The cytotoxicological responses to insect growth regulator (IGR), using tebufenozide as ecdysteroid mimic, were investigated in Drosophila Kc cells. Treatment of Kc cells with tebufenozide showed significant growth inhibition and striking morphological changes including aggregation and elongation of the cells. In order to understand the cellular mechanism underlying the response of Drosophila cells to tebufenozide, immunofluorescence microscopy was performed. We found that treatment of Kc cells with tebufenozide enhanced the reorganization of f-actin and stimulated the expression of hsp27. These data suggest a possible association of filamentous actin (f-actin) and hsp27 in the cytotoxicological mechanisms of growth regulators in Drosophila cells.

다중전송률 지원 이동 Ad Hoc 네트워크를 위한 위치 정보 기반 라우팅 프로토콜

이동 Ad Hoc 네트워크(MANET)에서 많은 영역에 걸쳐 다양한 라우팅 프로토콜들이 제안되어 왔다. 다중전송률은 현재 많은 무선 랜 카드에 의해 지원되고 있고, 그 효과에 관한 라우팅 프로토콜 알고리즘들이 연구되고 있다. 다중전송률에서의 high-rate을 이용한 데이터 전송은 일정한 시간 내에 더 많은 데이터를 전송할 수 있기 때문에 높은 throughput을 기대 할 수 있다. 그러나 high-rate을 이용한 데이터 전송의 경우 전송이 가능한 거리가 짧아져 목적노드까지 도달하는 데에 더 많은 홉을 거쳐야 한다. 그로 인해 패킷을 전달하는데 걸리는 시간이 늘어나 전체 네트워크의 성능을 저하시킬 수도 있다. 본 논문에서는 노드들의 이동으로 인한 네트워크의 토폴로지 변화나 채널의 상태에 따라 어떤 전송 속도로 데이터를 전송할 것인가를 결정하기 위해 다중전송률을 고려한 위치 정보 기반의 라우팅 프로토콜을 제안한다.

<P>Most researchers expect extreme ultraviolet lithography (EUVL) to be used to create patterns below 32 nm in semiconductor devices. An ultrathin EUV photoresist (PR) layer a few nanometers thick is required to further reduce the minimum feature size. Here, we show for the first time that pentacene molecular layers can be employed as a new EUV resist. Nanometer-scale dots and lines have been successfully realized using the new molecular resist. We clearly show the mechanism that forms the nanopatterns using a scanning photoemission microscope, EUV interference lithography, an atomic force microscope, and photoemission spectroscopy. The molecular PR has several advantages over traditional polymer EUV PRs. For example, it has high thermal/chemical stability, negligible outgassing, the ability to control the height and width on the nanometer scale, fewer residuals, no need for a chemical development process and thus a reduction of chemical waste when making nanopatterns. 1H NMR-based metabolite profiling of diet-induced obesity in a mouse mode

High-fat diets (HFD) and high-carbohydrate diets (HCD)- induced obesity through different pathways, but the metabolic differences between these diets are not fully understood. Therefore, we applied proton nuclear magnetic resonance (1H NMR)-based metabolomics to compare the metabolic patterns between C57BL/6 mice fed HCD and those fed HFD. Principal component analysis derived from 1H NMR spectra of urine showed a clear separation between the HCD and HFD groups. Based on the changes in urinary metabolites, the slow rate of weight gain in mice fed the HCD related to activation of the tricarboxylic acid cycle (resulting in increased levels of citrate and succinate in HCD mice), while the HFD affected nicotinamide metabolism (increased levels of 1-methylnicotineamide, nicotinamide-N-oxide in HFD mice), which leads to systemic oxidative stress. In addition, perturbation of gut microflora metabolism was also related to different metabolic patterns of those two diets. These findings demonstrate that 1H NMR- based metabolomics can identify diet-dependent perturbations in biological pathways. [BMB Reports 2012; 45(7): 419-424]
The Hsp27-Mediated IkBα-NFκB Signaling Axis Promotes Radiation-Induced Lung Fibrosis

<P><B>Purpose:</B></P><P>Lung fibrosis is a major side effect experienced by patients after lung cancer radiotherapy. However, effective protection strategies and underlying treatment targets remain unclear. In an effort to improve clinical outcomes, pharmacologic treatment of fibrosis is becoming increasingly popular; however, no ideal therapeutic strategy is yet available.</P><P><B>Experimental Design:</B></P><P>We used a mouse model to irradiate high focal (90 or 75 Gy) to 3-mm volume of the left lung. Lung tissues of mice were subjected to microarray, mRNA expression, and immunohistochemical analysis. Correlations of radiation (IR)-induced epithelial-mesenchymal transition (EMT) were validated in lung cell lines using appropriate treatments to activate or inhibit selected pathways.</P><P><B>Results:</B></P><P>The expression of Hsp27 was increased during IR-induced lung fibrosis in a mouse model. Inhibition of functional Hsp27 using shRNA and a synthetic small molecule inhibitor (J2) in lung cells alleviated IR-mediated EMT. The activation of NFkB pathways via direct interaction between Hsp27 and IkBα resulted in increased expressions of Twist, IL-1β, and IL-6 and facilitated IR-mediated EMT, which was identified as an underlying mechanism of Hsp27-mediated fibrosis after IR. J2 also inhibited IR-induced lung fibrosis in an orthotopic lung cancer model, and IR-induced lung fibrotic tissues from patients showed higher expression of Hsp27 than unirradiated lungs.</P><P><B>Conclusions:</B></P><P>Collectively, IkBα-NFkB signaling activation by Hsp27, which resulted in the facilitation of Twist, IL1β, and IL6 expression, is involved in the EMT process that is tightly connected to the development of IR-induced lung fibrosis. Our findings also suggest that inhibition of Hsp27 has the potential to become a valuable therapeutic strategy for IR-induced lung fibrosis.</P>
전산직 종사자의 인터넷 중독 정도가 우울감 및 직무스트레스에 미치는 영향

본 연구는 전산직과 비전산직 종사자의 인터넷 중독 정도가 직무스트레스 및 우울감에 미치는 영향을 파악하고자, 2009년 10월 25일부터 12월 9일까지 서울 및 대전에 소재한 2개의 공공기관 건물내 근무하고 종사자 500명을 대상으로 구조화된 설문지를 이용하여 일반적 특성, 직무스트레스, 우울감 정도, 일상적인 인터넷 이용, 인터넷 중독 자가진단을 조사하였다. 주요 결과는 첫째, 일반적 특성에 따른 직무스트레스 요인별 분류에서는 개인 관련요인이 낮은 연령과 미혼인 경우, 아파트 외 기타에서 거주하는 경우 높게 나타났고, 조직 외 요인의 직무스트레스에서는 전산직에 비해 비전산직이 통계적으로 높게 나타났다(p<0.05). 둘째, 인터넷 중독에 따른 직무스트레스에서는 직무 관련요인, 개인 관련요인, 조직 외 요인 모두 일반 사용자보다 잠재적 위험사용자가 높게 나타났다(p<0.05). 셋째, 우울감 정도에 따른 직무스트레스에서는 우울감 정도가 정상인 경우에 비해 경미함 및 중증도의 경우가 직무 관련요인, 개인 관련요인, 조직 외 요인 모두 높은 직무스트레스를 나타냈다(p<0.05). 넷째, 직무스트레스와 우울감을 종속변수로 한 다중회귀분석 결과를 보면 직무스트레스는 결혼여부, 가족구성원 수, 인터넷 중독이 유의한 변수였고, 25.6%의 설명력을 나타냈다. 우울감은 결혼여부와 인터넷 중독이 유의한 변수였고, 26.4%의 설명력을 나타냈다. 전산직 및 비전산직의 인터넷 중독과 직무스트레스 및 우울감은 관련성이 있는 것으로 나타났고, 인터넷 중독이 심할수록 직무스트레스가 높았으며, 우울감 정도가 심할수록 역시 직무스트레스가 높은 것으로 나타났다.
