• Annual Financial Impact of Thyroidectomies for Nodular Thyroid Disease in China

        Liu, Xiao-Yun,Zhu, Li-Jun,Cui, Dai,Wang, Zhi-Xiao,Chen, Huan-Huan,Duan, Yu,Shen, Mei-Ping,Zhang, Zhi-Hong,Wang, Xiao-Dong,Chen, Jia-Wei,Alexander, Erik Karl,Yang, Tao Asian Pacific Journal of Cancer Prevention 2014 Asian Pacific journal of cancer prevention Vol.15 No.14

A large proportion of patients with thyroid nodules in China undergo thyroidectomy in order to get confirmatory histology diagnosis. The financial impact of this modality remains to be investigated. To evaluate rationality of performing thyroidectomy without a routine FNA preoperatively from the economic perspective, we conducted a retrospective, observational study of all archival thyroidectomies with records of cost per stay (CPS), cost per day (CPD) and length of stay (LOS) from 2008 to 2013 in the First Affiliated Hospital of Nanjing Medical University. We compared all the parameters between cancer and non-cancer thyroidectomies. We recruited 6, 140 thyroidectomies with valid records of CPS, CPD and LOS in this period. The CPS of cancer thyroidectomy was significantly higher than non-cancer thyroidectomy. The percentage of cancer thyroidectomy increased from 26.5% to 41.6%. The percentage of annual cost of cancer thyroidectomies rose from 30.2% to 45.2%. The LOS for cancer and non-cancer thyroidectomy decreased while the CPD increased in the past six years. The estimated national cost in 2012 for all thyroidectomies would be USD 1.86 billion with USD 1.09 billion for non-cancer thyroidectomies. We have witnessed great improvement in the healthcare for patients with thyroid nodules in China. However, given limited healthcare resources, currently thyroid FNA for more precise preoperative diagnosis may help to curb the rapidly increasing demand in healthcare costs in the future for nodular thyroid disease in China.

Ginsenoside Re Increases Fertile and Asthenozoospermic Infertile Human Sperm Motility by Induction of Nitric Oxide Synthase

        Hong Zhang,Qing-Ming Zhou,Xiao-Da Li,Yi Xie,Xin Duan,Feng-Ling Min,Bing Liu,Zhi-Gang Yuan 대한약학회 2006 Archives of Pharmacal Research Vol.29 No.2

We investigated the effects of Ginsenoside Re on human sperm motility in fertile and asthenozoospermic infertile individuals in vitro and the mechanism by which the Ginsenosides play their roles. The semen samples were obtained from 10 fertile volunteers and 10 asthenozoospermic infertile patients. Spermatozoa were separated by Percoll and incubated with 0, 1, 10 or 100 µM of Ginsenoside Re. Total sperm motility and progressive motility were measured by computer-aided sperm analyzer (CASA). Nitric oxide synthase (NOS) activity was determined by the 3H-arginine to 3H-citrulline conversion assay, and the NOS protein was examined by the Western blot analysis. The production of sperm nitric oxide (NO) was detected using the Griess reaction. The results showed that Ginsenoside Re significantly enhanced both fertile and infertile sperm motility, NOS activity and NO production in a concentration-dependent manner. Sodium nitroprusside (SNP, 100 nM), a NO donor, mimicked the effects of Ginsenoside Re. And pretreatment with a NOS inhibitor Nω-Nitro-L-arginine methyl ester (L-NAME, 100 µM) or a NO scavenger N-Acetyl-L-cysteine (LNAC, 1 mM) completely blocked the effects of Ginsenoside Re. Data suggested that Ginsenoside Re is beneficial to sperm motility, and that induction of NOS to increase NO production may be involved in this benefit.

      • Rho-GTPase Effector ROCK Phosphorylates Cofilin in Actin-Meditated Cytokinesis During Mouse Oocyte Meiosis

        Duan, Xing,Jun-Liu,Dai, Xiao-Xin,Liu, Hong-Lin,Cui, Xiang-Shun,Kim, Nam-Hyung,Wang, Zhen-Bo,Qiang-Wang,Sun, Shao-Chen Society for the Study of Reproduction [etc.] 2014 BIOLOGY OF REPRODUCTION Vol.90 No.2

During oocyte meiosis, a spindle forms in the central cytoplasm and migrates to the cortex. Subsequently, the oocyte extrudes a small body and forms a highly polarized egg; this process is regulated primarily by actin. ROCK is a Rho-GTPase effector that is involved in various cellular functions, such as stress fiber formation, cell migration, tumor cell invasion, and cell motility. In this study, we investigated possible roles for ROCK in mouse oocyte meiosis. ROCK was localized around spindles after germinal vesicle breakdown and was colocalized with cytoplasmic actin and mitochondria. Disrupting ROCK activity by RNAi or an inhibitor resulted in cell cycle progression and polar body extrusion failure. Time-lapse microscopy showed that this may have been due to spindle migration and cytokinesis defects, as chromosomes segregated but failed to extrude a polar body and then realigned. Actin expression at oocyte membranes and in cytoplasm was significantly decreased after these treatments. Actin caps were also disrupted, which was confirmed by a failure to form cortical granule-free domains. The mitochondrial distribution was also disrupted, which indicated that mitochondria were involved in the ROCK-mediated actin assembly. In addition, the phosphorylation levels of Cofilin, a downstream molecule of ROCK, decreased after disrupting ROCK activity. Thus, our results indicated that a ROCK-Cofilinactin pathway regulated meiotic spindle migration and cytokinesis during mouse oocyte maturation.

Chk2 Regulates Cell Cycle Progression during Mouse Oocyte Maturation and Early Embryo Development

        Dai, Xiao-Xin,Duan, Xing,Liu, Hong-Lin,Cui, Xiang-Shun,Kim, Nam-Hyung,Sun, Shao-Chen Korean Society for Molecular and Cellular Biology 2014 Molecules and cells Vol.37 No.2

As a tumor suppressor homologue during mitosis, Chk2 is involved in replication checkpoints, DNA repair, and cell cycle arrest, although its functions during mouse oocyte meiosis and early embryo development remain uncertain. We investigated the functions of Chk2 during mouse oocyte maturation and early embryo development. Chk2 exhibited a dynamic localization pattern; Chk2 expression was restricted to germinal vesicles at the germinal vesicle (GV) stage, was associated with centromeres at pro-metaphase I (Pro-MI), and localized to spindle poles at metaphase I (MI). Disrupting Chk2 activity resulted in cell cycle progression defects. First, inhibitor-treated oocytes were arrested at the GV stage and failed to undergo germinal vesicle breakdown (GVBD); this could be rescued after Chk2 inhibition release. Second, Chk2 inhibition after oocyte GVBD caused MI arrest. Third, the first cleavage of early embryo development was disrupted by Chk2 inhibition. Additionally, in inhibitor-treated oocytes, checkpoint protein Bub3 expression was consistently localized at centromeres at the MI stage, which indicated that the spindle assembly checkpoint (SAC) was activated. Moreover, disrupting Chk2 activity in oocytes caused severe chromosome misalignments and spindle disruption. In inhibitor-treated oocytes, centrosome protein ${\gamma}$-tubulin and Polo-like kinase 1 (Plk1) were dissociated from spindle poles. These results indicated that Chk2 regulated cell cycle progression and spindle assembly during mouse oocyte maturation and early embryo development.

Ginsenoside $R_e$ Increases Fertile and Asthenozoospermic Infertile Human Sperm Motility by Induction of Nitric Oxide Synthase

        Zhang Hong,Zhou Qing-Ming,Li Xiao-Da,Xie Yi,Duan Xin,Min Feng-Ling,Liu Bing,Yuan Zhi-Gang The Pharmaceutical Society of Korea 2006 Archives of Pharmacal Research Vol.29 No.2

We investigated the effects of Ginsenoside $R_e$ on human sperm motility in fertile and asthenozoospermic infertile individuals in vitro and the mechanism by which the Ginsenosides play their roles. The semen samples were obtained from 10 fertile volunteers and 10 asthenozoospermic infertile patients. Spermatozoa were separated by Percoll and incubated with 0, 1, 10 or $100\;{\mu}M$ of Ginsenoside $R_e$. Total sperm motility and progressive motility were measured by computer-aided sperm analyzer (CASA). Nitric oxide synthase (NOS) activity was determined by the $^{3}H$-arginine to $^{3}H$-citrulline conversion assay, and the NOS protein was examined by the Western blot analysis. The production of sperm nitric oxide (NO) was detected using the Griess reaction. The results showed that Ginsenoside $R_e$ significantly enhanced both fertile and infertile sperm motility, NOS activity and NO production in a concentration-dependent manner. Sodium nitroprusside (SNP, 100 nM), a NO donor, mimicked the effects of Ginsenoside $R_e$. And pretreatment with a NOS inhibitor $N^{w}$-Nitro-L-arginine methyl ester (L-NAME, $100\;{\mu}M$) or a NO scavenger N-Acetyl-L-cysteine (LNAC, 1 mM) completely blocked the effects of Ginsenoside $R_e$. Data suggested that Ginsenoside $R_e$ is beneficial to sperm motility, and that induction of NOS to increase NO production may be involved in this benefit.

Correlation between Non-Alcoholic Fatty Liver Disease and Visceral Adipose Tissue in Non-Obese Chinese Adults: A CT Evaluation

        Ai-Hong Yu,Yang-Yang Duan-Mu,Yong Zhang,Ling Wang,Zhe Guo,Yong-Qiang Yu,Yu-Sheng Wang,Xiao-Guang Cheng 대한영상의학회 2018 Korean Journal of Radiology Vol.19 No.5

Objective: To investigate the correlation between non-alcoholic fatty liver disease and visceral adipose tissue in non-obese Chinese adults using computed tomography (CT). Materials and Methods: The study included 454 subjects undergoing abdominal CT scan. Degree of CT attenuation in liver and spleen, and the degree of fat infiltration in liver were evaluated according to three indices: the attenuation value of liver parenchyma (CTLP), the attenuation ratio of liver and spleen (LSratio) and the attenuation difference between liver and spleen (LSdif). Visceral fat area (VFA) and total fat area (TFA) at L2/3 and L4/5 levels were measured, and the abdominal subcutaneous fat area (SFA) was calculated. Bivariate correlation analysis was carried out to determine the correlation among these factors. Results: In men, VFA, SFA and TFA at L2/3 and L4/5 levels showed significant differences in terms of the three indices to distinguish fatty liver from non-fatty liver (all, p < 0.001). In men, all the three indices showed negative correlation with TFA, SFA and VFA (all, p < 0.001). The negative correlation between the three indices and VFA at the L2/3 level was higher than at L4/5 level (r = -0.476 vs. r = -0.340 for CTLP, r = -0.502 vs. r = -0.413 for LSratio, r = -0.543 vs. r = -0.422 for LSdif, p < 0.001, respectively). The negative correlation between LSratio, LSdif and VFA at L2/3 and L4/5 levels was higher than SFA at the corresponding level. In women, all the three indices showed negative correlation with VFA and TFA at L2/3 and L4/5 levels, and the negative correlation between CTLP and VFA was higher at L2/3 level than at L4/5 level (r = -0.294 vs. r = -0.254, p < 0.001). Conclusion: In non-obese Chinese adults, the degree of hepatic fatty infiltration showed a strong correlation with abdominal fat on CT. VFA at L2/3 level was more closely related to fatty liver compared with VFA at L4/5 level.

Characterization of free and glycosidically bound volatile compounds, fatty acids, and amino acids in Vitis davidii Foex grape species native to China

        Yi-Bin Lan,Xiao-Feng Xiang,Wei-Xi Yang,Bao-Qing Zhu,Hong-Tie Pu,Chang-Qing Duan 한국식품과학회 2020 Food Science and Biotechnology Vol.29 No.12

Berries of six Vitis davidii Foex (spine grape)cultivars (‘Baiputao’, ‘Gaoshan 1’, ‘Gaoshan 2’, ‘Seputao’,‘Miputao’, and ‘Tianputao’) were harvested from a commercialvineyard in Hunan Province in China. Free andbound volatile compounds and fatty acids were analyzedby GC–MS, and amino acids were analyzed by HPLC. ‘Tianputao’ and ‘Miputao’ were characterized by relativelyhigher concentrations of aromatic amino acids and lowerconcentrations of branched-chain amino acids. The majorfree volatile compounds of spine grapes were hexanal, (E)-2-hexenal, 1-hexanol, (E)-2-hexenol, (E)-b-damascenone,and benzeneacetaldehyde. The major glycosidically boundvolatile compounds identified were 1-hexanol, menthol, nerol, 1-butanol, 3-methyl-3-butenol, benzenemethanol, bphenylethanol, eugenol, and guaiacol. (E)-b-damascenone,benzeneacetaldehyde, guaiacol, and eugenol had odoractivity values (OAVs)[1 in all cultivar grapes. Partialleast squares discriminant analysis (PLS-DA) revealed‘Tianputao’ to be distinct from the other cultivars due to itsrelatively higher concentrations of major terpenoids, norisoprenoids, higher alcohols, and aromatic amino acids.

PREPARATION OF NANO-TATB BY SEMIBATCH REACTION CRYSTALLIZATION

        XUE-RONG TAN,XIAO-HUI DUAN,CHONG-HUA PEI,HONG-LIN XU 성균관대학교(자연과학캠퍼스) 성균나노과학기술원 2013 NANO Vol.8 No.5

The 1,3,5-triamino-2,4,6-trinitrobenzene (TATB) nanoparticles were prepared by using semibatch reaction crystallization method, and the influencing factors in close relationship with the grain size and crystal morphology control, such as the concentration of reaction system and categories of surfactants, were studied in this paper. The synthesized nano-TATB particles had been characterized by SEM, XRD, thermo gravimetric/differential scanning calorimetric (TG/DSC) and N2 physisorption. The grain size of TATB particles using nonionic surfactant as the additive ranged from 30 nm to 65 nm with a shape of spheres or ellipsoids. The broadening of the peaks and the weakening of the strength for nano-TATB were observed by XRD analysis. The corrected average particle size of nano-TATB was calculated using the Debye–Scherrer equation and the range was from 18 nm to 50 nm. TG and DSC curves revealed that thermal decomposition of nano-TATB occurred in the range of 361.5°C–385.0°C and its peak temperature was 373.7°C with a decrease of approximately 7°C compared with original TATB. Furthermore, the specific surface area (21.54 m2/g) of nano-TATB was calculated by BET method using N2 physisorption (at 77°C).

Structural Evolution of Sub-10 nm Octahedral Platinum–Nickel Bimetallic Nanocrystals

        Chang, Qiaowan,Xu, Yuan,Duan, Zhiyuan,Xiao, Fei,Fu, Fang,Hong, Youngmin,Kim, Jeonghyeon,Choi, Sang-Il,Su, Dong,Shao, Minhua American Chemical Society 2017 NANO LETTERS Vol.17 No.6

Octahedral Pt alloy nanocrystals (NCs) have shown excellent activities as electrocatalysts toward oxygen reduction reaction (ORR). As the activity and stability of NCs are highly dependent on their structure and the elemental distribution, it is of great importance to understand the formation mechanism of octahedral NCs and to rationally synthesize shape-controlled alloy catalysts with optimized ORR activity and stability. However, the factors controlling the structural and compositional evolution during the synthesis have not been well understood yet. Here, we systematically investigated the structure and composition evolution pathways of Pt-Ni octahedra synthesized with the assistance of W(CO)(6) and revealed a unique core-shell structure, consisting of a Pt core and a Pt-Ni alloy shell. Below 140 degrees C, sphere-like pure Pt NCs with the diameter of 3-4 nm first nucleated, followed by the isotropic growth of Pt-Ni alloy on the seeds at temperatures between 170 and 230 degrees C forming Pt@Pt-Ni core-shell octahedra with {111} facets. Owing to its unique structure, the Pt@Pt-Ni octahedra show an unparalleled stability during potential cycling, that is, no activity drop after 10 000 cycles between 0.6 and 1.0 V. This work proposes the Pt@Pt-Ni octahedra as a high profile electrocatalyst for ORR and reveals the structural and composition evolution pathways of Pt-based bimetallic NCs.
