Brown Guinea Pig와 Hairless Mouse를 이용한 광노화 모델에서 LGNC-5의 경구반복투여에 의한 피부주름발생 및 색소침착억제 효과

비타민과 불포화지방산, 그리고 폴리페놀의 복합처방인 LGNC-5 (LG생활건강 기술연구원 개발)가 건강기능식품의 개별인증을 받을 수 있는지의 여부를 확인하기 위해 hairless mice와 brown guinea pig 모델을 이용해 자외선에 의한 주름생성 및 색소침착 억제력 평가를 실시하여 다음과 같은 결괴를 얻었다. 1. Hairless mice를 대상으로 실시한 주름억제실험에서 육안관찰과 피부주형의 영상분석결과, LGNC-5군은 농도의존적인 주름발생 억제효과를 보였으며, 고용량군(x60)인 LGNC-5H군의 경우 10주후 정상군과 유사한 수준의 양호한 피부 상태를 보였다. Hematoxylin & Eosin stain 에서도 LGNC-5H군에서는 UV 대조군에 비해 표피 두께와 과각화가 현저하게 줄어드는 것으로 조사되어 우수한 주름발생억제 효과를 확인하였다. 2. Brown guinea pig을 대상으로 실시한 색소침착억제력 실험에서 육안관찰과 색차계를 이용한 측정결과, LGNC-5H군에서는 설험개시 7일 후부터 UV 대조군에 비해 색소침착이 억제되기 시작해, 설험 종료시까지 색소침착억제효과를 보였다(p<0.05). Fontana-masson silver stain 에서도 LGNC-5H군에서는 UV 대조군에 비해 melanin 침챡량이 현저하게 감소된 것으로 조사되어 LGNC-5H의 색소침착억제효과를 확인하였다. 3. 10주간의 주름억제실험과 4주간의 색소침착 억제실험에 사용한 모든 설험동물에서 사망 혹은 시험물질 투여로 인한 특이한 임상증상이 관찰되지 않았다. 시험물질 투여 기간 중 사료와 음수 소비량 역시 유의한 변화를 관찰할 수 없었으며, 대조군과 실험군 모두 정상적인 체중 증가를 나타내었다. 이상의 실험결과를 종합하면 LGNC-5은 안전하고 경구반복투여 시 UV조사로 인해 hairless mice에서 발생되는 주름과 brown guinea pig에서 생성되는 색소침착을 효과적으로 억제시키는 처방인 것으로 평가되었다. Chronic exposure of UV radiation in sunlight results in a number of biological responses, including erythema, sunburn cell formation, immune suppression, photoaging, and skin cancer. In this study, we investigated the effects of LGNC-5 (LG Nutricosmetics-5, the mixture of vitamins, unsaturated fatty acid and polyphenol) on UV-induced skin damage. Oral administration of LGNC-5 for 10 weeks resulted in inhibition of UVB-induced wrinkle formation in SKH-1 hairless mice skin compared to UV control group as a dose dependent manner. LGNC-5 treatment also inhibited UV radiation-induced skin pigmentation in brown guinea pigs examined by colorimeter and Fontana-masson silver staining. These results mean that LGNC-5 as a dietary supplement could be useful to attenuate solar UVB-induced melanogenesis and premature skin aging.
백수오(白首烏)와 비주화(啤酒花) 복합물의 에스트로겐 활성과 파골세포 분화 억제효과

Objectives : This study aimed to investigate the synergistic effect of combining Cynanchi Wilfordii Radix extract with Humuli Lupuli Flos extract on estrogenic and anti-osteoclastogenic activity. Methods : Estrogenic effect of a mixture of Cynanchi Wilfordii Radix extract and Humuli Lupuli Flos extract (CWHL), Cynanchi Wilfordii Radix extract, Humuli Lupuli Flos extract, caudatin (an active ingredient of Cynanchi wilfordii Radix extract) and 8-prenylnaringenin (an active ingredient of Humuli Lupuli Flos extract) were examined by proliferation E-screen assay and expression of estrogen inducible gene, pS2 via Real Time-PCR (RT-PCR) in MCF-7 estrogen responsive cells. And their estrogenic activities were investigated how to modulate Estrogen receptor 𝛽 by binding affinity assay. Inhibitory effect of CWHL, Cynanchi Wilfordii Radix extract, Humuli Lupuli Flos extract, caudatin and 8-prenylnaringenin on RANKL-induced osteoclast differentiation were tested by TRAP (Tartrate-resistant acid phosphatase) staining in osteoclastogenic RAW 264.7 cells. Results : CWHL, Humuli Lupuli Flos extract and 8-prenylnaringenin accelerated the proliferation of MCF-7 and the expression of pS2 in MCF-7. CWHL, Cynanchi Wilfordii Radix extract, Humuli Lupuli Flos extract, caudatin and 8-prenylnaringenin bind to estrogen receptor 𝛽. CWHL, Cynanchi Wilfordii Radix extract, Humuli Lupuli Flos extract, caudatin and 8-prenylnaringenin inhibited RANKL-induced osteoclastogenesis in osteoclastogenic RAW 264.7. CWHL is more effective for all markers than Cynanchi Wilfordii Radix extract or Humuli Lupuli Flos extract alone. Conclusions : CWHL may a potential therapeutic agent for menopause and osteoporosis as a natural food resource. CWHL as a natural food source has therapeutic potential in cases of menopause and osteoporosis.
마가자(馬家子) 추출물의 연골 보호 및 항염 효과에 관한 연구

Objectives : From this study, we sight to identify chondro-protective and anti-inflammatory effects of Sorbi Commixtae Fructus extract and its compound, chlorogenic acid. Methods : Sorbi Commixtae Fructus were extracted by 50% ethanol. And chlorogenic acid in Sorbi Commixtae Fructus 50% extract was quantified by high performance liquid chromatography (HPLC). To investigate chondro-protective effects, we treated Sorbi Commixtae Fructus 50% ethanol extract and chlorogenic acid in TNF𝛼-activated ATDC5 murine chondrogenic cells. After 24 hours, protein level of matrix metalloproteinase-3 (MMP3) and mRNA level of matrix metalloproteinase-13 (MMP13) were measured by using ELISA or reverse transcription PCR, respectively. To examine anti-inflammatory effects, we treated Sorbi Commixtae Fructus 50% ethanol extract and chlorogenic acid in LPS-induced RAW 264.7 murine macrophages. We measured the level of inflammatory mediators, such as Prostaglandin E2 (PGE2), Interleukin-6 (IL6) by ELISA and nitric oxide (NO) by Griess reagent assay. Results : A concentration of chlorogenic acid in Sorbi Commixtae Fructus 50% ethanol extract was 3.9 mg/g. Sorbi Commixtae Fructus 50% ethanol extract and chlorogenic acid attenuated protein level of MMP3 and mRNA level of MMP13 in TNF𝛼-activated ATDC5 cells. Sorbi commixtae Fructus 50% ethanol extract inhibited the level of PGE2, IL6 and NO in LPS-activated RAW 264.7 cells in dose dependent manner, but chlorogenic acid has no anti-inflammatory effects. Conclusions : These findings demonstrated that Sorbi Commixtae Fructus 50% ethanol extract has chondro-protective and anti-inflammatory effects showing possible therapeutics to ease the symptoms related with osteoarthritis.
골담초꽃 추출물의 연골 보호 및 염증 억제 효과에 관한 연구

Objectives : This research aimed to investigate chondro-protective and anti-inflammatory effects of Caraganae Sinicae Flos 50% ethanol extract and its compound, tilianin. Methods : Caraganae Sinicae Flos was extracted with 50% ethanol. Tilianin in Caraganae Sinicae Flos 50% ethanol extract was quantified by HPLC analysis method. To investigate chondro-protective effects of Caraganae Sinicae Flos 50% ethanol extract, ATDC5 chondrogenic cells were co-treated with Caraganae Sinicae Flos 50% ethanol extract (or tilianin) and tumor necrosis factor-𝛼 (TNF𝛼) for 24 hours. After treatement for 24 hours, media supernatant was used for quantifying protein level of matrix metalloproteinase-3 (MMP3) by ELISA and harvested cells were used for analyzing mRNA expression level of matrix metalloproteinase-13 (MMP13) by reverse transcription PCR. To identify anti-inflammatory effects of Caraganae Sinicae Flos 50% ethanol extract, RAW 264.7 macrophage cells were co-treated with Caraganae Sinicae Flos 50% ethanol extract (or tilianin) and lipopolysaccharide (LPS) for 24 hours. media was used for quantifying the level of prostaglandin E2 (PGE2), interleukin-6 (IL6) by ELISA and nitric oxide by Griess reagent asssay. Results : Caraganae Sinicae Flos 50% ethanol extract and tilianin attenuated protein level of MMP3 and mRNA expression level of MMP13 in TNF𝛼-activated ATDC5 cells. Caraganae Sinicae Flos 50% ethanol extract inhibited the level of PGE2, IL6 and NO in LPS-activated RAW 264.7 cells in dose dependent manner, though tilianin inhibited PGE2 only. Conclusions : These results presented that Caraganae Sinicae Flos 50% ethanol extract could be used as natural medicines for osteoarthritis.
유산균으로 발효한 침향공진단으로부터 분리한 Nodakenetin의 Neuraminidase 활성 억제 효능

The purpose of this study was to identify the changes in the components of unfermented Gongjin-dan (GD) and fermented Gongjin-dan (FGD) and to confirm whether GD or FGD has an inhibitory effect on viral neuraminidase (NA) activity. A major component of FGD was isolated and identified as nodakenetin, which is the aglycone of nodakenin. After fermentation, the nodakenetin content in FGD was approximately 10- fold higher than that in GD. Then, we examined the viral NA-inhibitory activity of GD, FGD, nodakenin, and nodakenetin. At a concentration of 500 μg/ml, FGD inhibited viral NA activity by 92% compared to the DMSO-treated control, while GD barely inhibited viral NA activity. In addition, 250 μg/ml of nodakenetin inhibited viral NA activity by 68% compared to the control, while nodakenin inhibited viral NA activity by only 4% at the same concentration as nodakenetin. Collectively, these results suggest that FGD has a more remarkable viral NA-inhibitory activity than GD because the content of the anti-viral component nodakenetin was higher in FGD due to the hydrolysis of nodakenin by Lactobacillus plantarum KCTC 3104.
