The replication sequences of Autographa californica nuclear polyhedrosis virus (AcNPV) ts-B1074 were comparatively visualized at time intervals from 18 to 48 hours postinfection at 25℃ and 32.5℃. The late stages of the multiplication sequence were quite different in the amounts of virogenic stroma formation and nucleocapsid formation, polyhedra occlusion body formation, and occlusion of enveloped virions at 25℃ and 32.5℃. The amounts of virogenic stroma formation decreased and the amounts of NOVs remained more in the nucleus at 32.5℃. The polyhedra appeared early by 18h postinfection at 32.5℃, but later by 24h postinfection. The formations of the polyhedra and the occlusion of the enveloped virions onto the polyhedra were more defective and temperature-sensitive at 32.5℃. Bundling of nucleocapsids and cytopathic effects of virions were not different at either temperature. But the amounts of extracellular NOVs were present much more at 25℃. The mutant had defects in the late genes.

The events in the middle multiplication sequences (10 to 18 hours postinfection) of Autographa californica nuclear polyhedrosis virus (AcNPV) ts-B1074) in Spodoptera frugiperda cells at 25℃ and 32.5℃ were comparatively visualized by electron microscopy. Early stages (0 time to 8 hours postinfection) of the AcNPV L-1 and ts-B1074 multiplication appeared to be the same pathways at both temperatures (Oh and Lee, 1987). The general aspects of the middle events in the multiplication were (1) the formations of the virogenic stroma, progeny nucleocapsid and prepolyhedra occlusion bodies, (2) bundling, enveloping and budding of the nucleocapsids, and (3) the hypertrophy of the nuclear membrane. The middle stages in the multiplication of the ts-B1074 at 25℃ and 32.5℃ were slightly different in the prepolyhedra, virogenic stroma and nucleocapsid formations at 10h to 18h postinfections which were stimulated and appeared early at 32.5℃. Amounts of nucleocapsid production and bundling and cytopathic effects not affected by temperatures. The amounts of extra-cellular nonoccluded nucleocapsids were greater at 25℃.
