        Exosomes from Tension Force-Applied Periodontal Ligament Cells Promote Mesenchymal Stem Cell Recruitment by Altering microRNA Profiles

        Background and Objectives: To investigate the role of exosomes from periodontal ligament cells (PDLCs) in bone marrow mesenchymal stem cell (BMSC) migration. Methods and Results: Human PDLCs were applied cyclic tension stretching. Exosomes were extracted from cultured PDLCs by ultracentrifugation, then characterized for their size, morphology and protein markers by NTA, TEM and western blotting. The process that PKH26-labeled exosomes taken up by BMSCs was assessed by confocal microscope. BMSC migration was examined by Transwell assay. Exosomes derived from PDLCs were identified. Cyclic tension stretch application on PDLCs can enhance the migration ability of BMSCs through exosomes. The exosomal miRNA expression profiles of unstretched and stretched PDLCs were tested by miRNA microarray. Four miRNAs (miR-4633-5p, miR-30c-5p, miR-371a-3p and let-7b-3p) were upregulated and six (miR-4689, miR-8485, miR-4655-3p, miR-4672, miR-3180-5p and miR-4476) were downregulated in the exosomes after stretching. Sixteen hub proteins were found in the miRNA-mRNA network. Gene Ontology and KEGG pathway analyses demonstrated that the target genes of differentially expressed exosomal miRNAs closely related to the PI3K pathway and vesicle transmission. Conclusions: The exosomes derived from cyclic tension-stretched PDLCs can promote the migration of BMSCs. Alternation of microRNA profiles provides a basis for further research on the regulatory function of the exosomal miRNAs of PDLCs during orthodontic tooth movement.

        Rosa roxburghii Tratt juice inhibits NF-κB and increases IL-2 to alleviates the Foxp3-mediated Tregs imbalance in the peripheral blood of arseniasis patients

        Arsenic can cause immune inflammation, which is the basis of arsenic-induced damage to multiple organs and systems. Forkhead box P3 (Foxp3)-labelled CD4+CD25+ regulatory T cells (Tregs) play an essential role in maintaining immune homeostasis. Nuclear factor-κb (NF-κB) and Interleukin-2 (IL-2) are critical regulators of Foxp3. Rosa roxburghii Tratt (RRT) is an edible medicinal plant with anti-inflammation effects. In this study, a control group (n = 41) and an arseniasis group (n = 209) were recruited, and screened subjects from the arseniasis patients for RRTJ (n = 46) or placebo (n = 43) to explore the possible mechanism by which RRT alleviates immune inflammation. The results indicated that RRTJ can inhibits NF-κB and increases IL-2, and alleviates the Foxp3-mediated Tregs imbalance in the peripheral blood of arseniasis patients. In summary, these findings suggest a novel intervention or therapeutic target for immune inflammation in arseniasis patients and provide new evidence that RRTJ inhibits immune inflammation.

        Landslide susceptibility mapping at Gongliu county, China using artificial neural network and weight of evidence models

        The aim of this study was to apply and to verify the use of artificial neural network (ANN) and weight of evidence (WoE) models to landslide susceptibility mapping in the Gongliu county, China, using a geographic information system (GIS). For this aim, in this study, a landslide inventory map was prepared using earlier reports and aerial photographs as well as by carrying out field surveys. A total of 163 landslides (70% out of 233 detected landslides) were randomly selected for model training, and the remaining 70 landslides (30%) were used for the model validation. Then, a total number of twelve landslide conditioning factors, such as slope angle, slope aspect, general curvature, plan curvature, profile curvature, altitude, distance to rivers, distance to roads, lithology, rainfall, normalized difference vegetation index (NDVI), and sediment transport index (STI), were used in the analysis. Landslide hazardous areas were analyzed and mapped using the landslide-occurrence factors by ANN and WoE models. Finally the output maps were validated using the area under the curve (AUC) method. The validation results showed that the ANN model with a success rate of 82.51% and predictive accuracy of 77.31% performs better than WoE (success rate, 79.82%; predictive accuracy, 74.59%) model. Overall, both models showed almost similar results. Therefore, the two landslide susceptibility maps obtained were successful and can be useful for preliminary general land use planning and hazard mitigation purpose.

        A method to characterize ink performance by line resistance measurement through modeling for direct pen writing

        The rational evaluation of carbon-based conductive ink performance is critical to both industrial production and applications. Herein, a model to evaluate writing performance of conductive ink by line resistance was proposed by investigating possible relations among different parameters and establishing relevant model to estimate ink writing performance. Bulk conductive inks were prepared and characterized to provide samples for model. To improve the precision of model, the impact of external factors including writing speed and angle was studied. Nonlinear regression and back propagation artificial neural network were employed to estimate line resistance, and cross check validation was conducted to prove robustness and precision of model. Most importantly, the investigation will open up a new path for the exploration of other carbon-based handwritten electronic devices.

        Determination Method for the Strength Model of a Jointed Rock Mass Based on the Geological Strength Index

        The mechanical characteristics of materials are the key to numerical calculation, thedetermination of material parameters of jointed rock mass is conundrum in academia. For thepurpose of more efficiently and accurately select the mechanical parameters of the jointedrock masses, in this paper, a new method is proposed which for constructing a numericalmodel of jointed rock masses, which based on the Geological Strength Index (GSI) and theprinciples of damage mechanics. With the help of basic concepts of damage mechanics, thejoint equivalent coefficient is proposed, establishing the relationship between GSI and jointspacing. The selection way for the mechanical parameters of a jointed rock masses is given incombination with the Hoek-Brown strength criterion. Then, a numerical calculation model isconstructed by utilizing 3DEC numerical simulation software to discuss the stability of theadjoining rock under the condition of different GSIs, which is successfully applied to theWangjialing coal mine project. The numerical results match quite closely with the fieldconditions, the maximum error is 18%, the minimum error is 5.6%, verifying the rationalityand accuracy of this way and providing a new way for the accurate numerical simulation ofjointed rock masses.

        Knee-driven many-objective sine-cosine algorithm

        When solving multi-objective optimization problems, the blindness of the evolution direction of the population gradually emerges with the increase in the number of objectives, and there are also problems of convergence and diversity that are difficult to balance. The many-objective optimization problem makes some classic multi-objective optimization algorithms face challenges due to the huge objective space. The sine cosine algorithm is a new type of natural simulation optimization algorithm, which uses the sine and cosine mathematical model to solve the optimization problem. In this paper, a knee-driven many-objective sine-cosine algorithm (MaSCA-KD) is proposed. First, the Latin hypercube population initialization strategy is used to generate the initial population, in order to ensure that the population is evenly distributed in the decision space. Secondly, special points in the population, such as nadir point and knee points, are adopted to increase selection pressure and guide population evolution. In the process of environmental selection, the diversity of the population is promoted through diversity criteria. Through the above strategies, the balance of population convergence and diversity is achieved. Experimental research on the WFG series of benchmark problems shows that the MaSCA-KD algorithm has a certain degree of competitiveness compared with the existing algorithms. The algorithm has good performance and can be used as an alternative tool for many-objective optimization problems.

        Heat treatments of peptides from oyster (Crassostrea gigas) and the impact on their digestibility and angiotensin I converting enzyme inhibitory activity

        The changes of protein digestibility, the peptidesin the digestive juice and angiotensin I converting enzyme(ACE) inhibitory activity after heating of oysters wereinvestigated. The digestibility of raw oysters was 71.1%,and that of oysters heated at 100 C was 67.9%. A total of169 and 370 peptides were identified from the digestion ofraw oysters and heated oysters, respectively. According toUPLC-Q-TOF-MS spectra, the peptides with a molecularweight below 2000 Da accounted for 87.6% of the totalpeptides of raw oysters and 94% of heated oysters. Testingthe ACE inhibitory activity in vitro, the IC50 values of rawoyster and cooked oyster were 6.77 lg/mL and 3.34 lg/mL, respectively. Taken together, the results showed thatheated oysters could produce more active peptides andprovide ACE inhibitory activity.

        A Light-Inducible Bidirectional Promoter Initiates Expression of Both Genes SHH2 and CFM3 in Brassica napus L.

        A bidirectional promoter is an intergenic region between a pair of adjacent and oppositely transcribed genes (‘head-to-head’ genes) that concurrently initiates the expression of both genes. In this work, we identified a bidirectional promoter PBn265 with a 265 bp sequence between transcription initiation codons (ATG) of two genes encoding the homeodomain protein SHH2 and chloroplast GROUP II intron splicing factor CFM3, respectively, in genome of Brassica napus. The histochemical staining and fluorescence microscopy results demonstrated that PBn265 possess bidirectional promoting activity in the various tissues of the transgenic Arabidopsis thaliana. Notably, the bidirectional expression activities of PBn265 were significantly increased following red or blue light induction. Furthermore, the gene structure, ‘SHH2-BiP-CFM3’, appears highly conservative among various species in Brassicaceae. Considering its short sequence length, high sequence conservation and light-inducible properties, this promoter from Brassica napus most likely has application potential in bioengineering and agricultural molecular breeding.
