      • 동물의 번식 효율증진을 위한 한약제의 활용

        유럽에서 돼지 번식 효율 증진과 비육을 위한 적절한 활용방법과 몇 가지 잘 조제된 한약제가 규격화되어 나왔다. 이 자료에는 한약제의 효능의 기준을 비롯하여 위생에 대해서도 제시하고 있다. 대학 및 연구기관, 임상 수의사들과 사료업체들이 수행했던 지난 수년간의 실험과 연구 결과를 토대로 돼지 번식효율 증진을 위한 한약제를 사료첨가제로 활용하여 최적화시켰다. 유럽의 수의분야에서 사용되었던 4가지의 특정 한약제를 기본으로 하여 돼지 사양에 있어서 가장 중요한 문제들의 대부분이 성공적으로 해결되었다. 즉 수정률, 호흡기질병, 설사와 스트레스에 관한 문제들인데 면역이 억압되고 저질의 자돈이 생산됨에 따라 생산성 저하에 따른 엄청난 경제적인 손실을 대처할 수 있다. 또한 일반적인 임상증상을 치료하면서 동시에 가장 중요한 것 중의 하나는 자기방어체계를 증진시킨다는 것이다. 이로써 동물의 면역력과 활력을 증가시키고 건강을 유지시켜서 폐사율을 감소시킨다. 게다가 여기에서 제시된 자료들은 일상적으로 사용되었던 항생제나 호르몬제제들의 사용을 대체할 수 있는 한약제들인데 고효율임이 증명되었다. 이러한 한약제들은 백신 프로그램이나 성장을 촉진시키는 인자들과 상호보완 상승작용도 가져 온다. In a comprehensive summary use and supplementation of some well defined natural herbal formulations in the European swine breeding and fattening is documented. Along with this data also the quality assurance, hygenic as well as the standards of efficacy of the formulations are presented. Series of experimental and development work over the last several years with universities, research institutes, veterinarians and feed industry proved that the application of such phylo-components (feed supplementation) result in to optimizing swine breeding practices. On the basis of 4 specific formulations tested and used in the European veterinary field most of the common problems of pig breeding and rearing can be successfully handled. Precisely, problems of fertility, respiration, diarrhea and stress; which are directly related to imerse losses in production, with poor quality of piglets associated with immune suppressions. Besides curing common symptoms, one of the most important aspect is of improvement in the self defence mechanism which results ultimately in to increased immunity, vitality and health of the animals; of course, thereby reducing the mortality which is the most economical problem observed in the animal health. Moreover, data presented here reflects on the fact that the use of such high quality alternative formulations successfully replace conventionally used antibiotics or hormonal therapies. They also work synergestically of as complementary in vaccination program or act as growth promoting agents on natural basis.

        Synergistic Effect of Essential Oils and Enterocin Produced by Enterococcus faecalis MSW5 against Foodborne Pathogens

        This study determines the combinatorial effect of enterocin MSW5 and five essential oils (EOs- Thymus vulgaris, Cymbopogon martini, Origanum vulgare, Cinnamomum zeylanicum, and Cymbopogon citrus) against Staphylococcus aureus, Listeria monocytogenes, and Salmonella Typhimurium. The Minimum Inhibitory Concentration of each antimicrobial agent was determined. The MIC of enterocin MSW5 against test pathogens was in the following order: S. aureus (0.362 ± 0.01), S. Typhimurium (0.362 ± 0.05 mg/ml), L. monocytogenes (0.725 ± 0.08 mg/ml). Among all EOs, maximum activity was observed in the case of C. zeylanicum against S. aureus (78.12 ± 0.04 ppm), S. Typhimurium (78.12 ± 0.08 ppm), and L. monocytogenes (39.00 ± 0.05 ppm). Further, the checkerboard assay was used to determine the synergistic effect between antimicrobial agents and enterocin MSW5 in combination with C. zeylanicum has shown significant synergism with the Fraction Inhibitory Concentration index (0.372) against test pathogens. Additionally, individual EOs and enterocin MSW5 have shown anti-biofilm activity, whereas their combined use has shown more significant antibiofilm activity. The maximum anti-biofilm activity was observed with the combination of enterocin MSW5 and O. vulgares against S. aureus (92.86 ± 0.06%) and S. Typhimurium (73.63 ± 0.23%) and a combination of enterocin MSW5 and C. citrus against L. monocytogenes (87.84 ± 0.15%). Therefore, combinations of antimicrobial compounds can control the growth of foodborne pathogens better than the individual agent.

        Transaminases for Green Chemistry: Recent Progress and Future Prospects

        Transaminase represents the most important biocatalysts used for the synthesis of chiral amines due to their stereoselectivity. They allow asymmetric synthesis with high yields and enantioselectivity from their corresponding ketones. Due to their environmentally friendly access for the preparation of chiral amines, they have attracted growing attention in recent times. Thus, the production of chiral compounds by transaminase catalysed reactions is considered as an important application in synthetic organic chemistry. Therefore, transaminase is considered to be an important enzyme in the pharmaceutical and chemical industries. ω-Transaminase holds great potential because of its wide substrate specificity thus making it a suitable enzyme to be used at an industrial scale. This review highlights the reaction mechanism, classification, substrate specificity, and biochemical properties. The review also showcases the application of ω-transaminase in organic chemistry with a focus on the production of active pharmaceutical ingredients (APIs).

        Application of Response Surface Methodology and Plackett Burman Design assisted with Support Vector Machine for the Optimization of Nitrilase Production by Bacillus subtilis AGAB-2

        Nitrilases are a hydrolase group of enzymes that catalyzes nitrile compounds and produce industrially important organic acids. The current objective is to optimize nitrilase production using statistical methods assisted with artificial intelligence (AI) tool from novel nitrile degrading isolate. A nitrile hydrolyzing bacteria Bacillus subtilis AGAB-2 (GenBank Ascension number- MW857547) was isolated from industrial effluent waste through an enrichment culture technique. The culture conditions were optimized by creating an orthogonal design with 7 variables to investigate the effect of the significant factors on nitrilase activity. On the basis of obtained data, an AI-driven support vector machine was used for the fitted regression, which yielded new sets of predicted responses with zero mean error and reduced root mean square error. The results of the above global optimization were regarded as the theoretical optimal function conditions. Nitrilase activity of 9832 ± 15.3 U/ml was obtained under optimized conditions, which is a 5.3-fold increase in compared to unoptimized (1822 ± 18.42 U/ml). The statistical optimization method involving Plackett Burman Design and Response surface methodology in combination with an AI tool created a better response prediction model with a significant improvement in enzyme production.

      • Factors Associated with Tobacco Use in Students Attending Local Government Schools in Mumbai, India

        Purpose: Factors associated with ever-use and differences between ever-users and non-users of tobacco among adolescent school students from low income families in Mumbai were assessed. Materials and Methods: A self-administered questionnaire, completed by 1918 students from grades 7, 8 and 9 in 12 schools managed by the city municipal corporation in July 2015, gathered data on socio-demographic characteristics, tobacco use and tobacco-related knowledge, attitudes and beliefs. Results: Although only 1% of respondents thought tobacco was cool, nearly 35% were unaware of associations between tobacco use and health problems. Male students were almost twice as likely (OR=2.5, P <= 0.05) to have ever used tobacco compared to females and Supari (areca nut) users were eight times more likely (OR=8.99, P < 0.001) than Supari non -users. Tobacco-users were more likely to agree with statements: 'People who use tobacco have more friends' (OR=2.8, P = 0.004), 'Smoking relieves stress' (OR=5.6, P = 0.002) and 'It is possible to purchase any tobacco product within 100 yards of school' (OR=10.8, P < 0.001). Conclusion: This study highlights the gains made by tobacco prevention campaigns in that almost all students did not consider tobacco as cool or a stress reliever. However, they still need education about health consequences of tobacco-use. In addition, Supari use has to be addressed in school-based tobacco prevention and cessation initiatives. Furthermore, programs must also address perceptions and norms related to peers and tobacco use and ensure active implementation of existing laws. Such integrated measures will help ensure tobacco-free spaces around schools.

        Evaluation of early versus delayed laparoscopic cholecystectomy in acute calculous cholecystitis: a prospective, randomized study

        Purpose: Uncertainty exists about whether early laparoscopic cholecystectomy (LC) is an appropriate surgical treatment for acute calculous cholecystitis. This study aimed to compare early vs. late LC for acute calculous cholecystitis regarding intraoperative difficulty and postoperative outcomes. Methods: This was a prospective randomized study carried out between December 2015 and June 2017; 60 patients with acute calculous cholecystitis were divided into two groups (early and delayed groups), each comprising 30 patients. Thirty patients treated with LC within 3 to 5 days of arrival at the hospital were assigned to the early group. The other 30 patients were placed in the delayed group, first treated conservatively, and followed by LC 3 to 6 weeks later. Results: The conversion rates in both groups were 6.7% and 0%, respectively (p = 0.143). The operating time was 56.67 ± 11.70 minutes in the early group and 75.67 ± 20.52 minutes in the delayed group (p = 0.001), and both groups observed equal levels of postoperative complications. Early LC patients, on the other hand, required much fewer postoperative hospital stay (3.40 ± 1.99 vs. 6.27 ± 2.90 days, p = 0.006). Conclusion: Considering shorter operative time and hospital stay without significant increase of open conversion rates, early LC might have benefits over late LC.

      • MSP1 triggers cell death and defense response in rice

        When the rice blast fungus attacks rice, fungal proteins are secreted into the plant apoplast to facilitate infection. The rice plant recognizes such secreted proteins, which result in the induction of defense responses. However, the molecular mechanisms of how rice plant recognizes secreted proteins remain elusive. Here, we report that a small, secreted protein, Magnaporthe oryzae snodprot1 homolog (MSP1), is recognized by rice plants and triggers host cell death and defense responses. Furthermore, pre-treatment of rice with Domain II, elicitor-active epitope of MSP1, induces resistance to the pathogen KJ301. We demonstrated that secretion of MSP1 into the apoplast is prerequisite for triggering cell death and activating defense-related gene expression, suggesting that it is recognized by a receptor in the host plasma membrane. Through comprehensively analysis of transcriptional profile in rice leaves and suspension cultured cells (SCCs) in response to exogenous MSP1 and Domain II treatment using 60K Agilent microarray chip, we found that 27 signaling genes, such as F-box(6), MAPK(4), protein kinase(11), transcription factor(6), were up-regulated in leaves and SCCs and six protein kinases were targeted into plasma membrane. Thus, we suggest that some of these genes may act as receptor of MSP1 in response to exogenous MSP1 treatment. Expression pattern of candidate genes was further checked in response to different environment cues using open rice data. These results demonstrate that these genes may be also involved in the signaling in response to cold stress, root-JA treatment and brown plant hopper (BPH) attack.
