The brown seaweed Ecklonia cava is known to be a rich source of phlorotannins that have diverse biological activities. Among the phlorotannins in E. cava, concentrations of dieckol and phlorofucofuroeckol-A, which were identified as major active components, were determined in different parts of the tissue. We compared the efficacy of different pretreatments for their extraction. A high-performance liquid chromatography (HPLC) method to determine phlorotannin concentrations showed good accuracy (92.64 and 94.02%, respectively), precision (3.92 and 3.94%, respectively), and linearity (r>0.996). Mature thalli contained 1.5-fold more dieckol (1.82 mg/g-dry tissue) than young thalli. In the tissues of E. cava, blade tissue contained more phlorotannins than the stipe or holdfast. Among differently dried thalli, approximately 90% or more dieckol and phlorofucofuroeckol-A were extracted from shadow-dried tissue as compared with lyophilized tissue. In sun-dried and oven-dried thalli, approximately 60% of the phlorotannins were extracted. Thalli washed with fresh water, boiled thalli, and steamed thalli showed reduced extraction of the compounds.

Abalone containing phlorotannins is produced by feeding the phlorotannin-rich brown seaweed Eisenia bicyclis after 4 days of starvation. Reverse-phase high-performance liquid chromatography (RP-HPLC) was used to isolate and quantify phlorotannins, which were identified by mass spectrometry and [1H]-nuclear magnetic resonance to be the P1 compound, 7-phloroeckol, and eckol. When E. bicyclis was used as feed, P1 compound accumulated to an average of 1.60 mg/g dry weight of abalone muscle tissue after 18 d, 7-phloroeckolol to 0.21 mg/g after 16 d, and eckol to 0.22 mg/g after 12 d. Saccharina japonica was used as a control feed, and the abalone showed little or no accumulation of phlorotannins in muscle tissue. Feed consumption and growth rate were very similar when either E. bicyclis or S. japonica was fed for 20 d. Half-maximal reductions in the levels of P1 compound, 7-phloroeckol, and eckol accumulation were attained in 1.5, 1.9, and 3.4 days, respectively, after the feed was switched from E. bicyclis to S. japonica. Value-added abalone containing bioactive phlorotannins can be produced by simply changing the feed to the phlorotannin-rich E. bicyclis 18 d prior to harvesting.

An improved strain of the red seaweed Porphyra suborbiculata containing an increased amount of the essential amino acid llysine was obtained through mutation and analog enrichment. Mutagenesis using a 10% lethal dose of ultraviolet irradiation and an enrichment culture with the l-lysine analog aminoethyl-l-cysteine (AEC) was repeated to select the most productive strain using monospores of P. suborbiculata. The concentrations of AEC required to produce 50 and 100% inhibition of survival were 60 and 115mM in the parent strain, and 72 and 135mM in the selected AEC-resistant strain, respectively. The AEC-resistant strain, L130, produced 1.74-fold more lysine compared to its parent strain. Thus, mutagenesis with analog enrichment shows promise for selecting seaweed strains that can overproduce this essential amino acid.

<P>Sargassum fulvellum (Turner) C. Agardh is an edible brown macroalgae having pharmacological importance. In previous reports, we described the screening of marine algae for their neuritogenic activity in developing hippocampal neurons and found that ethanol extract of S. fulvellum (SFE) possesses promising neurite-outgrowth-promoting activity. In this study, we evaluated whether the initial neurite promoting effect of SFE was followed on the further neuronal maturation and synapse formation. SFE exhibited dose-dependent effect on neurite maturation with an optimum concentration of 5?μg/mL. The initial neuronal differentiation is significantly promoted by SFE. Subsequently, compared with control culture, SFE increased the indices of axonal and dendritic developments such as the number and the length of primary processes, and branching frequencies. In addition to its effect on neurite development, SFE significantly increased the number of puncta for postsynaptic density-95, synaptic vesicle 2, and synapse (about 35%, 67%, and 125%, respectively, of control). Moreover, SFE dose-dependently protects neurons from naturally occurring death in normal culture condition. Taken together, our data demonstrate that SFE can promote neuronal maturation and synaptogenesis and support neuronal survival, suggesting the beneficial effect of this alga in nervous system.</P>
