        LAB Fermentation Improves Production of Bioactive Compounds and Antioxidant Activity of Withania somnifera Extract and Its Metabolic Signatures as Revealed by LC-MS/MS

        In this study we investigated the effect of lactic acid bacteria (LAB) fermentation on the ingredients and anti-oxidant activity of Withania somnifera extract. Four strains of LAB could proliferate normally in medium containing W. somnifera extract after the pH reached 3.1~3.5. LAB fermentation increased the content of alcohols and ketones, endowing the extract with the characteristic aroma of fermentation. Compared to the control, the DPPH and ABTS free radical scavenging rates in the fermented samples were significantly improved, ranging from 48.5% to 59.6% and 1.2% to 6.4%. The content of total phenols was significantly increased by 36.1% during the fermentation of mixed bacteria. Moreover, the original composition spectrum of the extract was significantly changed while the differentially accumulated metabolites (DAMs) were closely related to bile secretion, tryptophan metabolism and purine metabolism. Therefore, LAB fermentation can be used as a promising way to improve the flavor and bioactivity of the extracts of W. somnifera, making the ferments more attractive for use as functional food.

        Kinematic response of industrial robot with uncertain-but-bounded parameters using interval analysis method

        The uncertain parameters of the deviation in link dimensions and joint clearances caused by manufacturing and assembling errors have a strong impact on the degree of accuracy of the industrial robot. Understanding how these uncertain parameters influence the kinematic response of the industrial robot is critical for design. In this paper, the interval method is used for analysing the kinematic response of the industrial robot with uncertain-but-bounded parameters. The interval used for describing the uncertain parameters is estimated by the grayscale theory. The Denavit-Hartenberg (D-H) method is used for establishing the kinematic model of a 6-degree-of-freedom industrial robot. The kinematic response of the industrial robot calculated by the interval method with uncertain parameters is compared with the results obtained by the probabilistic approach. The comparison results show that the upper or lower bound obtained by interval method is far away from the ideal kinematic response than that obtained by the probabilistic approach. That is a true portrayal of the essence of interval method and probabilistic approach. The research contributes to choose the tolerance zone of uncertain parameters, such as the link dimensions and joint clearances, during the design process of industrial robots.

        From Mount Meru to Mind-Only: Chinese Buddhist Cosmology in the Fajie anli tu 法界安立圖 or An Illustrated [Guide to] the Established Order of the Dharma-realm

        Fajie anli tu (An Illustrated [Guide] to the Established Order of the Dharmadhatu 法界安立圖, X 972) is an orderly account of Buddhist cosmology composed by the monk Renchao 仁潮 in Hangzhou. This book reflects the most detailed account late imperial Chinese Buddhists had of Buddhist cosmology and the physical world around them. It was well received in China since its first publication in 1607 and had a far-reaching influence during its transmission to other parts of the world, especially Japan and Britain. However, this book has not been given the attention it merits in contemporary studies of Buddhist history. In this essay, I introduce its authorship, analyze its content and writing skills, as well as examine its reception among disparate groups of readers in China and abroad using a methodology developed in the field of book history. Through a close reading of texts from the work and its history of circulation, this essay demonstrates how Buddhist cosmological discourses evolved within the Chinese context from detailed descriptions of the physical cosmos to philosophical explanations of the universe emphasizing the supremacy of the “mind” (xin 心).

        Dual delivery of bone morphogenetic protein‑2 and basic fibroblast growth factor from nanohydroxyapatite/collagen for bone tissue engineering

        Background: In bone tissue engineering, the fabrication and biocompatibility of scaffold are crucial. Among many scaffold materials, nanohydroxyapatite (nHAP) and collagen (COL) are chosen as building materials of scaffold. At the same time, growth factors were also used to modify the scaffolds. Methods: In this study, blending and freeze drying methods were adopted together in order to build basic fibroblast growth factor (bFGF)-bone morphogenetic protein-2 (BMP-2)-nHAP/COL scaffolds. ELISA was applied to test the release of bFGF and BMP-2 on the scaffold. The flow cytometry was used to identify bone marrow mesenchymal stem cells (BMSCs). Scanning electron microscope was adopted to observe scaffolds and cells morphology. BMSCs were seeded on the scaffolds to test the biological compatibility in vitro. Cells were counted to detect early cell adhesion. Cell counting kit-8 assay was adopted to detect cell proliferation and alkalinephosphatase assay was applied to detect cell activity. Results: The characterization of bFGF-BMP-2-nHAP/COL scaffolds meets the requirements of ideal bone tissue engineering scaffolds. BMSCs that were isolated, purified and passaged satisfied the needs of further experiments. The growth status of cells on bFGF-BMP-2-nHAP/COL scaffolds was satisfactory. Cell adhesion was the highest in the bFGF-BMP-2-nHAP/COL scaffolds group. The cell viability and ALP activity of bFGF-BMP-2-nHAP/COL scaffolds group were the highest. Conclusion: Taken together, bFGF-BMP-2-nHAP/COL scaffolds have good biocompatibility in vitro and promote adhesion, proliferation, differentiation of BMSCs.

        Effects of coolant temperature on particulate deposition and cooling efficiency of EGR coolers

        Porous fouling layers formed by particulate deposition in the EGR cooler tubes degrade the cooling efficiency of the EGR coolers. This research has investigated the changes of the particulate concentration under varied coolant temperature conditions and the effects of the coolant temperature on the cooling efficiency of the cooler tubes by measuring the particulate concentration and size distribution at the cooler inlet and outlet. The experiment results indicate that for the particulates of different sizes, the thermophoretic force acting on them and the regeneration vary under the same experiment conditions, presenting distinctive effects on the particulate concentration distribution in the exhaust gas and causing concentrations of nuclear particulates and aggregated particulates to change, respectively. The mass of fouling layers inside the cooler tubes increases as the coolant temperature decreases. Decreasing the coolant temperature not only increases the quantity of fouling particulates but also forms more particulates of big sizes, causing cooling efficiency of the cooler tube per unit length to decrease with the decrease of coolant temperature and the increase of the tube length.

        An active learning hybrid reliability method for positioning accuracy of industrial robots

        Popsitioning accuracy is an important index for evaluating the capacity of industrial robots. As a mechanism with multi-degree of freedom, the uncertainties of industrial robots are diverse and analyzing the positioning accuracy reliability is time consuming. To improve computation efficiency, a new active learning method based on Kriging model is proposed for hybrid reliability analysis of positioning accuracy with random and interval variables. In this study, the updated samples were selected through U learning function in the vicinity of limit-state function. A new stopping criterion based on expected risk function was exploited to judge whether the accuracy of Kriging model is enough. Two numerical examples and one engineering example were provided to verify the efficiency and accuracy of the proposed method. The results indicate that the proposed method is accurate and efficient.

      • Chemopreventive Effects of Korean Angelica versus Its Major Pyranocoumarins on Two Lineages of Transgenic Adenocarcinoma of Mouse Prostate Carcinogenesis

        <P>We showed previously that daily gavage of <I>Angelica gigas</I> Nakai (AGN) root ethanolic extract starting 8 weeks of age inhibited growth of prostate epithelium and neuroendocrine carcinomas (NE-Ca) in the transgenic adenocarcinoma of mouse prostate (TRAMP) model. Because decursin (D) and its isomer decursinol angelate (DA) are major pyranocoumarins in AGN extract, we tested the hypothesis that D/DA represented active/prodrug compounds against TRAMP carcinogenesis. Three groups of male C57BL/6 TRAMP mice were gavage treated daily with excipient vehicle, AGN (5 mg per mouse), or equimolar D/DA (3 mg per mouse) from 8 weeks to 16 or 28 weeks of age. Measurement of plasma and NE-Ca D, DA, and their common metabolite decursinol indicated similar retention from AGN versus D/DA dosing. The growth of TRAMP dorsolateral prostate (DLP) in AGN- and D/DA-treated mice was inhibited by 66% and 61% at 16 weeks and by 67% and 72% at 28 weeks, respectively. Survival of mice bearing NE-Ca to 28 weeks was improved by AGN, but not by D/DA. Nevertheless, AGN- and D/DA-treated mice had lower NE-Ca burden. Immunohistochemical and mRNA analyses of DLP showed that AGN and D/DA exerted similar inhibition of TRAMP epithelial lesion progression and key cell-cycle genes. Profiling of NE-Ca mRNA showed a greater scope of modulating angiogenesis, epithelial–mesenchymal transition, invasion–metastasis, and inflammation genes by AGN than D/DA.         The standards of obstetrics and gynecology core outcome sets: A scoping review

        Background: Core outcome sets (COSs) are the minimum outcomes which should be measured and reported by researchers investigating a specific condition. The definition of standards of COSs vary across different health-related areas. This investigated the characteristics of COSs regarding obstetrics and gynecology (OG) and examined the reports and designs of standards of OG COSs. Methods: A comprehensive search was conduced on the COMET database on December 20, 2019 to identify systematic reviews on COSs. Two reviewers independently evaluated whether the reported OG COS met the reporting requirements as stipulated in the Core Outcome Set-STAndards for Reporting (COS-STAR) statement checklist and the minimum design recommendations as outlined in the Core Outcome Set-STAndards for Development (COS-STAD) checklist. Results: Forty-four OG COSs related to 26 topics were identified. None of them met all the 25 standards of COS-STAR statement which representing 18 items considered essential for transparent and complete reporting list for all COS studies (range: 6.0-24.0, median: 14.0). The compliance rates to 16 standards of methods and result sections ranged from 27.3%–68.2%. Total COS-STAR compliance items for OG COSs with the prior protocol was significantly higher than without prior protocol (MD = 3.846, 95% CI: 0.835–6.858, P = 0.012). None of the OG COSs met all the 12 criteria in the COS-STAD minimum standards (range: 3.0-11.0, median: 5.0). The compliance rates for all three standards of stakeholders involved and all four standards of the consensus process were lower than 60%. Conclusions: Methodological and reporting standards of OG COSs should be improved.
