        Production and Optimization of a Kiwi Pectin Methylesterase Inhibitor in Pichia pastoris GS115

        Gene sequence coding of a kiwi pectin methylesteraseinhibitor was optimized, synthesized, and expressedin Pichia pastoris GS115 based on P. pastoris preferredcodon usage. The expression level of the recombinantprotein (kwPMEI) increased by 89.74% after codonoptimization. Expression conditions of recombinant strainswere optimized. The highest production of kwPMEI wasachieved using 0.8% sorbitol (added every 24 h), 0.05%oleic acid (added at the beginning of induction), and 0.5%methanol (added every 12 h). kwPMEI was purified usingNi2+ chelating affinity chromatography and 17 mg of theprotein was harvested from 60 mL of a culture supernatant. Activity analysis showed that kwPMEI efficiently inhibitedthe activity of different plant PMEs. High expression levelsand purification of kwPMEI will promote applications infruit and vegetable juices.

        Calcium supplements and structure–activity relationship of peptide-calcium chelates: a review

        In recent years, the functional properties of peptides derived from food proteins have attracted considerable interest. Among them, bioactive peptides that can effectively bind metals have application prospects in the improvement of mineral bioavailability, and compensating for the shortcomings of the generally low bioavailability of inorganic mineral supplements. Although a reasonable understanding of structure activity relationship related to the calcium binding of peptides has been gained, physiological connections of peptides as mineral carriers to gastrointestinal uptake needs further research. Hence, this article reviews (1) the development of calcium supplements; (2) inorganic calcium sources and bone calcium; (3) source and acquisition of biologically active peptides; (4) calcium peptide chelation mechanism and structure–activity relationship; and (5) Methods for evaluating calcium bioavailability.

        Trace surface fluorination and tungsten-intercalation cooperated dual modification induced photo-activity enhancement of titanium dioxide

        General modification that utilizes insoluble tungstic acid or highly toxic hydrofluoric acid to improvecharge separation and transfer in TiO2 results in considerable issues such as uneven doping, significantimpacts on particle size and morphology, as well as environmental hazards. In the present work, an environmentalbenign one-pot dual-modification approach was demonstrated that uses soluble sodium tungstateand only trace-level sodium fluoride as replacements. The photoactivity efficiency for decolorationof methylene blue (MB) solution was improved by 33.6% and 119.5% under ultraviolet and visible light,respectively. Meanwhile, the photocurrent density reached an enhancement by 181.2% by this dualmodification. The dual-modification had negligible influence on the crystal structure and the surface areaof the TiO2 nanoparticles. Density functional calculation suggested the remarkable improvements of thephotocatalysis of TiO2 can be ascribed to the rapid charge separation and transfer owing to the downshiftof conduction band from the tungsten doping and localized spatial charge separation from the surfacefluorination enabled by the dual-modification approach.
