      • Divergent tree growth response to recent climate warming of <i>Abies faxoniana</i> at alpine treelines in east edge of Tibetan Plateau

        <P><B>Abstract</B></P><P>An anomalous reduction in tree growth indices has been detected in tree ring records from many circumpolar northern latitude sites in recent years, which was known as the divergence phenomenon. In order to understand whether the divergence phenomenon appeared at altitudinal treelines in east edge of Tibetan Plateau, we analyzed the correlation coefficients between tree growth and climate variables before and after recent climate warming, respectively. The results showed that divergence phenomenon took place and varied with sites. Compared with those in 1955–1982, the correlation coefficient of growth to mean temperature in June declined during 1983–2012. In Songpan site, more than half of the sampled trees showed an increasing positive correlation with temperature from July to September, while others turned to be a negative correlation. The positive responses to the mean temperature in previous‐year November and December declined in Markang site. Only in Miyaluo site, the positive correlation between tree ring index and temperature from July to September increased. Both the anomalous reduction or disappearance of the responses to the mean temperature and the divergent growth responses suggested that the divergence phenomenon was universal at altitudinal treelines in east edge of Tibetan Plateau, which had potentially significant influences on the estimation of forest productivity and climatic reconstructions based on tree ring.</P>

        Performance and Mechanism Analysis of Succinate Production under Different Transporters in Escherichia coli

        Succinic acid is a platform chemical with potential for bio-based synthesis. However, the production of bio-based succinate is limited because of insufficient succinate efflux capacity in the late stage of fermentation. In the present study, three different transporters, which have been reported to be responsible for C4-dicarboxylates transport, were employed for investigation of the transport capacity of succinate in Escherichia coli. After engineered strains were constructed, the fermentative production of succinic acid was studied in serum bottles and 3 L of fermentor. The results demonstrated that engineered strain showed better efflux capacity than control strain under high concentration of succinate. The highest production of succinate was 68.66 g/L, while the NCgl2130 transporter may be the best candidate for succinate export in E. coli. Further research showed that the expression levels and relative enzyme activities involved in the metabolic pathway all increased markedly, and the maximum activities of PPC, PCK, PYK, and MDH increased by 1.50, 1.38, 1.28, and 1.27-fold in recombinant E. coli AFP111/pTrc99a- NCgl2130, respectively. Moreover, the maximum level of intracellular ATP increased by 23.79% in E. coli AFP111/ pTrc99a-NCgl2130. Taken together, these findings indicated that engineered transporters can improve succinate production by increasing key enzyme activities and intracellular ATP levels. To the best of thew authors’ knowledge, this is the first report on a mechanism to improve succinate production by engineered transporters. This strategy set up a foundation for improving the biosynthesis of other C4-dicarboxylates, such as fumaric acid and malic acid.

        Amygdalin inhibits HSC-T6 cell proliferation and fibrosis through the regulation of TGF-β/CTGF

        Amygdalin is one of the nitrilosides that was widely used to treat cancer, inhibit liver fibrosis. In the present study, the aim was to determine the antifibrotic potential of amygdalin and examine its mechanisms of action in vitro. Firstly, we found amygdalin significantly inhibited HSC-T6 cells proliferation. Both mRNA and protein of transforming growth factor-β (TGF-β) were decreased in HSC-T6 cells during amygdalin treatment. Secondly, to investigate functional role of TGF-β, both TGF-β over-expression vector and siRNA against TGF-β were transfected into HSC-T6 cells respectively. The results showed that over-expression of TGF-β promoted proliferation of HSC-T6 cells, whereas TGF-β knockdown inhibited cell viability. Moreover, our data even indicated that TGF-β could promote cell proliferation independent of amygdalin treatment. Finally, we found amygdalin could inhibit expression of the classical fibrotic factor αSMA, which suggested an antifibrotic effect of amygdalin. While the TGF-β antagonized anti-fibrotic effect of amygdalin. To assess the mechanisms, we examined expression of CTGF in cultured HSC-T6 cells. Our results showed that CTGF was down-regulated in HSCT6 cell treated by amygdalin, but was up-regulated when exogenous TGF-β introduced. As CTGF was one of the downstream factors in the TGF-β pathway. These might suggest that amygdalin inhibited HSC-T6 cells proliferation and fibrosis via TGF-β/CTGF pathway.

        Nutrigenomics reveals potential genetic underpinning of diverse taste preference of Chinese men

        Background Taste preference varies geographically in China. However, studies on Chinese people’s taste preference in different regions of China are limited, and are lack of research on the mechanism of diferences in taste preference, especially in genetics. Objective This study aims to investigate the characteristics of taste preference of Chinese men, and estimate whether diverse taste preference in Chinese have genetic underpinning. Methods We conducted a questionnaire survey on taste preferences on 1076 males from 10 regions of China, and collected another 1427 males from the same regions which genotyped by microarray. We compared the correlation between diferent taste preference, and evaluated the correlation between the mutation frequency of inhouse database and diferent taste preference. The putative taste-preference-related genes were further utilized to estimate the candidate relationship on gene and gene network in diferent taste preference. Results There was a correlation between diferent taste preferences in Chinese men. We found 31 SNPs associated with 6 kind of taste preferences. These SNPs located within or nearby 36 genes, and the tastes associated with 4 of these genes (TRPV1, AGT, ASIC2 and GLP1R) are consistent with the previous studies. Moreover, in diferent tastes which were suggested to be associated with each other, some putative related genes were the same or in the same gene network, such as pathways related with blood pressure, response to stimulus and nervous system. Conclusions This study indicates that the diverse taste preference of Chinese men may have genetic underpinning.
