        Molecular Characterization and Physico-Chemical Analysis of a New Giant Embryo Mutant Allele (ge(t)) in Rice (Oryza sativa L.)

        The rice embryo is rich in lipid and protein bodies, bioactive chemicals such as dietary fiber, phytic acids, vitamin B and E, and gamma aminobutyric acid (GABA) than the endosperm. In this paper, we report a new giant embryo mutant, ge(t), induced from somaclonal variation derived by anther culture in rice. Sequence analysis of Cytochrome P450 of the ge(t) mutant revealed that ge(t) is a new allele of the GE gene with a single point mutation with substitution of amino acid, W395 to L395. The weight of the ge(t) mutant embryo was 3.7 times higher than normal embryo. Tocopherol and mineral content were also higher than the previously reported giant embryo rice variety, Keunnun. These results indicated that this new giant embryo rice (ge(t)) offers a promising source of genetic material in improving nutritional quality of rice especially tocopherol, essential minerals, and GABA.

        KOPIA 필리핀 센터의 벼 종자 생산 및 보급 시범마을사업 효과분석

        The KOPIA Philippines Center implemented a model village project on production and dissemination of rice seed from 2015 to 2017 for three years and subsequently operated a follow-up project from 2018 to 2019. In Iloilo and Bohol provinces, the top grain producers in the Philippines, the project supported production technologies tailored to the local environments, as well as produced and disseminated seeds of high-yield cultivars including ‘Milyang 23’, ‘NSIC Rc 222’, and ‘MS 11’. In Iloilo with limited irrigation channels and equipment, the project overhauled their water pumps and irrigation facilities. It also employed a set of marketing strategies for ‘MS 11’ jointly developed by the Rural Development Administration (RDA) and International Rice Research Institute (IRRI) which are highquality japonica type cultivars adapted to tropical Asia. By positioning them as a premium brand, the products are targeted at Korean, Japanese and Chinese tourists in the country. As a result, the participating farm households responded in a survey that they are satisfied with and willing to continue the project, with scales ranging from 5.4 to 6.7 points, which are relatively high. The farmers experienced increases in their incomes ranging from 20.6% to 47.3%, which averages 34.8%. According to the analysis of management performance of the model village project on multiplication and dissemination of quality rice seed in Iloilo province and Bohol province of the Philippines, the project provided needed technical cooperation and infrastructure in improving local rice productivity, which clearly shows the effectiveness of this Official Development Assistant (ODA) project. The participating farm households gave positive responses on the effectiveness of the model village project. If there would be another round of the same project in the future, they are willing to take part in again. KOPIA 필리핀 센터는 2015년부터 2019년까지 벼 우량종자증식과 보급 시범사업을 위해 Iloilo 지역 시범마을에 양수기지원 및 관개수로 확충을 통하여 Sinibaan 50 ha와 Tabucan 65 ha 등 총 115 ha 면적의 관개시설과 ‘Milyang 23’ 등 다수성 품종과 재배기술을 지원하였다. Bohol 지역 시범마을은japonica 벼 품종 ‘MS 11’의 고순도 종자를 시범마을 농가에분양하고 재배기술을 지원하였으며 대량 재배를 통해 주변 시장과 연계하는 고급브랜드화 사업을 추진하였다. Iloilo와 Bohol 2개 지역에서의 필리핀 벼 종자증식 및 보급시범마을사업의 성과분석 결과, 본 사업은 마을에서 요구하는벼 재배 맞춤형 기술지원과 적절한 기반시설 투입을 통해 다음과 같은 결과를 얻었다. 1. 2개 지역 시범마을사업 추진 결과 벼의 평균 생산량은25.6~29.7%가 증가하였다. 2. 시범마을사업 추진을 통해 2개 지역 평균 순소득은 2~3 배가 향상되었다. 3. 이중차분법(DID)을 이용하여 분석한 농가 소득은 Iloilo 주 건기 22.2%, 우기 0.2%, Bohol 주 건기 47.3%, 우기48.5%로 향상되었다. 4. 시범마을사업 참여 농가의 만족도와 계속 참여 의향은 7 점 만점에 5.4~6.7 범위로 향후 동일한 사업이 진행된다면 다시 참여하겠다는 농가 비율이 상대적으로 높은 경향을 보였다.

        Molecular re-confirmation and floral characteristics of drooping leaf (DL) mutants generated by insertional mutagenesis in rice

        Transposon tagging and insertional mutagenesis provide one of the most powerful tools in gene function studies. Here, we report a comparison between two novel drooping leaf (DL)mutants from transposon and T-DNA insertion lines of rice. DL is distinct from well-known ABC genes and a member of the YABBY gene family, and it is closely related to the CRABS CLAW (CRC) gene of Arabidopsis thaliana. Based on phenotypic analysis, DL regulated midrib formation by promoting cell proliferation in the central region of rice leaf and was necessary for the specification of carpel identity. We identified two DL mutants by screening the Ac/Ds and T-DNA insertional mutant pool of rice. Flanking sequence tag analysis indicated that both Ds and T-DNA segments were inserted in the promoter region at 3.4 kbs and 5.4 kb upstream, respectively,of the previously known OsYABBY domain. Interestingly, the progenies of DL lines of two different pools showed various degrees of leaf drooping and abnormal carpel formation. Flower structures revealed that there were more than two stigmas with normal stamens and pistils per panicle in the Ds-induced mutants. However, T-DNA induced mutant had extra stamens with staminoid carpels. These results indicate that the promoter region of DL plays an important function in regulating anther and carpel formation.
