An Arabidopsis Ca²?/H? antiporter (CAX1) contains an N-terminal autoinhibitory domain. Ectopic expression of the N-terminal truncated CAX1, sCAX1, increases Ca levels in several plants. In order to develop transgenic potato tuber with enhanced calcium content, the sCAX1 gene was introduced into potato under either CaMV 35S or patatin promoter. Transgenic potato plants were generated using an Agrobacteriummediated transformation system, in which they were selected on MS medium containing 2 ㎎ㆍL?¹ zeatin, 0.3 ㎎ㆍL?¹ IAA, 500 ㎎ㆍL?¹ carbenicillin, and 100 ㎎ㆍL?¹ kanamycin. Expression of sCAX1 gene in transgenic potato tubers was confirmed by Northern blot and RT-PCR analyses. The transcript level of CaMV 35S-sCAX1 fusion in transgenic potato tubers was higher than that of patatin-sCAX1 fusion. The transformants under CaMV 35S promoter contained more Ca than untransformants; however, sCAX1 transgenic potato plants regulated by patatin promoter were not.
