        Effect of temperature on the development, reproduction and survival of Tetramoera schistaceana (Lepidoptera: Tortricidae)

        Tetramoera schistaceana Snellen is an important stem borer of sugarcane, which causes severe damage to sug arcane in China. Temperature is a key factor affecting its development and reproduction. Development duration, reproduction and survival rate of T. schistaceana were determined at five different temperatures (20, 23, 26, 29 and 32 ◦ C) in this study. Results showed that temperature had significant effects on the development duration, survival rate and reproduction of T. schistaceana. The development rate of T. schistaceana for each stage increased linearly with increasing temperatures between 20 and 32 ◦ C. Egg hatching rate, larval pupation rate and pupal eclosion rate were the highest between 23 and 26 ◦ C. The lower thermal threshold (T 0 ) for egg, larva, pupa, preadult and whole generation were 9.98, 9.80, 9.47, 9.50 and 8.87 ◦ C, respectively and the thermal constant (K) for the development of the respective life cycle stages of were 89.29 for eggs, 400.00 for larvae, 151.52 for pupae, 625.00 for pre-adult and 666.67DD for whole generation, respectively. The average fecundity per female was also the highest between 23 and 26 ◦ C. Therefore the optimum temperature range for the development and reproduction of T. schistaceana was between 23 and 26 ◦ C.

      • Preparation and Antitumor Activity of a Tamibarotene-Furoxan Derivative

        Multi-target drug design, in which drugs are designed as single molecules to simultaneously modulate multiple physiological targets, is an important strategy in the field of drug discovery. QT-011, a tamibarotene-furoxan derivative, was here prepared and proposed to exert synergistic effects on antileukemia by releasing nitric oxide and tamibarotene. Compared with tamibarotene itself, QT-011 displayed stronger antiproliferative effects on U937 and HL-60 cells and was more effective evaluated in a nude mice U937 xenograft model in vivo. In addition, QT-011 could release nitric oxide which might contribute to the antiproliferative activity. Autodocking assays showed that QT-011 fits well with the hydrophobic pocket of retinoic acid receptors. Taken together, these results suggest that QT-011 might be a highly effective derivative of tamibarotene and a potential candidate compound as antileukemia agent.

      • Knowledge of Human Papillomavirus (HPV) Infection, Cervical Cancer, and HPV Vaccine and its Correlates among Medical Students in Southwest China: a Multi-center Cross-sectional Survey

        Background: Since cervical cancer can be prevented and controlled through human papillomavirus (HPV) vaccination, it is important to train health care providers and provide them with appropriate knowledge. This study aimed to understand the level of HPV related knowledge among medical students and correlates in Southwest China in order to address any potential gap in their knowledge base. Methods: We conducted a cross-sectional survey among medical students in six selected universities across Southwest China based on a pretested questionnaire regarding HPV infection, cervical cancer, and HPV vaccines. Results: We successfully surveyed 1, 878 medical students, of whom 32.1% were males and 67.9% were females. Their mean age was 20.8 (standard deviation: 1.3) years. 91.8% of them were ethnic Han Chinese, and 43.8% were students in clinical medicine. While 76.5% had heard of HPV only 48.8% knew that the prevention of cervical cancer was possible through HPV vaccination. Only 42.9% of the male and 49.2% of the female students correctly answered over 10 out of 22 questions on HPV related knowledge. Female students appeared to know more about HPV and HPV vaccination (OR: 1.39; 95% CI: 1.11-1.75). In addition, the student knowledge improved with the grade (p<0.001). University courses were the most selected source of knowledge about HPV vaccination (66.4%). 83.6% of males and 91.4% of females were willing to have more HPV related education by experts (p<0.001). Only 10.1% of the students acknowledged that people had asked for their advice regarding HPV vaccination. Conclusions: Our survey indicates that medical students from Southwest China have poor knowledge of HPV and HPV vaccination, but are willing to receive more relevant information. Targeted education should be incorporated into school courses to inform these future health care providers and ensure success of programs for cervical cancer control and prevention.

        Effects of temperature on the development and reproduction of sugarcane top borer Scirpophaga excerptalis (Lepidoptera: Pyralidae)

        To determine the effects of temperature on the development and reproduction of top borer Scirpophaga excerptalis Walker (Lepidoptera: Pyralidae), the durations of different developmental stages and reproductive capacity of S. excerptalis were measured at 20, 23, 26, 29 and 32℃. The results showed that the average durations of various developmental stages of S. excerptalis shortened with increasing temperatures from 20 to 32℃. The durations to complete one life cycle were 113.17 days at 20℃, and 39.50 days at 32℃. The developmental rate of each stage and generation was positively correlated with the temperature, which was consistent with the Logistic regression model. The temperature thresholds for egg, larval, pupal and preoviposition stages and the total cycle were 13.73, 14.73, 13.91, 13.66 and 14.10℃, respectively. The effective accumulative temperature was 112.62, 370.01, 188.17, 23.82 and 718.07 degree-days (DD), respectively. The adult longevity shortened with increasing temperatures, and the highest number of eggs laid per female was 204.74 eggs at 29℃. The survival rates of eggs, larvae and the whole generation were the highest at 26℃, which were 87.25%, 56.67% and 37.21%, respectively while the highest survival rate of the pupa was 76.69% at 29℃. There was no significant difference for the total cycle at 23-29℃. The results indicated that temperature was the key factor affecting the development and reproduction of S. excerptalis, and the optimum temperature for development and reproduction ranged from 23 to 29℃.

        Characterization, Cloning, and Heterologous Expression of a Subtilisin-Like Serine Protease Gene VlPr1 from Verticillium lecanii

        The entomopathogenic fungus Verticillium lecanii is a wellknown biocontrol agent. V. lecanii produces subtilisin-like serine protease (Pr1), which is important in the biological control activity of some insect pests by degrading insect cuticles. In this study, a subtilisin-like serine protease gene VlPr1 was cloned from the fungus and the VlPr1 protein was expressed in Escherichia coli. The VlPr1 gene contains an open reading frame (ORF) interrupted by three short introns, and encodes a protein of 379 amino acids. Protein sequence analysis revealed high homology with subtilisin serine proteases. The molecular mass of the protease was 38 kDa, and the serine protease exhibited its maximal activity at 40°C and pH 9.0. Protease activity was also affected by Mg2+ and Ca2+ concentration. The protease showed inhibitory activity against several plant pathogens, especially towards Fusarium moniliforme.
