        Molecular characterization of AIPL1 gene region in the Iranian population: application of novel informative haplotypes and detection of mutational founder effect

        Leber’s congenital amaurosis (LCA) is considered as one of the main causes of congenital blindness. In view of the genetically heterogeneous nature of the disease, indirect diagnosis using linkage analysis has proven to be useful in molecular diagnosis procedure. Mutations in AIPL1 gene are one of the leading causes of LCA. In the present study, the application of three single nucleotide polymorphic (SNP) markers related to the gene, including rs7212734, rs11658369 and rs8066853 was evaluated for the first time in the Iranian population. The markers were genotyped using tetra-primer ARMS PCR in 154 unrelated healthy individuals. Haplotype frequency and other characteristics of the markers were examined by using the GENEPOP, PowerMarker and Cubic Exact Solution software. The data indicated the presence of six different haplotypes in the Iranian population. Among them, three haplotypes showed high informativeness with frequencies C0.05. Three unrelated Iranian LCA families were analyzed. The p.W278* mutation in exon 6 of AIPL1 was found in all the families using APEX microarray chips. SNP analysis for the families showed the conservation of T-A-A haplotype linked to p.W278* mutation. All families bearing the mutation were from the Isfahan province and a founder effect was suggested in this population. Prenatal diagnosis using the markers led to the successful prediction of the fetus genotypes in all the at risk pregnancies and showed that rs7212734, rs11658369 and rs8066853 can be considered as the three informative markers for linkage analysis in carrier detection and molecular diagnosis of LCA in the Iranian population.

        Semi-field demographic performance of the invasive planthopper, Orosanga japonica (Hemiptera: Ricaniidae) on two host plants

        Orosanga japonica Melichar (Hemiptera: Ricaniidae) is an invasive polyphagous pest in Iran. The planthopper damaged host plants by sap-sucking and egg laying on new twigs. This study aimed to determine the life table parameters of O. japonica on Rubus ulmifolius Schoot (elm leaf blackberry) and Melia azedarach L. (chinaberry) under field conditions in Rudsar (North of Iran) during 2019–2020. The collected life history data were analyzed based on age-stage, two-sex life table theory. The results showed that the incubation period of O. japonica was too long (≈ 270 days) on both host plants. The nymphs emerged from mid-May 2020. The planthopper had five nymphal stages under field conditions. The nymphal duration was 41.39 ± 0.56 days and 38.13 ± 0.62 days on R. ulmifolius and M. azedarach, respectively. The total preadult survival rates were 53.2% and 51.7% on R. ulmifolius and M. azedarach, respectively. Based on the results, a significant shorter total developmental time was observed on M. azedarach. When adults emerged in late July, they laid eggs after 26.05 days and 26.75 days on R. ulmifolius and M. azedarach, respectively. The fecundity on M. azedarach (131.69 egg/female) was significantly higher than that on R. ulmifolius. However, the life table parameters were all not significantly different on the two host plants. The intrinsic rate of increase (r) were 0.0097 ± 0.0005 day − 1 and 0.0105 ± 0.0006 day − 1 on R. ulmifolius and M. azedarach, respectively. Generally, these findings could be helpful to establish an efficient management strategy for this exotic pest.

        Rotorcraft Flight Regime Recognition in Complex Maneuvers Using Adaptive Extended Kalman Filter

        The retirement time of helicopter components can be extended using the health and usage monitoring system (HUMS) and revisiting the flight spectrum with enough precision through fatigue damage analysis. The recognition of flight regimes for air vehicles with complex maneuvers is a major problem in determining the experienced spectrum. The relationship between flight variables and regimes is complex and coupled during a mission profile with multiple maneuvers that needs more efficient algorithms. In this paper, a logical framework is developed to identify the complex maneuvers based on the qualitative and descriptive interpretation of each maneuver by the pilot. The regime recognition criteria are implemented in the algorithm using an adaptive extended Kalman filter with both flight instruments and control movements as the measurements. The proposed algorithm classifies the flight regimes into the standard fatigue spectrum while needing no large amount of training data and not being sensitive to pilot behavior and mission variations. The proposed regime recognition algorithm is evaluated through extensive simulation of a comprehensive dynamic model of helicopter flight. The results show the improved accuracy of the proposed method in comparison with that of previous ones, while the proposed method is easier to embed in HUMS software without exhaustive training.
