        CAD/CAM-assisted total joint replacement for correction of facial asymmetry caused by large osteoma of mandibular condyle

        Osteoma of mandibular condyle is uncommon benign tumor that grows slowly. Growth of condylar osteoma can cause functional and morphological problems such as facial asymmetry and malocclusion. Surgical removal and concurrent temporomandibular condyle replacement is a strategy that can resolve functional and aesthetic problems associated with large osteoma of mandibular condyle. Total joint replacement (TJR) with computer-associated surgical simulation (CASS) and computer-aided design/computer-aided manufacturing (CAD/CAM) technology can effectively and accurately rehabilitate debilitated condyle. A 52-year-old female visited department of Oral and Maxillofacial Surgery at Seoul National University Dental Hospital with a chief complaint of facial asymmetry and discomfort of left temporomandibular joint (TMJ). Radiographic and CT images showed a large bony mass at left TMJ surrounding left condylar head. Clinical examination revealed mandibular shift to right side, causing malocclusion and facial asymmetry. TJR of left TMJ with customized alloplastic condyle prosthesis and Biomet fossa was planned for the patient. Condyle was designed and fabricated using CAD/CAM. Surgical guides and occlusal splint fabricated with CAD/CAM technology was well. After 2 years, patient still showed occlusal stability with restored facial symmetry. No complications or recurrence were observed. TJR with customized alloplastic condyle and stock fossa can be an effective treatment strategy for large symptomatic condylar osteoma in restoration of normal function and aesthetics.

        Temperature-dependent immune response of olive flounder (Paralichthys olivaceus) infected with viral hemorrhagic septicemia virus (VHSV)

        <P>Olive flounder (Paralichthys olivaceus) is one of the most economically important aquaculture fish. However, its production is often affected by various diseases, especially viral hemorrhagic septicemia virus (VHSV) that cause serious economic losses. In this study, we sequenced the whole transcriptome of the P. olivaceus using Illumina RNA-sEq. De novo assembly of control and virus-infected cDNA libraries of head kidney at 13 and 20 A degrees C was accomplished with 2,007,532,438 raw reads, resulting in 244,578 unigenes with an average length of 533 bp and found 65,535 candidate coding unigenes with homology to other species by BLAST analysis. DEG analysis among control and virus-infected head kidney samples of 13 and 20 A degrees C revealed that 1290 up-regulated and 162 down-regulated genes (p ae<currency> 0.01), linked to metabolism, virulence factors, adhesion and immune-response. We constructed an expressed gene catalog for the P. olivaceus to serve as a resource for marine environmental genomic and immuno-genetic/genomic studies focused on uncovering the molecular mechanisms underlying the responses of P. olivaceus to VHSV under different temperature.</P>

        Olive flounder CD276 (B7-H3) a coinhibitory molecule for T cells: Responses during viral hemorrhagic septicemia virus (VHSV) stimulation

        <P>Coinhibitory pathways in the B7-CD28 family provide critical inhibitory signals that regulate immune homeostasis, defense and protect tissue integrity. CD276 (B7-H3) is an important immune checkpoint member of this family, which is induced on antigen-presenting cells (APCs), and plays an important role in the inhibition of T cell function. We have characterized the CD276 gene of olive flounder, Paralichthys olivaceus. OfCD276 has an ORF of 912 bp that codes for 303 amino acids with a predicted molecular mass of 33 kDa. It is a type I trans membrane protein with a single extracellular V- and C-like Ig domains, a transmembrane region, and a highly diverse cytoplasmic tail. This gene was distinctly expressed in gill, spleen, and skin, and sparsely expressed in other tissues. Pathogen stimulation by VHSV revealed that transcription of OfCD276 was induced on early hours in liver and expressed late in head kidney, spleen, intestine and gill tissues. Flow cytometry analysis of leukocytes revealed the percentage of granulocytes and lymphocytes that expressed OfCD276 molecules on their cell surface was 85.1% and 3.1%, respectively. Our study shows a significant role played by this coinhibitory molecule that participate in the regulation of the cell mediated immune response.</P>

      • S-593 Impact of cyclophilin A in gastric cancer patients who received platinum-based chemotherapy

        Introduction: Cyclophilin A (CypA), which is a protein that mediates protein folding and trafficking, has known to be associated with cancer progression and drug resistance such as cisplatin. However, the actual effect in cancer patients has not been elucidated. Methods: We collected clinical data from the cohort of 289 gastric cancer patients. Subjects were selected as following criteria: 1) adjuvant or first-line palliative chemotherapy containing platinum, 2) available FFPE tissue for immunohistochemical stain for CypA. We divided the population into two groups; Group A was patients who received adjuvant chemotherapy and Group P was those of palliative chemotherapy-treated group. The score of CypA immunohistochemistry was calculated depending on the intensity of stain and the percentage of stained cells. We performed survival analysis based on CypA expression. Results: 1) Group A (n=147) Patients with lower expression of CypA (<vs. ≥ median IHC score) showed significantly worse progression-free survival (PFS, 2.4 years vs NR, p=0.043), and overall survival (OS, 3.4 months vs. NR, p=0.002). In subgroup analysis, level of CypA expression discriminated PFS significantly in patients with age less than 65, poorly differentiated histology, signet ring cell type, and OS for those with use of oxaliplatin, age less than 65, stage III, poorly differentiated histology, diffuse type of Lauren classification, and signet ring cell type favoring higher expression of CypA. 2) Group P (n=142) There was no relationship between expression level of CypA and PFS/OS in patients with palliative settings. However, in patients who were treated with oxaliplatin-containing chemotherapy, lower expression of CypA showed worse OS (8.3 vs 11.1 months, p=0.003) Conclusions: Lower level of CypA expression was associated with worse PFS and OS in patients with adjuvant setting, especially in case of advanced stage, and high grade histology. Although CypA did not have significant effect on survivals in palliative chemotherapy-received patients, those who received oxaliplatin showed worse OS with lower CypA.

        <P><B>Abstract</B></P> <P>Dietary garlic has been suggested to possess anticancer properties, and several attempts have been made to isolate the anticancer compounds. In this study, we efficiently synthesized <I>N</I>-benzyl-<I>N</I>-methyl-dodecan-1-amine (BMDA) by the reductive amination method. We evaluated the potential anticancer activities of BMDA against A549 lung cancer cells with cancer stem cell-like phenotypes due to the overexpression of cancer upregulated gene (CUG)2. <I>N</I>-Benzyl-<I>N</I>-methyl-dodecan-1-amine treatment sensitized A549 cells overexpressing CUG2 (A549-CUG2) to apoptosis and autophagy compared with those of the control cells. The treatment with BMDA also reduced tumor development in xenografted nude mice. Furthermore, BMDA inhibited cell migration, invasion, and sphere formation in A549-CUG2 cells, in which TGF-β signaling is involved. Further analysis showed that BMDA hindered TGF-β promoter activity, protein synthesis, and phosphorylation of Smad2, thus decreasing the expression of TGF-β-targeted proteins, including Snail and Twist. <I>N</I>-Benzyl-<I>N</I>-methyl-dodecan-1-amine also decreased Twist expression <I>in vivo</I>. In addition, BMDA inhibited Akt-ERK activities, β-catenin expression, and its transcriptional activity. These results suggest that BMDA can be a promising anticancer agent against cancer cells overexpressing CUG2.</P>

        Imidacloprid는 chloronicotinyl계 침투성 살충제로서 흡입해충(sucking insects), 토양해충(soil insects),흰개미(termites), 저작해충(chewing insects) 등을 방제할 목적으로 사용되고 있다. 그러나, 꿀벌(Apismellifera)에 대하여 독성이 매우 높고, LD50이 8 ng/bee로 보고 되어 있다. 그리고, 토양내에서 imidacloprid의 수착(sorption) 정도는 유기탄소 및 무기물질 등의 영향을 많이 받는 것으로 알려져 있다. 따라서, 본 연구에서는 서로 다른 토양에서 imdacloprid의 흡/탈착특성을 알아보고, 토양내 거동형태를 파악하고자 하였다. HCB-SL-PF (silty loam, pH 6.8, OC 33.0 g kg-1), Iowa Soil (silty loam, pH6.2, OC 8.5 g kg-1)의 두 종류의 토양을 선정하였으며, 흡/탈착실험을 위하여 토양과 용액비율을 1:3(w/v)으로 하여 Imidacloprid를 0, 5, 10, 50, 100, 500 mg kg-1 수준으로 처리하여 상온에서 20 rpm의 속도로 4시간 진탕하였다. 산출된 결과를Freundlich equation에 적용한 일차식으로 나타내었고, 흡착실험에서 HCB-SL-PF 토양은 기울기 0.807 (R2 0.980), KadsF 1.897, Iowa Soil 토양은 기울기 0.950 (R20.998), KadsF 1.359로 나타났다. 탈착실험에서는 HCB-SL-PF 토양은 기울기 0.582 (R2 0.924), KadsF3.461, Iowa Soil 토양은 기울기 0.977 (R2 0.917), KadsF 1.390로 나타났다. 상대적으로 유기탄소함량이 높은 HCB-SL-PF (OC 33.0 g kg-1) 토양이 Iowa Soil (8.5 g kg-1) 토양보다 흡착계수가 높게 나타났다.

        <P>Primary cardiac lymphoma (PCL) is an extranodal non-Hodgkin's lymphoma exclusively located in the heart and/or pericardium. It is rare in immunocompetent patients and represents 1.3% of primary cardiac tumors and 0.5% of extranodal lymphomas. The clinical behavior is aggressive and the early symptoms are cardiac failure, syncope, arrhythmia, or pericardial effusion. Although echocardiography, computed tomography (CT) scan, magnetic resonance image (MRI) are the mainly used imaging techniques to detect cardiac tumors, pathologic examination is always required to confirm the diagnosis. Diagnosis of PCL is difficult due to non-specific clinical manifestations and requires invasive approach to get histopathologic evidence. While surgery with systemic chemotherapy or in combination with irradiation has been attempted, the only effective treatment is chemotherapy. However, the prognosis remains poor. We report on a 42-year-old woman who is diagnosed histopathologically as PCL by cardiac catheterization assisted percutaneous endomyocardial biopsy and treated successfully by anthracycline based chemotherapy.</P>
