The recent scientific interest in acupuncture and its mechanisms shows a timewise coincidence with the discovery of the endogenous opioids, the endorphins and 5-HT. It has been commonplace to assume that acupuncture acts via release of these substances and research along this line has provided ample evidence for the existence of such a connection. However, it has also become clear that other neuronal systems are involved. This is not unexpected in view of the complex and integrated nature of the nervous system. It may therefore be more appropriate to discuss pathways and circuits activated by acupuncture rather than single systems.

The purpose of this study was to examine the effects of Citrus Reticulata(CR) on the Cell Detachment and Apoptosis in Human Gastric Cancer SNU-668 Cells. The effect of CR on apoptosis was investigated through MTT assay, DAPI staining, and TUNEL assay. We also performed RT-PCR for apoptotic genes including BCL-2, BAX, and caspase-3, the caspase-3 activity assay, and western blotting for pro-CASP-3. Then, to detect that adhesion of cell to ECM was reduced by CR, we investigated mRNA expression of CDH1 and PTK2 using RT-PCR, and their protein expressions using western blotting, and immunocytochemistry in SNU-668 cells. In this study, the results showed that treatment of CR induced time and dose-dependent cell death in SNU-668 cells. Downregulated mRNA expression of BCL-2, and upregulated mRNA expressions of BAX and CASP-3 indicated that the cell death was due to apoptosis. Protein expression of inactivated CASP-3, and caspase-3 activity assay also showed that apoptosis was induced in CR-treated cells.

Aqueous Extract of Ma huang Suppresses Neuropeptide Y Expression in Food-Deprived Rat Hypothalamus

Ma huang, the dried plant stem of Ephedra Intermedia Schrenk et C.A., contains an ephedrine-type alkaloid and has been used for weight loss. Neuropeptide Y (NPY), a 36-amino acid peptide, is concentrated in the hypothalamus and stimulates feeding desire. In this study, the effect of Ma huang an the expressions of NPY in the hypothalamus of rats was investigated using immunohistochemistry. Food-deprivation enhanced the NPY expression in the hypothalamus. Ma huang suppressed the food-deprivation-induced enhancement of NPY expression. Present results suggest that Ma huang curbs the food desire by suppressing the NPY expression under food-deprivation conditions.

<P>Ephedrae Herba (EH) is obtained by drying the stems of Ephedra sinica Stapf (family Ephedraceae). EH has been used clinically to treat colds and to reduce edema in skin. The effects of EH on melanogenesis were studied in B16 murine melanocytes. The tyrosinase activity and melanin content were measured after incubation with EH. Both reverse transcription-polymerase chain reaction and cDNA microarray analysis were used to study the mechanism of EH action. EH decreased tyrosinase activity and melanin content in a dose-dependent manner. EH decreased microphthalmia-associated transcription factor (MITF) and tyrosinase gene expression. These effects were controlled by MITF-mediated regulation of tyrosinase gene expression. EH also altered the expression of about 100 other genes. These results suggest that EH may be used clinically to treat freckles and liver spots. Copyright © 2006 John Wiley & Sons, Ltd.</P>

Background: School-aged children born very preterm have been suggested to have worse cognitive and behavioral outcomes than children born full-term. Executive function (EF) is a higher level of cognitive function related to academic achievement. The present study aimed to evaluate the cognitive (including EF) and behavioral outcomes of Korean children born extremely preterm (EP) and to analyze any biological or socioeconomic risk factors for poor cognitive outcomes in this population. Methods: A total of 71 infants weighing < 1,000 g at birth or born before 30 weeks of gestation (EP group) who were admitted to the neonatal intensive care unit from 2008 to 2009 were included in this study and compared with 40 term-birth controls. The Korean Wechsler Intelligence Scale for Children-Fourth Edition, Advanced Test of Attention (ATA), Stroop test, Children's Color Trails Test (CCTT), and Wisconsin Card Sorting Test (WCST) were used. Additionally, the Korean Child Behavior Checklist (K-CBCL) and Korean ADHD Rating Scale (K-ARS) were completed. Perinatal and demographic data were collected and analyzed. Results: The mean full-scale intelligence quotient (FSIQ) score in the EP group was significantly lower than that of the term control group (89.1 ± 18.3 vs. 107.1 ± 12.7; P < 0.001). In the EP group, 26 (37%) children had an FSIQ score below 85, compared to only one child (3%) in the control group. Furthermore, the EP group showed significantly worse EF test results (ATA, Stroop test, CCTT, WCST). Except for the higher social immaturity subscore in the EP group, the K-CBCL and K-ARS scores were not different between the two groups. EP children who received laser treatment for retinopathy of prematurity (ROP) had an 8.8-fold increased risk of a low FSIQ score, and a 1-point increase in the discharge weight Z-score decreased the risk of a low FSIQ score by approximately half in this EP cohort. Conclusion: This is the first Korean study to investigate the cognitive and behavioral outcomes of school-aged children born EP. In the study cohort, EP children exhibited significantly lower FSIQ scores and EF than their full-term peers, and 37% of them had cognitive problems. Nonetheless, except for social immaturity, the behavioral problems were not different in EP children. Severe ROP and low discharge weight Z-score were identified as independent risk factors for low FSIQ score after adjusting for birth weight.

Ginseng radix, the root of Panax C. A. MEYER (Araliaceae), is one of the best-known Oriental medicinal herbs with numerous therapeutic applications. To investigate whether Ginseng radix possesses a protective effect against 1-methyl-4-phenylpyridine (MPP^(+))-induced cytotoxicity in neuronal cells, 3-(4,5-dimethylthiazol-2yl)-2.5-diphenyltetrazolium bromide (MTT) assay, flow cytometry, DNA fragmentation assay, reverse transcription-polymerase chain reaction (RT-PCR), Western blotting, and caspase-3 enzyme assay were performed on PC12 neuronal cells. Cells treated with MPP^(+) exhibited various apoptotic features, while cell pretreated with Ginseng radix prior to MPP^(+) exposure showed a decrease in the occurrence of apoptotic features. These results suggest that Ginseng radix may exert a protective effect against MPP^(+)-induced apoptosis in PC12 cells.

The possibility has been raised that Folium mort is clinically effective for the treatment and prevention of diabetes In the present study, the effects of Folium mori on cell proliferation and expression of neuropeptide Y(NPY) in the dentate gyrus of rats with streptozotocin (STZ)-induced diabetes were investigated by 5-bromo-2'deoxyuridine (BrdU) immunohistochemistry and NPY immunohistochemistry. In rats with STZ-induced diabetes, cell proliferation and NPY expression in the dentate gyrus were suppressed, and treatment with Folium mori was shown to increase new cell formation and NPY expression in the gyrus in both normal rats and those with STZ-induced diabetes. In light of previous studies, this result appears to indicate that increased expression of NPY in the dentate gyrus induced by treatment with Folium mori is associated with the observed effect of Folium mori extract on cell proliferation. Based on the present results, it is suggested that Folium mori treatment may aid in the recovery from the central nervous system complications of diabetes mellitus.

In this study, effects of acupuncture at zusanli acupoint on cell proliferation and expression of neuropeatide Y(NPY) in the dentate gyrus (DG) of streptozotocin-induced diabetic rats were investigated. Sprague-Dawley rats were divided into six groups: the control group, the non-diabetic and Zusanli-acupunctured group, the non-diabetic and non-acupoint-acupunctured group, the streptozotocin-induced-diabetes group, the streptozotocin-induced-diabetes and Zusanli-acupunctured group, and the streptozotocin-induced-diabetes and non-acupoint-acupunctured group. In the streptozo-tocin-induced diabetes group, the mean 5-bromo-2'-deoxyuridine-positive and NPY-positive cell counts in the DG were significantly decreased compared to the control group. Stimulation of the Zusanli resulted in increased cell proliferation and neuropeptide Y levels in the diabetic group. In the present results, it can be suggested that acupuncture may affert cell proliferation in the DG of streptozotocin-induced diabetic rats by regulating neuropeptide Y expression. ⓒ 2002 Elsevier Science Ireland Ltd.
