        Silencing of Heat Shock Protein 27 Expression Accelerates Doxazosin-induced Apoptosis in Prostate Cancer Cell Line PC-3

        Purpose: Heat shock proteins (HSPs) are highly expressed during stress responses and cellular adaptation to environmental changes. One such protein is HSP27, a 27kDa protein that prevents cell death induced by many pro-apoptotic agents. Therefore, the aim of this study was to investigate the correlation between HSP27 expression and apoptosis induced by doxazosin treatment in prostate cancer cell line PC-3. Materials and Methods: RT-PCR, Western blotting, and immunocytochemical staining were performed to determine whether HSP27 mRNA and protein are expressed in PC-3 cells. Next, to investigate the effects of doxazosin on apoptosis and HSP27 protein expression in PC-3 cells, the cells were stained using a TUNEL kit (to detect apoptotic cells) and with HSP27 antibody (to assess HSP27 protein expression) 6, 12, 24, and 48h after treatment with 25μM doxazosin. In addition, to determine whether HSP27 mRNA interference accelerates doxazosin-induced apoptosis of PC-3, we knocked down HSP27 with siRNA and then evaluated the rate of apoptosis after doxazosin treatment. Results: HSP27 mRNA and protein were expressed in PC-3 cells. Furthermore, HSP27 mRNA and protein levels increased until 12 hours after 25μM doxazosin treatment, whereas the rate of apoptosis did not increased dramatically. After 12 hours, HSP27 expression decreased and then apoptosis was accelerated. In addition, siRNA-mediated knockdown of HSP27 induce higher apoptosis rate of PC-3 cells even before 12hrs after doxazosin treatment. Conclusions: By inhibiting apoptosis, HSP27 expression might play an important role in inhibiting progression to castration-refractory prostate cancer and resistance to anti-cancer treatment.

        Plastic house의 형태, 재배양식 및 시설내 위치에 따른 기상환경의 차이가 암면재배 오이의 생장에 미치는 영향

        재배시기, 시설형태 그리고 시설내 계배위치에 따른 가상환경의 차이와 이에 따른 양액계배 오이의 생장 반응을 알아보기 위하여 여름재배와 억제재배, 1-2W형 하우스와 무기둥 하우스에 양액계배를 통해 오이를 패배한 후 시설내 부위별 기온, 상대습도, 일사량과 생장해석을 통해 얻어진 생리적 형질들을 조사하여 비교한 결과는 다음과 같다. 하우스 내부의 평균기온이나 평균상대습도는 하우스 형태간 유의한 차이가 없었으나 일사량은 1-2W형 하우스가 무기둥 하우스에 비해 유의하게 높았다. 일누적일사량은 1-2W형에서는 남서쪽이 무기둥 하우스에서는 북서쪽이 대체로 높았다. 초장과 엽수는 억제재배보다 여름재배에서 높게 나타났으며 엽면적은 억제재배에서 많았다. 상대생장율은 순동화율 및 엽면적비율과 고도로 유의한 정의 상관관계를 보였으며 상대생장율에 대한 기여도는 순동화율이 엽면적비율보다 높은 것으로 나타났다. 작물생장율은 엽면적지수와 고도로 유의한 정의 상관을 보여 엽면적지수의 증대가 단위면적당 생산량의 증대에 크게 영향하는 것으로 나타났다. 전 생육기간의 일평균기온는 상대생장을 및 순동화율과 고도로 유의한 정의 상관관계가 있었다. 억제재배에서는 전 생육기간의 누적일사량과 상대생장을 및 순동화율과 유의한 정의 상관관계가 있었다. This experiment was conducted to investigate the effects of climatic factors varied due to the type of plastic house, cultural season and location in the house on the growth of cucumber plants grown by nutrient solution. There were two growing periods, summer culture and retarding culture, two types of plastic houses, 1-2W type house and post-less house. Air temperature, relative humidity and amount of solar radiation in the plastic houses were measured. Also, dry weight of leaves and stems, plant height, number of leaves per plant, leaf area per plant and fresh weight of fruits per plant were observed. Plant growth analysis were conducted and interrelationships between climatic factors and physiological characteristics were investigated. The results were as follows. There were no differences between the type of plastic houses in the average air temperature and average relative humidity in the plastic house, but amount of solar radiation in 1-2W type house was significantly higher than that of postless house. Daily cumulative solar radiation were highest in southwest side of 1-2W type house and northwest side of postless house. Plant height and number of leaves per plant were higher in summer culture than retarding culture, while leaf area per plant was higher in retarding culture than summer culture. Relative growth rate (RGR) showed highly significantly positive correlations with net assimilation rate (WAR) and leaf area ratio (LAR). Contribution of NAR to RGR was much higher than that of LAR. Crop growth rate (CGR) showed highly significantly positive correlations with leaf area index (LAI). It appeared that increase of LAI was important to increase productivity of cucumber. Average daily air temperature for the whole growing period showed highly significantly positive correlations with RGR and NAR. Furthermore, cumulative solar radiation for the whole growing period in retarded culture showed significantly positive correlation with RGR and NAR.

        Effect of single nucleotide polymorphism on the total number of piglets born per parity of three different pig breeds

        Objective: To determine the effects of genomic breeding values (GBV) and single nucleotide polymorphisms (SNP) on the total number of piglets born (TNB) in 3 pig breeds (Berkshire, Landrace, and Yorkshire). Methods: After collecting genomic information (Porcine SNP BeadChip) and phenotypic TNB records for each breed, the effects of GBV and SNP were estimated by using single step best linear unbiased prediction (ssBLUP) method. Results: The heritability estimates for TNB in Berkshire, Landrace, and Yorkshire breeds were 0.078, 0.107, and 0.121, respectively. The breeding value estimates for TNB in Berkshire, Landrace, and Yorkshire breeds were in the range of -1.34 to 1.47 heads, -1.79 to 1.87 heads, and -2.60 to 2.94 heads, respectively. Of sows having records for TNB, the reliability of breeding value for individuals with SNP information was higher than that for individuals without SNP information. Distributions of the SNP effects on TNB did not follow gamma distribution. Most SNP effects were near zero. Only a few SNPs had large effects. The numbers of SNPs with absolute value of more than 4 standard deviations in Berkshire, Landrace, and Yorkshire breeds were 11, 8, and 19, respectively. There was no SNP with absolute value of more than 5 standard deviations in Berkshire or Landrace. However, in Yorkshire, four SNPs (ASGA 0089457, ASGA0103374, ALGA0111816, and ALGA0098882) had absolute values of more than 5 standard deviations. Conclusion: There was no common SNP with large effect among breeds. This might be due to the large genetic composition differences and the small size of reference population. For the precise evaluation of genetic performance of individuals using a genomic selection method, it may be necessary to establish the appropriate size of reference population.

        Pathogenesis of Human Norovirus Genogroup II Genotype 4 in Post- Weaning Gnotobiotic Pigs

        Norovirus is the most common cause of acute gastroenteritis. Its pathogenesis is poorly understood owing to the difficulty of establishing viral infection in animal models. Here, postweaning gnotobiotic pigs were infected with human norovirus genogroup II genotype 4 (HuNoV GII.4) to investigate the pathogenesis and replication of the virus. Three groups of four pigs were infected with 1 × 10<sup>5</sup>, 1 × 10<sup>6</sup>, or 1 × 10<sup>7</sup> genomic equivalent (GE) copies of HuNoV GII.4. Four pigs were used as negative controls. Blood and rectal swab samples were collected after viral infection, and gross legions were examined after necropsy. Diarrhea was induced in 25% and 75% of pigs infected with 1 × 10<sup>6</sup> and 1 × 10<sup>7</sup> GE copies, respectively. Viral shedding was detected in 50%, 75%, and 50% of pigs infected with 1 × 10<sup>5</sup>, 1 × 10<sup>6</sup>, and 1 × 10<sup>7</sup> GE copies, respectively. Viremia was detected in 25% of pigs infected with either 1 × 10<sup>6</sup> or 1 × 10<sup>7</sup> GE copies. When gross lesions of gastroenteritis were investigated, the ileum walls of the infected pigs were thinner than those of the controls. Villi atrophy and inflammatory cell infiltration were identified in the ileum of each infected pig. Viral capsid was identified in the jejunum, ileum, colon, spleen, and mesenteric lymph node. Virus replication was newly verified in the spleen and mesenteric lymph nodes by detection of negative-sense viral RNA. In conclusion, HuNoV GII.4 could induce acute gastroenteritis and replicate in the extraintestinal lymphoid tissues in post-weaning gnotobiotic pigs. Therefore, such pigs would be a suitable animal model for studying the pathogenesis and replication of HuNoV.

        로봇 보조 복강경 방광적출술 및 요로전환술의 초기 경험

        Purpose: Robot assisted laparoscopic radical cystectomy (RARC) is one of the options for minimal invasive treatment of muscle invasive bladder cancer or uncontrolled non muscle invasive bladder cancer. We detailed our operative techniques and initial experiences of robot assisted laparoscopic radical cystectomy with urinary diversion. Materials and Methods: From April 2010 to December 2011, 10 consecutive patients with bladder cancer who were performed robot assisted laparoscopic radical cystectomy with urinary diversion were enrolled. Patient’s demographics, intra-, perioperative data and oncologic outcome were analyzed. Urinary diversions including 9 ileal conduit and 1 ileal orthotopic neobladder were underwent extracoporeally. Results: Mean patient age was 65.1 years. The mean body mass index was 22.7kg/m². The mean operation time was 588.1min including 319.5min for robot assisted laparoscopic cystectomy (RARC), 165.0min for extracorporeal urinary diversion and 53.1min for lymph node dissection. The mean estimated blood loss was 244.7ml. The mean number of retrieved lymph node was 17.2. The pathologic report showed transitional cell carcinoma in all cases and adenocarcinoma of prostate in 2 male patients. No intra-operative complication was occurred. Conclusions: Robot assisted laparoscopic radical cystectomy could minimize surgery related complications without jeopardizing oncologic outcome. RARC may be effective treatment option for advanced bladder cancer.
