Cross-reactive material 197 (CRM<sub>197</sub>) is a non-toxic mutant of diphtheria toxin containing a single amino acid substitution of glycine 52 with glutamic acid. CRM<sub>197</sub> has been used as a carrier protein for poorly immunogenic polysaccharide antigens to improve immune responses. In this study, to develop a sandwich ELISA that can detect CRM<sub>197</sub> and CRM<sub>197</sub> conjugate vaccines, we generated a human anti-CRM<sub>197</sub> monoclonal antibody (mAb) 3F9 using a phage-displayed human synthetic Fab library and produced mouse anti-CRM197 polyclonal antibody. The affinity (K<sub>D</sub>) of 3F9 for CRM<sub>197</sub> was 3.55 nM, based on Bio-Layer interferometry, and it bound specifically to the B fragment of CRM<sub>197</sub>. The sandwich ELISA was carried out using 3F9 as a capture antibody and the mouse polyclonal antibody as a detection antibody. The detection limit of the sandwich ELISA was <1 ng/ml CRM<sub>197</sub>. In addition, the 3F9 antibody bound to the CRM<sub>197</sub>-polysaccharide conjugates tested in a dose-dependent manner. This ELISA system will be useful for the quantification and characterization of CRM<sub>197</sub> and CRM<sub>197</sub> conjugate vaccines. To our knowledge, this study is the first to generate a human monoclonal antibody against CRM197 and to develop a sandwich ELISA for CRM<sub>197</sub> conjugate vaccines.
A metazoan ortholog of SpoT hydrolyzes ppGpp and functions in starvation responses

In nutrient-starved bacteria, RelA and SpoT proteins have key roles in reducing cell growth and overcoming stresses. Here we identify functional SpoT orthologs in metazoa (named Mesh1, encoded by HDDC3 in human and Q9VAM9 in Drosophila melanogaster) and reveal their structures and functions. Like the bacterial enzyme, Mesh1 proteins contain an active site for ppGpp hydrolysis and a conserved His-Asp??box motif for Mn<SUP>2+</SUP> binding. Consistent with these structural data, Mesh1 efficiently catalyzes hydrolysis of guanosine 3??5??diphosphate (ppGpp) both in vitro and in vivo. Mesh1 also suppresses SpoT-deficient lethality and RelA-induced delayed cell growth in bacteria. Notably, deletion of Mesh1 (Q9VAM9) in Drosophila induces retarded body growth and impaired starvation resistance. Microarray analyses reveal that the amino acid??starved Mesh1 null mutant has highly downregulated DNA and protein synthesis??related genes and upregulated stress-responsible genes. These data suggest that metazoan SpoT orthologs have an evolutionarily conserved function in starvation responses.

<P>To confirm the beneficial effects of alpha (1,2)-fucosyltransferase (FUT1) M307 (A) on piglet survival on commercial farms, we performed PCR-RFLP analysis of FUT1 M307 in successfully marketed (n?=?245) and disease affected/deceased pigs during weaning (n?=?252) at a commercial farm. We also evaluated the FUT1 genotypes of 190 healthy pigs from three different genetic backgrounds. The distribution of genotypes differed between the successfully marketed and disease affected/deceased pig groups. The frequency of the A allele, associated with resistance to edema and post-weaning diarrhea, was higher in the post-weaning survival group (0.21) than in the non-survival group (0.16, P?<?0.05). The odds ratio for piglet survival between AA and GG genotypes was 1.98; thus, piglet survival for individuals with the AA genotype was almost two-fold greater than for GG individuals. The FUT1 gene polymorphism can be used as an effective marker for selection programs to improve post-weaning piglet survival.</P>

<P>In this study, we sought to identify novel antimicrobial peptides (AMPs) in Python bivittatus through bioinformatic analyses of publicly available genome information and experimental validation. In our analysis of the python genome, we identified 29 AMP-related candidate sequences. Of these, we selected five cathelicidin-like sequences and subjected them to further in silico analyses. The results showed that these sequences likely have antimicrobial activity. The sequences were named Pb-CATH1 to Pb-CATH5 according to their sequence similarity to previously reported snake cathelicidins. We predicted their molecular structure and then chemically synthesized the mature peptide for three putative cathelicidins and subjected them to biological activity tests. Interestingly, all three peptides showed potent antimicrobial effects against Gram-negative bacteria but very weak activity against Gram-positive bacteria. Remarkably,Delta Pb-CATH4 showed potent activity against antibiotic-resistant clinical isolates and also was observed to possess very low hemolytic activity and cytotoxicity. Delta Pb-CATH4 also showed considerable serum stability. Electron microscopic analysis indicated that. Delta Pb-CATH4 exerts its effects via toroidal pore preformation. Structural comparison of the cathelicidins identified in this study to previously reported ones revealed that these Pb-CATHs are representatives of a new group of reptilian cathelicidins lacking the acidic connecting domain. Furthermore, Pb-CATH4 possesses a completely different mature peptide sequence from those of previously described reptilian cathelicidins. These new AMPs may be candidates for the development of alternatives to or complements of antibiotics to control multidrug-resistant pathogens.</P>
Effects of Aquarobic Exercise on Body Composition and Cardiovascular Index in Elderly Women

Objective: The community has implemented a variety of exercise programs for the elderly population. However, studies comparing changes in the cardiovascular system through body composition and blood analysis after applying aqua aerobic exercise are insufficient. The purpose of this study was to investigate the effect of a 12 weeks aqua exercise on body composition and cardiovascular index in elderly women. Design: Non-equivalence pre-post design. Methods: Thirty elderly women were assigned to aquarobic group(n=15) and control group(n=15). The aquarobic exercise was 60 minutes per every session, 2times per week, for 12weeks. Weight, skeletal muscle mass, body fat percentage, total cholesterol, triglyceride, HDL cholesterol, and LDL cholesterol were measured before and after the exercise program. Paired and independent t-tests were performed using SPSS program. Results: The result showed was weight(p<0.05), body fat percentage(p<0.05), total cholesterol(p<0.05), triglyceride(p<0.05), HDL cholesterol(p<0.05), and LDL cholesterol(p<0.05) were significantly decreased and skeletal muscle mass(p>0.05) was not significantly increased after aquatic exercise program. Conclusions: According to the study result, aqua exercise program made a positive effect in the improvement of obesity and cardiovascular function in elderly women. Therefore, we suggest that you actively consider implementing the Aquarobic exercise program when operating the community health promotion program in the future. In addition, in future studies, comparative studies according to various ages and genders and studies on the effects of aqua aerobic exercise as a community exercise program for chronically illness patients are needed.
