        16S rRNA 유전자 계통분석에 의한 한강수계의 세균 다양성

        한강의 본류와 만나는 탄천과 중랑천에서 16S rDAN를 증폭하고 부분적인 염기서열 분석을 통하여 한강의 세균 다양성을 결정하였다. 총 27개의 클론을 분리하였으며 RFLP를 이용하여 7개의 group으로 나누었다. 탄천의 15개 클론은 4개의 group으로 나뉘어졌으며 가장 많은 클론을 포함하는 group(HT-1 클론)은 class Proteobacteria의 ${\delta}$-subdivision에 속하는 Acrobacter cryaerophilius와 높은 유사도를 보였으며, 다른 두 group(HT-6과 HT-9 클론)은 모두 clas Cytophagales에 속하였다. 중랑천의 12개의 클론은 3개의 group으로 나뉘어졌으며 가장 많은 클론을 보이는 group(HJ-1 클론)은 class Proteobacteria의 ${\alpha}$-subdivision에 속하는 Sphingomonas sp. 와 높은 유사도를 나타내었다. 전체적으로는 Proteobateria(alpha, beta and delta subdivision), Cytophagales와 Actinomycetales가 검출되었다. Bacterial diversity was determined by amplification and sequencing of 16S rDNA at Tancheon and Jungrang in Han river. Twenty-seven clones constructed were divided 7 groups using RFLP. Fifteen clones were classified 4 groups in Tancheon and the group (HT-1 clone) including many clones was affiliated a high similarity with Aerobacter cryaerophilus (the class Proteobacteria including members of the delta subdivisions). The other two groups (HT-6 and HT-9 clone) including several clones were classified with the class Cytophagales in Tancheon. Twelve clones were classified 3 groups in Jungrang and the group (HJ-1 clone) including many clones was affiliated a high similarity with Sphingomonas sp. (the class Proteobacteria including members of the alpha subdivisions). As a whole results, the class Proteobacteria (alpha, beta and delta subdivision), the order Cytophagales, and the order Actinomycetales were detected.

        호수에서의 종속영양세균에 의한 질소고정능

        Limiting factors for the nitrogen fixation potentials by heterotrophic bacteria are not ammonia but carbon and phosphate. The nitrogen fixation potentials were investigated by acetylene reduction method that was added carbon, phosphate, and ions. The nitrogen fixation potentials ranged 0∼520.6μM-ethylene/L/day in Lake Soyang and 0∼615.3μM-ethylene/L/day in Daechung Reservoir. The nitrogen fixation potentials showed positive correlation with water temperature in Lake Soyang (r=0.75, P<0.01) and Daechung reservoir (r=0.71, P<0.01). As a result, the nitrogen fixation potential by heterotrophic bacteria were mainly affected by water temperature in Lake Soyang and Daechung reservoir.

        대청호의 세균분포에 미치는 환경요인

        From June 1991 to November 1992, biological and physio-chemical environmental factors were investigated 18 times at 5 stations in the upper area of Daechung reservoir. The ranges of heterotrophic bacteria were 2.5×10 exp (2)∼7.3×10 exp (4), proteolytic bacteria were 1∼6.7×10 exp (3), lipolytic bacteria were 0.6×10 exp (2)∼1.9×10 exp (4), spore forming bacteria were 2∼55(CFU/㎖), total coliform bacteria were ND∼1.1×10 exp (3) and fecal coliform bacteria were ND∼2.9×10 exp (2) (MPN/100㎖)during the investigated period. All bacterial distribution of station 1 was affected by the precipitation and the bacterial distribution of stations located on downstream it was affected by the water temperature instead of the precipitation. On the total result, monthly variation of bacterial distribution was stronger than stational variation, therefore the important environmental factor affecting to the distribution of bacteria was analysed as water temperature.

        대청호에서의 질화작용

        The nitrification rate and environmental factors were estimated 12 times at 4 stations from March 1992 to January 1994 in Daechung reservoir. The number of nitrifying bacteria and nitrification rate were measured with MPN and ^14C-dark incorporation method, respectively. The distribution of ammonia-oxidizing bacteria was 0.5∼220MPNml^-1(mean, 13MPNml^-1) and that of nitrite-oxidizing bacteria was ND-42MPNml^-1(mean,3MPNml^-1). The range of nitrification rate was 0.01∼7.15㎍-Nl^-1day^-1(mean, 0.74㎍-Nl^-1day^-1) and the highest value was 7.15㎍-Nl^-1day^-1 at station 3 where lots of fishfarm were located. Being showed through multiple regression analysis from all data estimated, the distribution of nitrification was affected by change of ammonia concentration, but not affected by nitrifying bacterial population.
