Studies on Histones from Mouse Peritoneal Macrophages

여러종류의 생물체에 대한 전 histone 및 histone 분획에 관한 연구는 많지만 macrophage에 관한 것은 거의 찾아 볼 수 없다. 본 연구는 마우스 복강에서 유인한 macrophage의 단백성분을 Johns 방법에 의거하여 그 histone 분획 즉 $f_1$, $f_{2a1}$, $f_{2a2}$, $f_{2b}$ 및 $f_3$를 추출하여 조사하여 보았다. 이 다섯 분획은 모두 polyacrylamide disc gel 전기 영동 및 amino 산 조성의 분석을 행하여 관찰하였고 그 결과의 요약은 아래와 같다. 1. 마우스 복강 macrophage에서 얻은 전 histone의 양은 23.80 mg/g packed cells 였다. 2. 다섯가지 주분획의 비교적 수획율은 $f_1$, $f_{2a1}$, $f_{2a2}$, $f_{2b}$ 및 $f_3$에서 각각 10.9%, 11.9%, 18.4%, 36.2% 및 22.6%였다. 3. 마우스 복강 macrophages 에서의 $f_{2a2}$ 치는 마우스 간장이나 소 흉선에서의 그것과 유사하였으나 마우스 복강 macrophages에서는 $f_1$이 현저히 강소되어 있었고 또 $f_{2b}$가 현저하게 증가되어 있었으며 동시에 $f_{2a1}$과 $f_3$치가 얼마간 감소되고 있음을 볼 수 있었다. 4. Histone의 전체량과 이의 분획상 수률은 lysine-rich 분획인 $f_1$과 arginine-rich 분획인 $f_{2a}$만 제하고는 정상인 백혈구와 유사하였다. Histone의 전체량은 정상조직인 마우스간이나 소흉선 또는 병적 세포인 Sarcoma 180 복수세포들 보다 많았으나 사람의 백혈병 백혈구 보다는 적었다. 5. 전기영동상 histone의 각 분획은 정상 조직과 유사한 성장을 보여 주었다. 6. Histone 각 분획의 amino 산 분석에 있어서는 대체적으로 정상인 백혈구의 조성과 유사한것 같았으나 얼마간의 이질성도 동시에 보여 주고 있었다. In an attempt to investigate the pattern of macrophages, histone fractions f1, f2a1, f2a2, f2b and f3 were prepared from the precipitated mouse peritoneal macrophage protein by the procedure of Johns. These five major fractions were all characterized by polyacrylamide disc gel electrophoresis and amino acid analysis. The results of the experiment were as follows: 1. Whole histone obtained from mouse peritoneal macrophage was 23.80 mg/g packed cells. 2. The relative amounts of five major fractions: f1, f2a1, f2a2, f2b and f3 were 10.9%, 11.9%, 18.4%. 36.2% and 22.6% respectively. 3. The amount of f2a2 from mouse peritoneal macrophages was almost similar to mouse liver and calf thymus, but there were marked decrease in f1 and marked increase in the amount of f2b with concomitant decrease in f2a1 and f3 fractions present. 4. Total amount of histones and relative amount of histone fractions were similar to that of normal human leukocytes except lysine-rich fl fraction and arginine-rich fraction f2a. The amount of total histones was much larger than in normal tissue such as mouse liver or calf thymus and a certain tumor (Sarcoma 180 ascites cells) but was lesser than human leukemic leucocytes. 5. The electrophoretic pattern of histone fraction gave similar patterns to those mouse liver or calf thymus. 6. Amino acid analysis of the individual histone fractions showed that the overall compositions were similar but not identical to those of corresponding fractions from human leucocytes and other cells.
