Controversy exists on blood supply to the auxiliary graft being provided by the splanchnic venous circulation or a comparable systemic venous inflow being suitable. As a preliminary study, in order to asses whether or not normal portal venous flow is necessary to maintain the function of auxiliary hepatic graft, end-to-side portacaval shunts were performed in mature Sprague-Dawley male rats, weighing 250 to 300gms, by suture technique of Lee's method. All animals in these experiments were sacrificed 2, 5, 10 and 30 days subsequent to the surgical procedure. The DNA and RNA contents as well as GDA activity in the liver of shunt-operated animals were determined and the serum LDH and GDA activities were also assayed along with the follow ups of serum A/G ratio, cholesterol, alkaline phosphates and GPT. The results were as follows; 1) The reduction of liver weight and liver atrophy were observed by the shunt operation. 2) The nucleic acid contents in the liver of shunt-operated animals were increased during 30 days following the operation. 3) The GDA activity of the shunt-operated animals was increased in both the serum and liver, reaching to the control levels 30 days after the operation. 4) The serum LDH activity was increased on the 5th day after the operation, returning to the control level at the end of the experimental period.
