<P><B>Abstract.</B></P><P>We investigated the 47-kDa outer membrane protein (OMP), which is a periplasmic serine protease and an antigenic major surface protein of <I>Orientia tsutsugamushi</I>, as a vaccine candidate. We developed a conventional subunit vaccine expressing recombinant 47-kDa OMP (rec47) and a DNA vaccine (p47). In mouse immunization experiments, intranasal immunization with rec47 alone or with rec47 plus heat-labile enterotoxin B subunit from <I>Escherichia coli</I> or plus cholera toxin (CT) as adjuvants induced a higher amount of rec47-specific antibodies than intramuscular immunization with p47 alone or with p47 plus pBOOST2-samIRF7/3 (pB) as adjuvant. Moreover, the combination of rec47 and CT induced a strong cellular immune response to 47-kDa OMP, as demonstrated by a spleen cell proliferation assay, and also induced Th1- and Th2-type cytokine production, as demonstrated by a cytokine enzyme-linked immunosorbent assay. Intranasal immunization with rec47 plus CT was the most effective method for the induction of humoral and cell-mediated immune responses. Furthermore, relatively strong protection against homologous <I>O. tsutsugamushi</I> strain Boryong challenge was observed in mice immunized with rec47 plus CT. Therefore, 47-kDa OMP is an attractive candidate for developing a prophylactic vaccine against scrub typhus by <I>O. tsutsugamushi</I> infection.</P>

Predictive Factors for the Mortality of Cardiovascular Patients at Coronary Care Unit

BACKGROUND: Recently the incidence of coronary artery disease has been increased rapidly in Korea. After the introduction of coronary care unit, the mortality rate of cardiovascular patients has been decreased. The predictive factors for mortality in patients admitted at Coronary Care Unit (CCU) are important in the management of acutely ill cardiovascular patients. METHODS: One thousand one hundred and thirty patients (64.8+/-14.5 years), who were admitted at CCU from January 2002 to June 2003, were analyzed. The patients were divided into two groups according to mortality: the survived group (Group I: n=1055, 63.3+/-13.3 years) and the moribund group (Group II: n=75, 64.8+/-14.1 years). Clinical characteristics, risk factors, clinical diagnosis, laboratory, echocardiographic and coronary angiographic findings were compared between the two groups. RESULTS: The overall mortality at CCU was 6.6%, 75 out of 1130 patients. Age and sex were not different between both groups. Coronary artery disease was the most common cause of admission (886 out of 1130 patients) and death (46 out of 75 patients). Coronary angiographic findings were not different between the two groups. Protective Immunity of 56-kDa Type-Specific Antigen of Orientia tsutsugamushi Causing Scrub Typhus

Scrub typhus, caused by infection with Orientia tsutsugamushi, is a mite-borne zoonotic disease endemic to the Asian-Pacific region. In Korea, the incidence of this disease has increased with climate changes, and over 10,000 cases of infection were reported in 2013. Although this infection is treatable with antibiotics such as doxycycline and azithromycin, an effective prophylactic vaccine against O. tsutsugamushi would be more desirable for preventing scrub typhus in endemic areas. In this study, we investigated the 56-kDa type-specific antigen (TSA56), which is a major outer membrane protein of O. tsutsugamushi, as a vaccine candidate. Intranasal immunization of recombinant TSA56 (rec56) induced a higher level of TSA56- specific IgG than that induced by intramuscular immunization of tsa56-expressing DNA (p56). Both types of immunization induced a cell-mediated immune response to TSA56, as demonstrated by the splenic cell proliferation assay. Mice immunized with p56, followed by rec56 plus heat-labile enterotoxin B subunit from E. coli, had a stronger protection from a homologous challenge with the O. tsutsugamushi Boryong strain than with other combinations. Our preliminary results suggest that an effective human vaccine for scrub typhus can include either recombinant TSA56 protein or tsa56-expressing DNA, and provide the basis for further studies to optimize vaccine performance using additional antigens or different adjuvants.

32인치 WXGA급 (해상도 1366$\times$768) LCD-TV용 백라이트를 구성하는 외부전극 형광램프 및 각 광학 부품들에 대한 휘도 특성을 조사하고, 광학시트들의 빛의 투과율을 계측하였다. 12등식과 18 등식의 백라이트에 대한 휘도 특성을 조사하였다. 휘도 균일도는 12등식 백라이트에서도 85 %의 휘도 균일도를 갖지만, 광학 부품들의 휘도 값과 백라이트의 효율은 18 등식의 백라이트에 비하여 낮다. 외부전극 형광램프 12 등식과 18 등식의 백라이트에 대하여 램프의 휘도를 100 %로 규격화하면, 확산판, 확산시트, 프리즘시트 (BEF) 및 편광시트 (DBEF)의 상대 휘도는 램프의 휘도 값과 무관하게 정량화된다. 광학 부품의 상대 휘도 값은 12등식은 18등식보다 낮다. 백라이트의 휘도 100 %에 대한 액정 패널의 투과도는 DBEF와 BEF를 사용한백라이트는 7.14 %, DBEF만을 사용하면 6.12 %, 그리고 BEF만을 사용하면 3.21 %이다. 본 실험에서 얻어진 광학 부품들의 상대 휘도의 정량화 데이터와 액정의 투과도는 백라이트 설계를 위한 기초 자료이다. Luminance properties of external electrode fluorescent lamps and optical components in backlight unit (BLU) and optical transmission rates of optical sheets, are investigated for LCD-TV of 32" in diagonal with WXGA level resolution (1366$\times$768). The luminance is measured in 12-lamps and 18-lamps of BLU. The luminance uniformity preserves about 85 % in the 12-lamps backlight, while the luminance of optical components and the efficiency of backlight in the 12-lamps are lower than those in the 18-lamps backlight. When the lamp luminance in BLU having 12-lamps and 18-lamps is normalized as 100 %, the relative luminance of a diffusion plate, a diffusion sheet, a prism sheet (BEF), a polarization sheet (DBEF), has a constant value without dependence on a lamp luminance. The relative luminance of optical components in 12-lamps BLU is lower than that in 18-lamps backlight. The light transmission rate, the relative luminance of liquid crystal display panel with the luminance 100 % of backlight, is 7.14 % in the use of DBEF and BEF, 6.12 % in the use of only DBEF, and 3.21 % in the use of only BEF. Those Data obtained in this experiment for the lamps and optical components, are the design parameters for the LCD backlight.
