Slight vapor deficit accelerates graft union healing of tomato plug seedling

<P>The application of grafting in tomato production has substantially improved tomato quality and yields. It has been demonstrated that humidity plays an important role in the graft healing of seedlings. This study focuses on the optimum relative humidity (RH) conditions for scion and rootstock healing of grafted tomato (Solanum lycopersicum L.) seedlings. Two tomato cultivars, 'Super Sunload' and 'Super Dotaerang', grafted onto 'B-Blocking' rootstock were subjected to one of three RH regimens: 70-80, 80-90, or 90-100%. The results showed that the scions of both cultivars showed apparent wilting under the 70-80 and 80-90% RH treatments. On this basis, the 90-100% RH treatment was subdivided into 95-96, 97-98, and 99-100% RH treatments, which were then applied. Among these subdivided RH treatments, the fresh weights of the scions and rootstocks significantly increased in response to the treatments of 97-98 and 99-100% RH, and the graft union connection of both cultivars was also enhanced after two days of healing. Furthermore, lower levels of endogenous H2O2 and less activity of antioxidant enzymes were observed in both cultivars in response to treatment with 95-96 or 97-98% RH, which indicated that less oxidative stress occurred. Overall, it is suggested that 97-98% is the optimal RH level for the graft healing of tomato seedlings.</P>
Effect of Relative Humidity on Physiology and Antioxidant Metabolism of Grafted Watermelon Seedlings

본 연구는 수박 접목묘의 활착에 미치는 상대습도의 영향을 알아보고자 수행하였다. 2품종(‘스피드’, ‘삼복꿀’)의 수박(Citrullus vulgaris Schrad.)을 ‘RS-동장군’ 박 대목(Lagenaria siceraria Stanld.)에 접목하고, 접목묘는 주간 25℃/야간 18oC, 광주기는 1일 16시간 기준으로 몰리어다이어그램을 따라 3수준의 상대습도, 95-96%[1.1-0.8(주간) 또는 0.8-0.6(야간) g·m<SUP>-3</SUP>수분부족분(VPD)], 97-98%[0.7-0.4(주간) 또는 0.5-0.3(야간) g·m<SUP>-3</SUP> VPD], 또는 99-100%[0.3-0.0(주간) 또는 0.2-0.0(야간) g·m<SUP>-3</SUP> VPD]로 유지하였다. 상대습도 처리 중 97-98% 처리는 대목과 ‘스피드’ 접수의 생체중과 초장을 가장 크게 증가시키며 접목 2일 후에 접합부의 연결을 향상시켰다. 그러나 ‘삼복꿀’의 경우 상대습도 99-100% 처리에서 경경과 초장이 증가하였다. 또한 상대습도 95-96%와 97-98% 처리에서 재배한 2 품종 모두 과산화수소가 덜 발생하고 산화적 스트레스의 지표인 항산화 효소의 활성이 덜 나타났다. 따라서 이 연구의 결과는 수박묘의 접목활착에 가장 적절한 상대습도는 97-98%였다. This study investigated the effect of different relative humidity (RH) regimes on graft healing of grafted seedlings of watermelon (Citrullus vulgaris Schrad.). Two watermelon cultivars (‘Speed’ and ‘Sambok Honey’) were grafted onto the ‘RS-Dongjanggun’ bottle gourd rootstock (Lagenaria siceraria Stanld.) and the grafted seedlings were maintained under one of three relative humidity regimes, 95-96% [1.1-0.8 (day) or 0.8-0.6 (night) g·m<SUP>-3</SUP> vapor pressure deficit (VPD)], 97-98% [ 0.7-0.4 (day) or 0.5-0.3 (night) g·m<SUP>-3</SUP> (VPD)], or 99-100% [0.3-0.0 (day) or 0.2-0.0 (night) g·m<SUP>-3</SUP> (VPD)] according to the Mollier diagram based on the air temperature of 25℃ day/18℃ night with 16 h photoperiod per day. Among the RH treatments, 97-98% significantly increased plant height and fresh weight of the rootstock and scion of the ‘Speed’ and it also enhanced the graft union connection of both cultivars after two days of grafting. However, plant height and thickness of the scion of ‘Sambok Honey’ was increased by the 99-100% RH treatment. Furthermore, both cultivars grown in the 95-96 and 97-98% RH treatments consisted of lower levels of endogenous H₂O₂ and less activities of antioxidant enzymes which illustrated the occurrence of less oxidative stress. Hence, the results of this study identified the optimal RH level for the graft healing of watermelon seedlings.
Shear behavior of reinforced HPC beams made of a low cement content without shear reinforcements

High-performance concrete (HPC) usually has higher paste and lower coarse aggregate volumes than normal concrete. The lower aggregate content of HPC can affect the shear capacity of concrete members due to the formation of smooth fractured surfaces and the subsequent development of weak interface shear transfer. Therefore, an experimental investigation was conducted to study the shear strength and cracking behavior of full-scale reinforced beams made with low-cement-content high-performance concrete (LcHPC) as well as conventional HPC. A total of fourteen flexural reinforced concrete (RC) beams without shear reinforcements were tested under a two-point load until shear failure occurred. The primary design variables included the cement content, the shear span to effective depth ratio (a/d), and the tensile steel ratio (ρw). The results indicate that LcHPC beams show comparable behaviors in crack and ultimate shear strength as compared with conventional HPC beams. Overall, the shear strength of LcHPC beams was found to be larger than that of corresponding HPC beams, particularly for an a/d value of 1.5. In addition, the crack and ultimate shear strength increased as a/d decreased or ρw increased for both LcHPC beams and HPC beams. This investigation established that LcHPC is recommendable for structural concrete applications.

<OL><LI><P>Plasma concentrations of sibutramine and its two active metabolites after single oral dose of sibutramine were determined in Korean healthy male subjects with different CYP2B6 genotypes (CYP2B6*1/*1, *1/*6 and *6/*6), either alone or after four-day pretreatment with clopidogrel or clarithromycin.</P></LI><LI><P>The pretreatment with clopidogrel and clarithromycin raised the mean area under the concentration-time curve (AUC) of sibutramine by 163% and 255%, respectively.</P></LI><LI><P>Co-administration of clarithromycin, combined with <I>CYP2B6*6/*6</I> genotype, led to highest concentration of sibutramine.</P></LI><LI><P>The molar sum AUC (M1 + M2) was raised by 35% in the clopidogrel phase but not significantly affected by clarithromycin or CYP2B6 genotype.</P></LI><LI><P>The <I>CYP2B6*6/*6</I> subjects in the clopidogrel phase showed the highest molar AUC (M1 + M2) among three genotype groups throughout the three phases.</P></LI><LI><P>The exposure of sibutramine and its metabolites seemed to be associated with the CYP2B6 genotype.</P></LI><LI><P>The treatment of clopidogrel significantly altered the disposition of active metabolites as well as sibutramine, but clarithromycin only affects the disposition of sibutramine.</P></LI><LI><P>These results suggest that the perturbation of CYP2B6 activity may contribute to the inter-individual variation of sibutramine drug responses although the clinical relevance is remained to be established.</P></LI></OL>

<P>We developed a method for determining pravastatin or pitavastatin, 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase inhibitors, in plasma using liquid chromatography and tandem mass spectrometry (LC–MS/MS). Pravastatin, pitavastatin and the internal standard fluvastatin were extracted from plasma with solid-phase extraction columns and eluted with methanol. After drying the organic layer, the residue was reconstituted in mobile phase (acetonitrile:water, 90:10, v/v) and injected onto a reversed-phase C<SUB>18</SUB> column. The isocratic mobile phase was eluted at 0.2 mL/min. The ion transitions recorded in multiple reaction monitoring mode were m/z 423 → 101, 420 → 290 and 410 → 348 for pravastatin, pitavastatin and fluvastatin, respectively. The coefficient of variation of the assay precision was less than 12.4%, the accuracy exceeded 89%. The limit of detection was 1 ng/mL for all analytes. This method was used to measure the plasma concentration of pitavastatin or pravastatin from healthy subjects after a single 4 mg oral dose of pitavastatin or 40 mg oral dose of pravastatin. This is a very simple, sensitive and accurate analytic method to determine the pharmacokinetic profiles of pitavastatin or pravastatiny. Copyright © 2007 John Wiley & Sons, Ltd.</P>

Background: Weight loss during chemotherapy has not been exclusively investigated. Macrophage inhibitory cytokine-1 (MIC-1) might play a role in its etiology. Here, we investigated the prognostic value of weight loss before chemotherapy and its relationship with MIC-1 concentration and its occurrence during chemotherapy in patients with advanced esophageal squamous cell carcinoma (ESCC). Materials and Methods: We analyzed 157 inoperable locally advanced or metastatic ESCC patients receiving first-line chemotherapy. Serum MIC-1 concentrations were assessed before chemotherapy. Patients were assigned into two groups according to their weight loss before or during chemotherapy:>5% weight loss group and ${\leq}5%$ weight loss group. Results: Patients with weight loss>5% before chemotherapy had shorter progression-free survival period (5.8 months vs. 8.7 months; p=0.027) and overall survival (10.8 months vs. 20.0 months; p=0.010). Patients with weight loss >5% during chemotherapy tended to have shorter progression-free survival (6.0 months vs. 8.1 months; p=0.062) and overall survival (8.6 months vs. 18.0 months; p=0.022), and if weight loss was reversed during chemotherapy, survival rates improved. Furthermore, serum MIC-1 concentration was closely related to weight loss before chemotherapy (p=0.001) Conclusions: Weight loss both before and during chemotherapy predicted poor outcome in advanced ESCC patients, and MIC-1 might be involved in the development of weight loss in such patients.
