        Genome Sequencing Highlights the Plant Cell Wall Degrading Capacity of Edible Mushroom Stropharia rugosoannulata

        The basidiomycetous edible mushroom Stropharia rugosoannulata has excellent nutrition, medicine, bioremediation, and biocontrol properties. S. rugosoannulata has been widely and easily cultivated using agricultural by-products showing strong lignocellulose degradation capacity. However, the unavailable high-quality genome information has hindered the research on gene function and molecular breeding of S. rugosoannulata. This study provided a high-quality genome assembly and annotation from S. rugosoannulata monokaryotic strain QGU27 based on combined Illumina-Nanopore data. The genome size was about 47.97 Mb and consisted of 20 scaffolds, with an N50 of 3.73 Mb and a GC content of 47.9%. The repetitive sequences accounted for 17.41% of the genome, mostly long terminal repeats (LTRs). A total of 15,726 coding gene sequences were putatively identified with the BUSCO score of 98.7%. There are 142 genes encoding plant cell wall degrading enzymes (PCWDEs) in the genome, and 52, 39, 30, 11, 8, and 2 genes related to lignin, cellulose, hemicellulose, pectin, chitin, and cutin degradation, respectively. Comparative genomic analysis revealed that S. rugosoannulata is superior in utilizing aldehyde-containing lignins and is possible to utilize algae during the cultivation.

        Prediction of aspect ratio of a micro hole drilled by EDM

        Micro electrical discharge machining (EDM) has the ability to drill micro holes with high accuracy in metallic materials. The aspect ratio of a micro hole generated by micro EDM is usually higher than those by other processes such as etching, mechanical drilling, and laser. However, it was found that the drilling speed of micro EDM slows down and even stops when the aspect ratio of a micro hole reaches a certain value. To understand this phenomenon, a theoretical model is proposed based on the fluid mechanics and surface tension. Experiments under different machining conditions are carried out to verify this model. Experimental results agree with the theoretical values, which indicate the validity of the proposed model. The difference between the theoretical values and the real values might be caused by debris, temperature and rotating of electrode in the discharge gap, which are ignored in the model.

        Hierarchical Al2O3/SiO2 fiber membrane with reversible wettability for on-demand oil/water separation

        This work presents an effective method of fabricating hierarchical Al2O3/SiO2 fiber membrane with reversible wettability for on-demand oil/water separation. In this strategy, the superhydrophilic/underwater superoleophobic surfaces are fabricated by in-situ growing hierarchical Al2O3 nanosheets on the SiO2 fiber surfaces that can be used as water-removing materials for oil/water separation. Then, the superhydrophobic/oleophilic surfaces are obtained by surface chemical modification with sodium laurate, which can be used as oil-removing materials for oil/water separation. Interestingly, the reversible wettability transformation of Al2O3/SiO2 fiber membrane can be controlled by the annealing and modification treatment alternately. The as-prepared Al2O3/SiO2 fiber membrane, combining the advantages and overcoming disadvantages of two modes, achieves reversible wettability transitions and on-demand oil/water separation. In addition, the Al2O3/SiO2 fiber membrane shows a significant chemical stability and super-wettability even after five annealing and surface modification cycles, indicating its excellent durability. The separation efficiency in both oil-removing mode and water-removing mode is over 95% for various oil/water mixtures through multiple recycle separation processes. This work not only provides a simple and cost-effective method to fabricate separation membrane with reversible wettability, but also shows great potential in remediation of large-scale oil spillage or organic solvents discharge at different environmental conditions.

        Genome-wide identification of the valine-glutamine motif containing gene family and the role of VQ25-1 in pollen germination in Brassica oleracea

        Background The plant-specific valine-glutamine (VQ) motif containing proteins tightly regulate plant growth, development, and stress responses. However, the genome-wide identification and functional analysis of Brassica oleracea (B. oleracea) VQ genes have not been reported. Objective To identify the VQ gene family in B. oleracea and analyze the function of Bo25-1 in pollen germination. Methods The Hidden Markov Model (HMM) of VQ family was used to query the BoVQ genes in the B. oleracea genome. The BoVQ genes preferentially expressed in anthers were screened by qRT-PCR. Subcellular localization of VQ25-1 was observed in Nicotiana benthamiana (N. benthamiana) leaves. To analysis the role of BoVQ25-1 in pollen germination, the expression of BoVQ25-1 was suppressed using antisense-oligonucleotides (AS-ODN). Results A total of 64 BoVQ genes were identified in the B. oleracea genome. BoVQ25-1 was found to be preferentially expressed in the B. oleracea anthers. BoVQ25-1 was cloned from the anthers of the B. oleracea cultivar ‘Fast Cycle’. BoVQ25-1 is localized to the nucleus. The pollen germination rate significantly decreased after AS-ODN treatment. Conclusion Sixty-four BoVQ genes were identified in the B. oleracea genome, of which BoVQ25-1 plays an important role in pollen germination.
