      • 器內 ??-線 處理에 의한 배 突然變異 誘起 및 選拔

        배 器內生育 중인 新稍에 ??-선을 처리하여 돌연변이를 유기하고 여기서 黑斑病 抵抗性 變異體를 선발하는 시험을 실시하였다. 적정 ??-선 처리 線量(LD??)은 新水와 二十世紀 품종 공히 6krad는 과다하고, 4krad는 약간 낮은 감이 있었다. γ-선 처리선량이 높아지면 신초 및 腋芽의 증식 배수가 줄어들었다. 기내 생육 일을 이용한 흑반병 저항성 조기 검정법으로서, 菌絲배양에 의해 얻어진 독소액으로 검정하면 반응이 없었으나, 균사를 접종하면 병반이 발생했다. 기내신초를 마사토에 삽목하여 발근하면 기내에서 발근하는 것보다 發根率이 더 높아졌으며, 높은 감마선 선량이 照射된 신초는 발근율이 낮았다. 포장에서 생육한 ??-선 처리 신수 품종의 나무는 접목묘에 비해 樹高와 신초 발생수에 있어 차이가 있었으나 이십세기 품종은 차이를 보이지 않았다. 신수 품종 11계통 중 2계통과 이십세기 품종 22계통 중 1계통이 실내독소 검정과 포장의 생육 잎의 관찰에서 저항성으로 나타났다. In vitro shoots were irradiated with gamma rays to induce mutants resistant to pear black spot disease(Alternaria kikuchiana Tanka) from oriental pear cultivars, Shinsui and Nijisseiki. ??(50% lethal dose) of in vitro shoots was about 5 krad between 4 and 6 krad in two tested cultivars. In vitro shoots obtained after 4∼5 subcultures were more well rooted in ex vitro than in vitro. Rooting rate of shoots decreased in the treatment of gamma-ray of high dose. For the early test of resistance to pear black spot disease, AK toxin inoculation on in vitro leaf was not desirable for little response, but the inoculation of hypha on in vitro leaf has a possibility as it developed necrotic spots. In vitro irradiated trees grown for 2 years in the field showed shorter in the plant height and more branching than non-irradiated trees. After grown in the field for 2 years, AK toxin test of leaf and observations of leaf at the field showed that 2 lines of Shinsui and 1 line of Nijisseiki were resistant to black spot disease.

      • 배 기내 신초의 증식과 기내 및 기외 발근

        Meristems(0.6-0.9mm) obtained from shoot tips of pear were cultured on MS media containing various concentrations of BA to induce multiple shoots and multiplicated shoot was rooted in vitro and ex vitro using some kinds of methods. About 50% inoculated meristems were survived in the first culture. Medium containing 1∼2mg/L BA as a growth regulator was the most suitable for multiplication of in vitro shoots. Rooting was poor in vitro, while ex vitro rooting were effective when irrigating 2ml/L concentration EM solution. Rooted shoots grew up to about 90cm during one growing season.

      • 무, 배추 및 갓의 원형질체의 나출과 융합

        This study was carried out to get fundamental information for the production of somatic hybrids among Raphanus sativus, Brsssica campestris ssp. pekinensis and Brassics juncea. 1. Enzyme mixture of 1∼2% Cellulase R-10 with CPW, 0.5% Macerozyme, 0.7M mannitol and pH 5.8 was suitable for protoplast isolation from all the materials. Incubation time for 6 hrs. was best for the species to isolate protoplasts from plastic house cultured young leaves. Optimal concentration of mannitol was 0.7M for R. sativus and Brassica juncea but 0.6M for B. campestris ssp. pekinensis when the protoplasts were isolated from leaf mesophyll. pH 5.8 was adequate for protoplast isolation of the three species. 2. Source materials used were in vitro cultured cotyledons and leaves, plastic house cultured cotyledons and young leaves. In vitro cultured leaf mesophyll was the most suitable to get the protoplasts of Brassica campestris ssp. pekinensis. As in vitro cultured leaf was not used for the isolation of protoplast of R sativus and B. juncea, protoplast yield was high from plastic house cultured young leaf mesophyll. 3. The rate of heteroplasmic fusion was 12% from R. sativus + B. campestris ssp. pekinensis, 14% from B. campestris ssp. pekinensis + B. juncea, and 11% from R. sativus + B. juncea.

      • 경단 절편 배양에 의한 바나나 급속 증식

        바나나의 흡아에서 채취한 정단, 경단의 1/2절편, 그리고 1/9절편을 배양한 결과 BA를 5mg/ℓ를 첨가한M 바나나 흡아의 경단을 2등분 한 것, 6등분한 것, 그리고 경정을 BA가 다른 농도별로 첨가된 MS배지에 배양하여 효과적인 multiple shoot를 유기하기 위한 방법을 개발하였다. 부정아가 9등분한 경단절편에서 가장 많이 형성되었으며 이들 부정아는 엽초와 엽초사이 조직에서 발생되었다. 경단 절편배양에서 배지에 첨가된 BA 농도가 5mg/ℓ일 때 경단 절편으로부터 발생한 부정아의 수가 절편당 6.3개로 가장 많았으며 부정아의 생장도 빨랐다. 발생한 부정아로부터 얻어진 신초는 계대배양에 의해 증식할 수 있었으며, 계대배양 회수가 늘어남에 따라 배양한 신초당 다시 발생한 부정아 및 신초의 수는 더 많아졌다. 발근된 묘는 순화후 온실에서 2개월 뒤에 30∼40cm 크기로 생장했다. The shoot tips, and 1/2 and 1/9 shoot apex segments obtained from suckers of banana were cultured on MS media containing 0.0, 0.5, 1.0, 3.0, 5.0 and 7.0mg/ℓ BA, respectively, to induce multiple adventitious shoots in vitro. 6.2 adventitious shoots per segment were developed from 1/9 shoot apex segment when cultured on MS media containing 5mg/ℓ BA for 4 weeks. The number of developed adventitious buds and their growth were best on MS medium added with 5.0mg/ℓ BA. By progressing subculture, the number of developed shoots per explant increased gradually. Rooted plantlets grew up to 30∼40cm in 2 months in the plastic house.

        A methodology of using the first consonant character of korean language for operation of various instruments was developed. Adding the first consonant character of Hangul on numeric buttons of various electric products such as phone dialing, door key and remote controller, we can use them more conveniently then before. First, instead of using old fashioned system of dialing number to find company or institute, we can simply use the first consonant character(ㄱ, ㄴ, ㄷ, ...., ㅎ) of Hangul character. We can also convert a numeric telephone number into meaningful words or phrase, adding vowel or vowel and consonants to the first consonant characters properly. Then we can memorize the converted words or phrase instead of memorizing telephone number. Second, we can extend telephone functions in phone number memory, ARS or beeper system. Third, we can also apply this methodology to door key system which unlock the door by accepting numeric password. With this method, we can unlock the door by pressing the first consonant of passwords which used to numbers. This methodology will make our life better, as it is easy to apply this new system to various electric instruments such as air conditioner, TV, video player, etc., which all have numeric control system to. Thus, all of the products with the first consonants of Hangul can be controlled by the first consonant pressing system which presses the first consonant of words or phrase meaning their functions.

      • 배 기내 증식 배지의 질소원 종류별 신초 생육

        This study was carried out to know the optimum concentration of NH_(4)NO_(3) and KNO_(3) added MS media for the culture of Oriental pear (Pyrus pyrifolia) in vitro. Meristems (0.6-0.9mm) obtained from axillary bud of pear were cultured on MS media containing 3mg/L BA for establishing shoots in vitro. The initial culture of meristem on the solid media was better than that on the liquid media at the point of view of shooting and vitrification of culture. The best result was obtained from the cultures in the MS media containing 20mM NH_(4)NO_(3) and 20mM KNO_(3) as the nitrogen sources. Fresh weight and shoot length obtained were highest after 5 week culture in the media containing 20mM NH_(4)NO_(3) and 20mM KNO_(3) in the culture of Oriental pear in vitro.

      • 배에 발생하는 괴저반점병과 이상반점 증상의 구별

        괴저반점병과 이상반점 증상은 거의 동일한 시기에 발생하였는데, 병반이 5월 중순경에 나타나기 시작하여 6월 중순에 가장 많이 발생하고 그 후 감소하다가 고온기인 8월에는 거의 나타나지 않았다. 괴저반점병의 병반 형태는 부정다각형이지만 이상반점 증상은 원형이었다. 발생초기에 괴저반점병은 흑색, 이상반점증상은 적자색이지만 둘 다 진전되면 회백색으로 되었다. 두 병의 병반은 발육지의 하부의 중간부위 잎에서 가장 많이 발생하고 발육지의 상부의 잎에 거의 발생하지 않으나, 괴저반점병의 병반은 이상반점 증상의 것보다 과총잎과 발육지의 기부 잎에 더 많이 발생하였다. 괴저반점병은 실생 개체에서 발생하지 않으나 이상반점 증상은 실생에서도 발생하였다. 이상반점 증상 발생나무에 괴저반점병 지표품종 HN-39를 고접해도 괴저반점병 증상은 없었다. 괴저반점병은 현재 많은 농가에서 발생하고 있으나 이상반점 증상은 1996년과 1997년 사이의 조사에 의하면 이병잎의 비율이나 병반수가 현저히 줄어들고 있다. This study was carried out to make a distinction between pear necrotic spot virus(PNSV) known as a virus disease and PNSV-like disease called as abnormal spot disease (ASD) in pear (Pyrus pyrifolia Nakai). Two diseases, PNSV and ASD, developed symptoms on the leaves of shoot at almost same time. They started to develop their spots on the middle part of May, and showed the peak on the middle part of June and decreased on the middle part of July. The shapes of spots were irregular rectangle and polygon in the symptom of PNSV, while a circular shape in ASD. In the early stage of formation of spots, PNSV developed generally black rectangular spots, but ASD developed red-purple and circular spots on the leaf of shoots. Distribution of diseased leaves on the shoots showed very similar pattern in the point that they formed diseased spots most at leaves of middle-low part of shoots from both PNSV and ASD, and that they little developed disease spots on the above leaves of shoots. Observing seedlings, there were no PNSV symptoms in all the seedling trees, while there were found ASD symptoms in most of seedlings. In the recent year, there was observed a obvious decrease in occurrence of ASD. But PNSV is prevailing on the pear orchards where cultivar 'Niitaka' was planted or Niitaka was high-grafted on the old cultivars, Chojuro and Okusankich for renewal of cultivars.
