        Fatigue Resistance of a BFRP-Encapsulated Long-Gauge FBG Strain Sensor under Cyclic Train Loads

        To verify the performance of a basalt fiber reinforced polymer (BFRP) fiber-encapsulated long-gauge strain sensor in railway bridge health monitoring, this paper studies the fatigue resistance and durability of the BFRP fiber-encapsulated FBG sensor under train loads. First, the influences of the length of the anchorage section and the length ratio of the sensing section on the accuracy of the sensor were studied. Then, the BFRP sensor was applied to a sleeper for 2 million cycles of tension fatigue testing. The strain-time history of the whole fatigue test was monitored and compared. After the test, a calibration test was carried out to verify the accuracy and repeatability of the sensor. Finally, the slip and fatigue cracking of the fiber in the anchorage section of the sensor were observed by electron microscopy. The results show that the gap between the anchoring section and the bare optical fiber was filled with epoxy resin, and there was no slip behavior. No fatigue cracking occurred in the fiber, and the straincoefficient and linearity of the sensor showed no obvious changes after 2 million cycles of loading. The long-gauge strain sensor encapsulated by BFRP fibers exhibited good fatigue resistance and can meet long-term monitoring requirements under train loads.

        Efficacy and Safety of the Safe Triangular Working Zone Approach in Percutaneous Vertebroplasty for Spinal Metastasis

        Objective: This study aimed to assess the technical feasibility, efficacy, and safety of the safe triangular working zone (STWZ) approach applied in percutaneous vertebroplasty (PV) for spinal metastases involving the posterior part of the vertebral body. Materials and Methods: We prospectively enrolled 87 patients who underwent PV for spinal metastasis involving the posterior part of the vertebral body, with or without the STWZ approach, from January 2019 to April 2022. Forty-nine patients (27 females and 22 males; mean age ± standard deviation [SD], 57.2 ± 11.6 years; age range, 31–76 years) were included in group A (with STWZ approach), accounting for 54 vertebrae. Thirty-eight patients (18 females and 20 males; 59.1 ± 10.9 years; 29–81 years) were included in group B (without STWZ approach), accounting for 57 vertebrae. Patient demographics, procedure-related variables, and pain relief as assessed using the visual analog scale (VAS) were collected at different time points. Tumor recurrence in the vertebrae after PV was analyzed using Kaplan–Meier curves. Results: The STWZ approach was successful from T1 to L5 without severe complications. Cement filling was satisfactory in 47/54 (87.0%) and 25/57 (43.9%) vertebrae in groups A and B, respectively (p < 0.001). Cement leakage was not significantly different between groups A and B (p = 1.000). Mean VAS score ± SD before and 1 week and 1, 3, 6, 9, and 12 months after PV were 7.6 ± 1.8, 4.2 ± 2.0, 2.7 ± 1.9, 1.9 ± 1.5, 1.7 ± 1.4, 1.7 ± 1.1, and 1.6 ± 1.3, respectively, in group A and 7.2 ± 1.7, 4.0 ± 1.3, 3.4 ± 1.6, 2.4 ± 1.2, 1.8 ± 1.0, 1.4 ± 0.5, and 1.7 ± 0.9, respectively, in group B. Kaplan–Meier analysis showed a lower tumor recurrence rate in group A than in group B (p = 0.001). Conclusion: The STWZ approach may represent a new, safe, alternative/auxiliary approach to target the posterior part of the vertebral body in the PV for spinal metastases.

      • IL-12 Regulates B7-H1 Expression in Ovarian Cancer-associated Macrophages by Effects on NF-κB Signalling

        Background and Aim: B7-H1, a co-inhibitory molecule of the B7 family, is found aberrantly expressed in ovarian cancer cells and infiltrating macrophage/dendritic-like cells, and plays a critical role in immune evasion by ovarian cancer. IL-12, an inducer of Th1 cell development, exerts immunomodulatory effects on ovarian cancer. However, whether IL-12 regulates B7-H1 expression in human ovarian cancer associated-macrophages has not been clarified. Therefore, we investigated the effects of IL-12 on the expression of B7-H1 in ovarian cancer-associated macrophages and possible mechanisms. Methods: PMA induced THP-1-derived macrophages or human monocyte-derived macrophages were treated with recombinant IL-12 (rIL-12) or infected with adenovirus carrying human IL-12 gene (Ad-IL-12-GFP) for 24 h, then cocultured with the SKOV3 ovarian cancer cell line for another 24 h. Macrophages were collected for real-time PCR and Western blot to detect the expression of B7-H1, and activation of the NF-${\kappa}B$ signaling pathway. Moreover, supernatants were collected to assay for IL-12, IFN-${\gamma}$ and IL-10 by ELISA. In addition, monocyte-derived macrophages treated with IFN-${\gamma}$ were cocultured with SKOV3 and determined for the expression of B7-H1. Furthermore, the expression of B7-H1 in monocyte-derived macrophages was also evaluated after blocking NF-${\kappa}B$ signaling. Results: The expression of B7-H1 was significantly upregulated in monocyte-derived macrophages treated with rIL-12 or Ad-IL-12-GFP compared with the control groups (p<0.05), accompanied by a remarkable upregulation of IFN-${\gamma}$ (p<0.05), a marked downregulation of IL-10 (p<0.05) and activation of NF-${\kappa}B$ signaling. However, the upregulation of B7-H1 was inhibited by blocking the NF-${\kappa}B$ signaling pathway (p<0.05). Expression of B7-H1 was also increased (p<0.05) in monocyte-derived macrophages treated with IFN-${\gamma}$ and cocultured with SKOV3. By contrast, the expression of B7-H1 in THP-1-derived macrophages was significantly decreased when treated in the same way as monocyte-derived macrophages (p<0.05), and IL-10 was also significantly decreased but IFN-${\gamma}$ was almost absent. Conclusions: IL-12 upregulates the expression of B7-H1 in monocyte-derived macrophages, which is possible though inducing the secretion of IFN-${\gamma}$ and further activating the NF-${\kappa}B$ signal pathway. However, IL-12 downregulates the expression of B7-H1 in THP-1-derived macrophages, associated with a lack of IFN-${\gamma}$ and inhibition of expression of IL-10.

        Prediction and Monitoring of the Construction Vibration Effect on an Adjacent Old Long Span Double-Convex Arch Bridge

        Ground vibrations caused by the construction of impact drilling piles may affect adjacent bridges, so corresponding prediction and monitoring are necessary to ensure the safety of bridges. In this study, the construction near an existing long-span double-convex arch bridge is taken as a case study to investigate different layouts of construction machines emanating harmful ground vibrations to the existing adjacent bridge by numerical simulations and monitoring. The peak acceleration and velocity acquired from numerical simulations, and monitoring were successfully used to implement impact drilling pile construction near the existing bridge. The results show that there are good consistencies between the monitoring and numerical simulation results, and the vibration acceleration of the foundation of abutments and piers are mainly derived from the vibration caused by the construction within a distance of 50 m, while the construction beyond 50 m has little effect on the old bridge. The numbers and locations of machines near the same foundation may have a weakening or strengthening effect on the peak acceleration and velocity due to destructive and constructive interference of waves, which can help to control the effects of the vibrations of the existing adjacent bridge.
