      • 고관절 표면 치환술 환자의 LATERAL 검사법에 대한 고찰

        Purpose : During the follow- up test of resurfacing hip joint arthroplasty patients, the lateral screening test is usually taken to distinguish wear, tear, debris, or osteolysis, when the boundaries of the bone and the implant are described as a single line. So far, however, there is no reported radiologic data about effective test method for accurate information processing and image description, so far. Therefore, we comparatively analyzed the effectiveness of methods by measuring time of lateral screening test useful for diagnosis and temporizing translateral test. Matherials and Methods : 200 follow-up patients (152 men, 48 women) of resurfacing hip joint arthroplasty, who had visited our hospital from April 2009 to January 2010 were subjected for this study, and their average age was 44.9 years (20~78 years old). ① 12 university hospitals including us were subjected for the survey of whether the resurfacing hip joint arthroplasty was held or not, and about the lateral test method. ② We classified and measured the femur angle at the hip AP(Internal rotation 15°) position into 5 groups (A:120°~125°, B:125°~130°, C:130°~135°, D:135°~140°, E:140°~145°) by PACS and analyzed the distribution. We also classified the incident beam into 4 groups (Cranial 30°, 35°, 40°, 45°), tested the effective incident beam(Tube angle) for diagnosis by femur angle, asked for screening by faculties, residents, and radiologists of the department of radiology and orthopedics, and finally analyzed statistically by Chi- square verification. ③ We comparatively analyzed the diagnostic effectiveness and work efficiency of temporizing translateral test with the published experimental method, during the lateral test for resurfacing hip joint arthroplasty patients. Results : ① 5 of 12 university hospitals provided resurfacing hip joint arthroplasty treatment, while only our hospital was proved to conduct an independent test method. ② The result of Chi-square test(P-Value=0.001) showed the maximal femur angle distribution at 125°~140°, and the most useful image for diagnosis was obtained from the result of 129 of 200 patients showing the incident beam(Tube angle) at Cranial 35°~40°. ③ Both groin lateral test and translateral test provided useful images for diagnosis. But, the groin lateral test method could improve the problems of translateral test method about the reproducibility of incident beam and the low repeat rate of measurement. Also, the efficiency of test was much higher by reducing average measurement time for more than 2 times (40.85 sec vs 82.85 sec). Conclusion : From this study subjecting the follow-up patients of resurfacing hip joint arthroplasty, the result of lateral screening test showed the best image with the incident angle by Cranial 35°~40° to find out wear, tear, debris, or osteolysis between the bone and the implant. In addition, the time also reduced by more than 2 times compared to the temporizing method.

        Electrostatic Spray를 이용한 Synbiotic 소재 함유 곡류 팽화 제품 개발

        본 연구에서는 다양한 생리활성을 가진 GABA를 함유하며 장내 미생물에 유익한 영향을 줄 수 있는 synbiotic 소재를 함유하는 곡류 팽화 제품을 개발하고자 하였다. 김치에서 분리한 유산균 동정을 위해 API 50 CHL kit에서 당 발효실험을 진행하여 API web program에 입력한 결과 L. brevis로 판명되었으며, 정확한 species 확인을 위해 16s rRNA sequencing을 진행한 결과 L. brevis(I.D. 99%)로 동정되어 L. brevis CFM21로 명명하였다. 최적 생장조건을 확인하기 위해 L. brevis CFM21을 MRS broth에서 배양한 결과 37°C, 24시간이었다. 균주의 GABA 생성량은 311.03 ㎍/mL였으며 전구체인 MSG를 0.8%(w/v) 첨가했을 때 926.42 ㎍/mL의 최대 생산량을 보였다. 프로바이오틱스 특성 실험결과 pH 2에서 내산성을 나타내지 않았으며, 담즙산 내성은 확인되었다. 팽화 제품의 제조에 이용되는 곡물 소재를 열수추출한 후 분리 균주와 배양하여 생육증진 효과를 확인하였으며, CLSM의 Z-stack 기능을 이용하여 곡물 소재에 균주가 부착한다는 것을 시각화하여 곡물 소재의 프리바이오틱스 특성을 확인하였다. 4종의 에센셜 오일을 10%(v/v)의 농도에서 paper agar diffusion method로 항균 활성을 나타내는 Citrus sinensis oil을 선별하였으며, 5종의 균주에 대해 최소저해농도를 측정했을 때 C. perfringens 균에 대해 가장 높은 항균 활성을 확인하였고 1.25%(v/v)를 장내유해균을 선택적으로 억제할 수 있는 농도로 선정하였다. 미강 추출물에 탄소원으로 dextrose 2%(w/v), 질소원으로 soytone 2.5%(w/v), 무기염류로 potassium chloride 0.2%(w/v)를 첨가하여 배지로 사용하였으며, 고농도 GABA 생산을 위해 MSG를 첨가한 결과 0.6%(w/v)의 농도로 첨가했을 때 524.77 ㎍/mL로 가장 높은 GABA 생성량을 확인하여 최적화 배지로 사용하였다. 최적화 미강 배지에 L. brevis CFM21을 배양시켜 고농도의 GABA를 생산하였으며, 장내 유해균 억제를 위해 선별한 Citrus sinensis oil을 첨가하여 곡물 소재 팽화 제품에 적용하였다. Electrostatic spray 처리를 통해 팽화 제품에 적용한 결과 GABA 함량이 10.78 ㎍/g에서 112.10 ㎍/g으로 증가하였다. 결론적으로 본 실험에서는 유익균 증진 활성을 지니는 곡물 소재를 이용한 팽화 제품에 electrostatic spray 처리로 L. brevis CFM21과 미강 배지를 통해 생산한 GABA 및 Citrus sinensis oil을 코팅함으로써, 장내 미생물 군총의 균형 유지에 도움이 되는 synbiotic 기능성을 지닌 제품의 제조 가능성을 확인하였다. This study aimed to develop synbiotic materials and apply them to puffed grain products using electrostatic spraying technology. Lactic acid bacteria were isolated from kimchi and selected through tests of acid resistance, bile-salt resistance, and γ-aminobutyric acid (GABA) content. The isolated Lactobacillus brevis CFM21 produced the highest GABA production up to a concentration of 926.42 ㎍/mL when grown in MRS broth containing 0.8% MSG. The possibility of coated grains as a prebiotic material was evaluated using confocal laser scanning microscopy (CLSM). Rice bran extract containing 2% dextrose, 2% soytone, 0.2% potassium chloride, and 0.6% MSG produced 524.77 ㎍/mL of GABA. Citrus sinensis oil showed the highest antibacterial activity against Clostridium perfringens. Electrostatic spray showed much higher effectiveness than conventional spray in coating the puffed grain product through CLSM. Applying a rice bran culture and Citrus sinensis oil to puffed grain product using an electrostatic spray can help promote the intestinal health of consumers.

      • 국내산 곡물과 장내개선 효과 규명을 통한 synbiotic 소재 개발 및 산업화

        The purpose of this study was to select the lactic acid bacteria (LAB) through acid resistance,???? bile salt resistance ,heat resistance, organic acid production and examine the LAB to attach to grains, ability of fermentation conditions. Confocal scanning laser microscopy (CLSM) was used to confirm the adhesion of grains and Lactobacillus spp. The selected strains were fermented in corn extract to confirm the optinum ratio of carbon and nitrogen source. The optimum ratio was applied to the mixed grain extracts to examine the growth curve and organic acid production. After fermentation activity, antioxidant of mixed grain extracts were measured. As results, Lactobacillus salivarius CFM 22 and Lactobacillus plantarum CFM 23 showed the highest potential as LAB. The selected strains were observed to adhesion to grains through CLSM. When the optimum conditions of fermentation obtained from the corn extract were applied to the mixed grain extracts, the growth curve and organic acid production were increased. After fermentation, antioxidant level in mixed grain extracts was increased. As results, we could confirm the possibility of the selected strains and grains as synbiotic food material.
