Metallothioneins are a class of small cysteine-rich proteins that have been associated with increased tolerance to metal and oxidative stresses in animals, plants, and fungi. We investigated a metallothionein-like (mt-like) gene shown previously to be upregulated in fruiting bodies of the fungus Agaricus bisporus in response to post-harvest storage. Analysis of an A. bisporus genomic DNA cosmid library identified two similar mt-like genes (met1 and met2) arranged as a bidirectional gene pair transcribed from the same promoter region. The promoter contained regulatory elements including 9 metal responsive elements and a CAAT box region 220bp downstream of met1 that showed striking similarity to a feature in Coprinopsis cinerea mt-like gene promoters. Transcriptional analysis showed that both met genes are significantly and rapidly (within 3 hours) upregulated during post-harvest storage and expression is significantly greater in stipe and cap tissues compared with the gills. However, a strong directionality of the promoter was demonstrated, as transcript levels of met1 were at least two orders of magnitude greater than those of met2 in all samples tested.

Chronic liver diseases are worldwide health issues. The causes for chronic liver disease include hepatitis viral infection, chronic alcohol consumption, environmental pollutants and cryptogenic factors. Hepatocellular carcinoma (HCC) is the most common primary cancer of liver. It is responsible for approximately one million deaths per year. HCC has been a malignancy of the underdeveloped and developing countries but the incidence in the developed countries is on the rise, Irrespective of its causative agents, cirrhosis per se is known to predispose an individual to the development of HCC. Eighty to ninety percent of HCC cases are developed on a background of liver cirrhosis. However, the molecular basis of pathogenesis of liver cirrhosis and HCC are not well known. Using two-dimensional gel electrophoresis and ProteinChip (arrays system, we have generated panels of "protein patterns" from normal and diseased samples. In combination with advanced pattern recognition based bioinformatic tools, we are expecting to classify liver diseases based on unique protein patterns. Differentially expressed proteins will be identified and thus be used for providing insights in molecular pathogenesis of liver diseases.

Few butterflies are pests of economic significance, but some may be locally destructive, such as the papilionid Graphium agamemnon Linnaeus, which is known to feed on the commercially important soursop (Annona muricata Linnaeus) in Vietnam. This paper documents the life history and ecology of G. agamemnonand investigates commonly used control measures in south-east Vietnam. A novel method of controllingG. agamemnon infestation is described. If soursop basal rootstock (Annona glabra Linnaeus) is encouraged to shoot during times of peak butterfly activity, ovipositing G. agamemnon females are preferentially attracted to the new growth where the resulting early stages may be easily detected and removed by hand.

The fig wax scale Ceroplastes rusci (Linnaeus) is a serious pest of fruit trees in many countries. In the present study we investigated the extent of C. rusci infestation and the range of host species, particularly in fruit orchards, in south-east Vietnam. Captive populations of C. rusci were established to record life history parameters and to investigate the efficacy of an endemic predacious moth, Eublemma amabilis Moore, as a potential biocontrol agent. Heavy infestation of C. rusci (up to 100%) was recorded in soursop and other fruit orchards, and the culturally important Hoa Mai flower was also affected. Captive breeding trials found E. amabilis to be an efficient predator of C. rusci, but an unusual hyperactive trait in early instars of E. amabilis resulted in lower than expected survival rates.The implications of this trait in terms of the laboratory environment, augmentative release protocols and as a survival strategy are discussed.

Young plants of the commercially important timber species Hopea odorata Roxb. are seriously damaged by the psyllid Trioza hopeae Burckhardt & Vu in nurseries in South Central and Southern Vietnam. Farmers routinely use Class 1 toxic insecticides to control the psyllids but these pose serious health risks in the urban environment. As a precursor to developing a pest management strategy, we studied the life cycle of T. hopeae and trialed several control measures. Trioza hopeae is polyvoltine with overlapping generations peaking in mid wet season. Females laid approximately 135 eggs; however, there was a natural attrition rate of about 80% in first-instar nymphs due to the nymph’s inability to find a suitable feeding site. Feeding nymphs induced pit galls on young H. odorata leaves and were present on all trees examined during the study. The pre-adult life cycle lasted approximately 13 days and adult life span approximately seven days. Several toxic insecticides were found to be effective for controlling T. hopeae, but a novel non-toxic alternative was equally as effective.We discuss these methods and potential biological control measures.

Objectives: Obstructive sleep apnea (OSA) increases the risk of perioperative adverse events in children. While polysomnography (PSG) remains the reference standard for OSA diagnosis, oximetry is a valuable screening tool. The traditional practice is the manual analysis of desaturation clusters derived from a tabletop device using the McGill oximetry score. However, automated analysis of wearable oximetry data can be an alternative. This study investigated the accuracy of wrist-worn oximetry with automated analysis as a preoperative OSA screening tool. Methods: Healthy children scheduled for adenotonsillectomy underwent concurrent overnight PSG and wrist-worn oximetry. PSG determined the obstructive apnea-hypopnea index (OAHI). Oximetry data were auto-analyzed to determine 3% oxygen desaturation index (ODI3) and visually scored as per McGill criteria. The logistic regression model assessed the predictive performance of ODI3 for detecting the presence and severity of OSA after adjusting for covariates. Results: Seventy-six children (34 females), aged (mean±standard deviation) 5.7±1.6 years were classified, based on PSG-derived OAHI, as no OSA (n=31), mild (n=31), and moderate-severe OSA (n=14). Oximetric ODI3 was identified as the sole predictor of moderate-severe OSA (OAHI≥5 events/h) (odds ratio 1.38, 95% confidence interval 1.15, 1.65, <i>p</i>=0.001). The best diagnostic performance was at ODI3=5 events/h (78.6% sensitivity, 75.8% specificity [receiver operating characteristic-area under the curve {ROC-AUC}=0.857]). ODI3 was also more sensitive than the McGill oximetry score in diagnosing moderate-severe OSA (78.6% by ODI3 vs. 33.0% by McGill). The performance was suboptimal for any level of OSA (OAHI≥1 event/h) (75.6% sensitivity, 61.3% specificity [ROC-AUC=0.709]). Conclusions: Wrist-worn oximetry-derived automated ODI3 can reliably identify moderate-severe OSA in children undergoing adenotonsillectomy, making it a potentially useful preoperative OSA screening tool.
