyze the influence of harmonic wear wheels on wheelset lateral displacement. Then according to macroscopic dimension change of the wheel profile from lateral and radial directions of high-speed train induced by harmonic wear wheels, wheel profiles under different wear condition were selected. And wheel/rail contact geometry under different harmonic wear stages was calculated to discuss the influence of harmonic wear wheel on wheel/rail contact geometry. The results show that harmonic order and wave depth of harmonic wear wheel have small influence on the lateral displacement of wheelset, but the influence of wave depth is greater than harmonic order. The average difference of lateral displacement between harmonic wear wheel and no harmonic wear wheel increases with the increase of wave depth and order. In a harmonic wave length, the maximum value of both equivalent conicity and contact angle appear in deepest wave depth. The greater the wave depth, the more obvious the fluctuations, and equivalent conicity changes are less obvious.

        BSA‑carbon dots a promising “off–on” fluorescence probe for detecting glyphosate residues in agricultural products

        The widespread and extensive use of glyphosate in agriculture has raised concerns about its potential impact on the quality and safety of agricultural products. Conventional detection methods require long analysis times, making them impractical for the rapid detection of large quantities of samples. Therefore, developing a fast and simple detection system for glyphosate pesticide residues is urgent. In this study, the development of a facile fluorescence probe synthesized using a simple one-pot hydrothermal method for the determination of glyphosate is an important step toward addressing the need for a fast and simple detection system. The present sensor was created using bovine serum albumin (BSA) as a precursor, and the sensor operates by producing an “off–on” fluorescent signal. The bovine albumin-derived BSA-CDs emitted light yellow fluorescence, but this fluorescence was quenched (or suppressed) by the presence of Cu2+ ions. However, the fluorescence can be restored by the presence of glyphosate, which interacts with the Cu2+ ions to form a complex and release the BSACDs from suppression. The functional groups in glyphosate can capture Cu2+ and break the BSA-CDs/Cu2+ combinatorial system. The BSA-CDs/Cu2+ fluorescence quenching system had good selectivity for glyphosate. The detection limit of the BSA-CD/Cu2+ fluorescence sensor was 0.05 μg/mL. This developed method was utilized to successfully detect glyphosate in Chinese wheat. The average recoveries ranged from 98.9 to 100.7%, with a relative standard deviation < 3.0%, showing good prospects for practical applicability.
