Aims: Liver cancer stem cells represent a small fraction of cells in liver cancer tissues so that studying these cells is very hard. Generation of liver cancer stem cells considered as one of the most important issue in cancer biology research. For this reason, we tried to generate liver cancer stem cells from induced pluripotent stem (iPSCs). Methods: First of all, CM was collected from confluent culture of Huh7 cells. Then, mouse iPSCs cells without MEF feeder cells were cultured in the presence of 50% CM for 4 weeks. The medium was changed every day with fresh medium containing 50% of CM. Mouse iPSCs cultured is the complete medium with LIF were used as a control. The survived cells (5x105 cells) were suspended in HBSS and injected into the liver of BALB/ c nude mice. After 25 days malignant tumor was formed in the liver while benign teratoma was formed by the injection of iPSCs. Tumors were then excised and partly fixed in 10% neutral formalin buffer solution for HE staining and immunohistochemical analysis. The rest of tumors were subjected to rt-qPCR anaylsis and primary culture. Results: Immunohistochemical analysis with liver cancer associated markers and cancer stem cell marker showed that malignant liver tumor was developed. These results indicate that the primary cells from the malignant tumor are rich in CSCs. Conclusions: This model will be very important and useful to assess the significant molecular mechanisms necessary to maintain liver cancer stem cells, which will help in defat liver cancer.

Dynamic behavior of TLP's supporting 5-MW wind turbines under multi-directional waves
Abou-Rayan, Ashraf M.,Khalil, Nader N.,Afify, Mohamed S. Techno-Press 2016 Ocean systems engineering Vol.6 No.2
Over recent years the offshore wind turbines are becoming more feasible solution to the energy problem, which is crucial for Egypt. In this article a three floating support structure, tension leg platform types (TLP), for 5-MW wind turbine have been considered. The dynamic behavior of a triangular, square, and pentagon TLP configurations under multi-directional regular and random waves have been investigated. The environmental loads have been considered according to the Egyptian Metrological Authority records in northern Red sea zone. The dynamic analysis were carried out using ANSYS-AQWA a finite element analysis software, FAST a wind turbine dynamic software, and MATLAB software. Investigation results give a better understanding of dynamical behavior and stability of the floating wind turbines. Results include time history, Power Spectrum densities (PSD's), and plan stability for all configurations.

Aims: Cancer stem cells (CSCs), also known as tumor-initiating cells (TICs), are suggested to be responsible for drug resistance and cancer recurrence. Current treatments with conventional chemotherapy are not highly efficient against the cancer stem cells (CSCs). The combination of anticancer drugs, of which functions are different from the other, enhances efficiency compared to the mono-therapy because it targets cancer cells in a synergistic or an additive manner. In this study, the effect of paclitaxel and sorafenib on cancer stem cells (CSCs) developed from mouse iPSCs in very low concentration was evaluated. Methods: To investigate the effect of combination therapy, CSCs were exposed to paclitaxel and/or sorafenib at different concentrations of 1, 2 and 4 nM, respectively. Cell viability was assessed with 3-(4,5-dimethylthiazolyl)-2,5-diphenyltetrazolium bromide (MTT). The same concentrations of the agents were assessed for the effect on the self-renewal potential of CSCs subpopulation by sphere formation ability. Results: As a result, a combination of sorafenib and paclitaxel significantly reduced the resistance while the CSCs exhibited drug resistance against paclitaxel alone. Also, combination of these agents reduces the self-renewal potential of CSCs when compared to single treatment. Simultaneously, combination significantly suppressed not only the colony formation but also the tube formation of the Cancer stem cells. Conclusions: These results suggest the combination therapy of paclitaxel and sorafenib in low doses be an attractive approach to target cancer stem cells in the future.

Imidacloprid (IMI), the main component of neonicotinoid insecticides, promotes oxidative stress and genotoxicity in mammals. The aim of this experiment is to assess oxidative stress in liver cells and genotoxicity of erythrocytes for rats exposed to sub-lethal doses of IMI and the protective effects for Rhodophyta as antioxidant material versus imidacloprid. A total of 30 adult male albino rats (average body weight, 190-200 g) were divided into six groups (n=5) as follows: group 1 served as the control, group 2 received 200 mg/kg red algae, group 3 received 45 mg/kg IMI (high-dose group), group 4 received 22.5 mg/kg IMI (low-dose group), group 5 received 200 mg/kg red algae +45 mg/kg IMI, and group 6 received 200 mg/kg red algae +22.5 mg/kg IMI. After 28 d of treatment, the antioxidant activity of the crude extract of red algae was assessed in terms of free radical scavenging activity and found to be higher in TCA (75.57%) followed by DPPH (50.08%) at concentration 100 μg extract and a significant increase in lipid peroxidation and reductions in glutathione were observed in liver cells were intoxicated with high and low doses of IMI. Moreover decreases in catalase and glutathione peroxidase parameters in same previous groups which indicated oxidative stress. In addition significant increases in micronucleus frequency (MN) in the bone marrow of the rats as a genotoxicity marker which indicated DNA damage in erythrocytes cells with alterations in the histopathology of liver cells were also noted such as necrosis, inflammatory cells, infiltration, and necrobiotic changes. Whereas Rhodophyta succeeded in alleviation the oxidative damage and genotoxicity induced by the insecticide. In conclusion, IMI demonstrates hazardous effects, such as alterations in antioxidant status and mutagenicity of erythrocytes and polysaccharides from Rhodophyta has good antioxidant activity in vivo model systems against imidacloprid.

The Sasaq community in Lombok, Indonesia has been recognized as a peasant community with its unique and strong social capital. Sources of social capital recognition can be derived from common terms or expressions and institutions practiced in community daily life. However, there is a trend of neglecting and ignoring those values by the community, especially the youth. Through action research, we would like to revitalize social capital of the community in supporting social and economic development in the rural level. In this paper, we introduce a Strategic Leadership and Learning Organization (SLLO) approach to build community participation in solving social and economic problems. Through regular dialogue, communities come with common agreements and collective action that are perceived as emergence property. Several common agreements are intended to solve community problems actually in line with the objectives of government designated development.

Clostridium difficile infection (CDI) is a rapidly emerging infection that may have devastating consequences. Prompt and accurate diagnosis is crucial for management and control. The aim of this study was to determine the incidence of C. difficile associated diarrhea among hospitalized patients, and to compare different diagnostic laboratory methods for detection of toxin producing strains in clinical specimens. The study was conducted at a university hospital in Cairo during the period from May 2013 till June 2015. Subjects were under antibiotic therapy and presented with hospital-acquired diarrhea. Four hundred and sixty-five stool specimens were processed by different microbiological methods. C. difficile was recovered in culture in 51 of stool specimens. Of these, 86.3% to 98% were positive for toxin production by 2 different methods. This study showed that antibiotic intake is the major risk factor for development of hospital-acquired diarrhea. We evaluated different microbiological methods for diagnosis of C. difficile. We recommend the use of toxigenic culture as a gold standard for microbiological diagnosis of C. difficile.
