열목어(Brachymystax lenok) transferrin cDNA의 構造解析 및 分子進化學적 系統分類에 관한 연구

In the present study, we report the molecular cloning, of liver transferrin cDNA in lenok and using its sequence molecular phylogeny is identified in salmonids. Lenok transferrin cDNA identified a 2,411 bp which amplified from liver cDNA with polymerase chain reation(PCR). Amplified clone has a open reading frame encoding 690 amino acids, contain a polyadenylation signal (AATAAA) located at position 2,338∼2,343 bp and 32 cysteine residues, posses one argininclinked glycosylation site located at 476∼478aa. The deduced amino acid sequence of lenok transferrin cDNA has 62% to 88% amino acid identities with other fish species. The results obtained indicate that lenok can be separated from the other 3 groups in 10 species of salmonids. Comparison of these 4 groups show that the lenok group is closer to Salmo and Salvelinus phylogenetic groups than the Oncorhynchus group. Such molecular phylogenetic results support at present phylogenic trees in salmonids.
