      • Characterization of a Napthoquinone Type Toxic Compound in Cottonseed Gland Material

        면화씨로 부터 gossypol 과 구조가 유사한 갈색 색소를 분리 정제하였다. 이 갈색 색소는 silicic acid column에서 gossypol 뒤에 그러나 gossyverdurin 앞에 유출 되었으며 acetic acie adduct로 결정화하였고 그 물리 화학적 특성을 조사하였다. 이 유해한 갈색 색소는 naphtoquinione이나 naphthydroquinone 형의 화학구조를 가진 것으로 생각되며 구조식은 6,6'-dihydroxy-5,5'-diisopropyl-3.3'-dimethyl-(2.2'-binaphthalene)-1,1',7,7'-tetrone-8, 8'-dicarboxaldehyde일 것으로 사료된다. A brown pigment structually similar to gossypol has been isolated from cottonseed pigment gland material. It was purified and crystallized as the acetic acid adduct and certain chemical and physical characteristics of this compound have been determined. Further work is needed to prove the structure of this compound but data from this study indicate the compound may be a more highly oxidized form of gossypol. A naphthoquinone or naphthydroquinone type structure has been indicated by the infrared spectra. The structure of this compound is proposed as 6,6'-dihydroxy-5,5'-diisopropyl-3,3'-dimethyl-(2,2'-binaphthalene)-1,1',7,7'-tetrone-8,8'-dicarboxaldehyde.

      • Effects of Starvation on Cardiac and Renal Protein Metabolism

        $1-^{14}C$-glutamate을 쥐에게 준후 5일간 굶주리게 했을때 심장과 신장의 총 단백질과 aldolase 양의 변화를 측정하였다. 총 단백질과 aldolase의 합성 분해속도를 계산하였다. 기아 상태에서 심장과 신장 aldolase는 다른 반응을 보였다. 신장 aldolase는 분해합성 속도가 다 늦추어짐으로 control과 비슷한 수준을 보이는 반면 심장 aldolase는 감소했다. Adult rats were starved for 5 days after the pulse administration of $1-^{14}C$-glutamate. The radioactivities of pure aldolase and mixed total proteins from heart and kidneys were determined. The rates of protein synthesis and degradation were calculated for cardiac and renal proteins and aldolases. Kidney and heart aldolase responded differently in starvation. Kidney aldolase activity was sustained while cardiac aldolase activity decreased.

      • A Study on the Binding Characteristics of Naloxone Binding Opiate Receptors

        Sprague Dawley 쥐 뇌의 opiate receptor의 날록손 결합 양상을 Scatchard plot 하였을때 high affinity binding과 low binding을 나타내었다. High affinity binding sites의 Bmax는 8.79 fmol/mg protein이었고 low affinity binding sites의 dissociation 상수 (Kd)는 9.09 nM 이었다. Brain membrane을 phospholipase $A_2$와 반응시켰을때 Bmax는 7.13 fmol/mg으로 Kd는 0.32 nM로 감소하였다. 1, 6, 12주 된 쥐 뇌막에서의 phosphatidylserine 양, lipid peroxide 형성과 날록손 결합 양상에 대한 변화를 관찰하였다. Apparant two binding sites, high and low affinity, were shown when Scatchard plotted after the rat brain membrane was analyzed for $^3H$-naloxone binding characteristics with opiate receptors. The maximum number of high binding sites (Bmax) was 8.79 fmol/mg brain membrane protein while the low affinity binding sites displayed the apparent dissociation equilibrium constant (Kd) of 9.09 nM. When the brain membrane was treated with phosphalipase A, Bmax has reduced to 7.13 fmol/mg and Kd to 0.32 nM. The changes in binding parameters with 1, 6, 12 weeks old rat brain were observed under various experimental conditions.

        무지개송어의 성숙에 따른 식품성분 및 지질과산화물의 변화

        양어장에서 양식된 무지개송어의 성장에 따른 식품성분과 지질과산화물의 변화를 측정하여 다음의 결과를 얻었다. 치어와 성어의 조지방 함량과 수분 함량은 3.54%, 9.59% 및 77.40%, 69.02%로 성장에 따라 조지방의 함량은 증가되고 수분은 감소하였다. 지방산 조성은 치어에서 saturates 30.18%, monoenes 37.81%, polyenes 25.09%이고, 성어에서 30.77%, 40.41%, 23.45%로 성장에 따라 monoenes의 비율은 증가하고 polyenes은 감소하였다. 구성아미노산조성은 성장에 따라 큰 차이를 보이지 않았으며, 모두 glutamic acid, aspartic acid, histidine이 전체 아미노산의 약 50%를 차지하였다. 지질과산화의 지표로 측정한 malondialdehyde, lipofuscin은 성장에 따른 유의적인 증가를 보이지 않았다. The present study was carried out to compare the differance of proximate composition and lipid peroxides in juvenile(80~120g) and adult(670~690g) rainbow trout(Oncorhynchus mykiss). There was a marked increase in lipid content with growth. The fatty acids of juvenile rainbow trout was composed of saturates, monoenes and polyenes of 30.18%, 37.81% and 25.09%, respectively. Adult rainbowtrout showed higher content in monoenes and lower content in polyenes. The composition of amino acid was similar. In both groups, glutamic acid, aspartic acid, histidine were abundant. The lipid peroxides, malondialdehydes and lipofuscin, were not increased significantly with growth.

        트립토판 보강식이를 섭취한 마우스에서 serotonin 대사와 morphine 진통기작 관련성에 대한 연구 ( 2 )

        N/A In this study we fed control diet and tryptophan supplemented diets containing 0.35% tryptophan to ICR mice for 2 weeks. The concentrations of serotonin and 5-HIAA were changed by injection of the serotonin synthesis inhibitor, p-CPA and the serotonin precursor, serotoninP and the change of brain serotonin concentration negatively correlated with that of pain sensitivity, and p-CPA and serotoninP also changed the analgesic effect of morphine. The injection of naloxone, the opiate antagonist, resulted in an increase in the writhing frequency, but its antagonistic effect was not significant. The concentration of 5-HIAA elevated in mice brain at least 3hr after administration of morphine hydroxide indicates that the changes in brain serotonin metabolism may be associated with the acute effects of morphine analgesia. In short, these results not only suggest that tryptophan supplemented diet suppress pain sensitivity in mice, but also indicate that at least in part analgesic mechanism of serotonin may be associated with morphine analgesia.

        두릅나무와 황백피의 혼합추출물 P55A 의 랫트 및 개에 대한 경구투여 급성독성

        The current study was performed to determine the acute oral toxicity of P55A, a crude extract of 1:1 mixture of Phellodendron amurense cortex and Aralia elate cortex, in SD rats and beagle dogs. 5 rats of each sex were treated with a single dose of P55A orally at doses of 0 and 5,000 mg/kg respectively. Also 2 dogs of each sex were treated with a single dose of P55A orally at doses of 0 and 2,000 mg/kg, respectively. After the treatment, clinical signs, and body weight change were observed for 14 days. All rats survived during the study and did not show any clinical sign. Body weight gain showed no significant difference between the control and treated rats. Grossly, no lesion was observed in the rats. All dogs survived during the study. In clinical signs, dark stool was observed in the 2,000 mg/kg treated dogs at day 1 after administration. The animals recovered from general signs at day 2 after administration. Body weight gain showed no significant difference between the control and treated dogs. Grossly, no lesion was observed in the dogs. It is suggested that the LD_(50) of P55A by oral administration was estimated to be over 5,000 mg/kg in both sexes of rats and 2,000 mg/kg in both sexes of beagle dogs.
