Background/Aims: Vedolizumab is indicated for moderately-to-severely active ulcerative colitis (UC) and Crohn’s disease (CD). Because multiple factors may result in different pharmacokinetics and clinical efficacies, understanding determinants of vedolizumab clearance may enhance dose and treatment strategies. The aim was to characterize vedolizumab pharmacokinetics in Asian and non-Asian UC and CD patients. Methods: Population pharmacokinetic analysis for repeated measures, using data from 5 studies, was conducted using nonlinear mixed-effects modeling. A Bayesian estimation approach in NONMEM 7.3 was utilized to leverage the predominantly sparse data available for this analysis with results from a prior population pharmacokinetic analysis of vedolizumab. Results: Vedolizumab pharmacokinetics were described by a 2-compartment model with parallel linear and nonlinear elimination. Using reference covariate values, linear elimination half life of vedolizumab was 24.7 days for anti-vedolizumab antibody (AVA)-negative patients and 18.1 days for AVA-positive patients; linear clearance (CLL) was 0.165 L/day for AVA-negative patients and 0.246 L/day for AVA-positive patients; central (Vc) and peripheral compartment volumes of distribution were 3.16 L and 1.84 L, respectively. Interindividual variabilities (percent coefficient of variation) were 30.8% for CLL and 19% for Vc; interoccasion variability on CLL was 20.3%; residual variance was 17.8%. For albumin, body weight and AVA, only extreme values were identified as potentially clinically important predictors of CLL. The effect of race (Asian/non-Asian) and diagnosis (UC/CD) on CLL was negligible and likely not of clinical importance. Conclusions: Pharmacokinetic parameters were similar in Asian and non-Asian patients with moderately-to-severely active UC and CD. This analysis supports use of vedolizumab flat-fixed dosing in these patients. (Clinicaltrials.gov Identifiers: NCT00783718 (GEMINI 1); NCT00783692 (GEMINI 2). Inhibition of Langerhans cell function by UVB radiation
Okamoto, Hiroyuki,Mizuno, Kana,Horio, Takeshi Korean Society of Photoscience 2002 Journal of Photosciences Vol.9 No.2
The functional disruption of Langerhans cells (LC) by UVB radiation is involved in antigen-specific immunosuppression of contact hypersensitivity. We tested whether UVB radiation inhibits the endocytotic activity of LC, which leads to impaired subsequent migration and maturation. Human monocyte-derived LC that took up lucifer yellow (L Y) or FITC-dextran (Fd) exclusively migrated in response to 6Ckine and matured. Exposing LC to 10-40 mJ/cm$^2$ of UVB radiation reduced their endocytotic activity in fluid phase pinocytosis (measured by uptake of LY) and in receptor-mediated endocytosis (measured by uptake of Fd). Membrane ruffling and CD32 expression were also suppressed by UVB radiation. UVB-irradiated, endocytosing LC had less movement towards 6Ckine, expressed less CD54 and CD86, and had less effective stimulatory activity in allo-MLR than nonirradiated, endocytosing LC. Endocytosis up-regulated TNF-$\alpha$ production by LC, but prior UVB radiation inhibited this enhancement. The finding that impaired endocytosis of LC by UVB radiation inhibits subsequent migration and maturation was also confirmed in murine epidermal cells obtained from unirradiated and 2OmJ/cm$^2$ of UVB-irradiated skin.

The author found out that there were some artificial points in research papers of the local history of Isahaya City, which have been overcome at the level of the view of history. However, the problems of historical research have given wrong contents to children through comic books and school education.

YAMAKAMI Takuju (1855-1931) and YOKOKAWA Umeko(1853-1926) were disciples of NAKAMURA Masanao (1832-91) who was the owner and headteacher at Doninsha School, one of the most famous schools in Tokyo in the latter half of 19th Century. Yamakami, born in the northern part of Motohachioji Village, made“Catholic Church School” for discriminated children and adults, and Yokokawa, born in the southern part of it, established the first girls’ school and nursery school in Tama Area. It is clear that Both of them had great influences on their home village, though there is no clear evidence of their contact. Yamakami’s intimate comrade and Yokokawa herself made efforts to compromise villagers when there happened a confrontation between north and south on the plan of uniting two primary schools in the village at the beginning of 20th Century. And we can find how significant Burakumin’s presence was, seeing the articles of the local press.

This study is about YAMAKAMI Takuju (1855-1931), an evangelist of Catholicism, teacher, businessman and activist of the Movement for Civic Rights and Freedom in the 1880s, who was born as a well-off but discriminated Burakumin. The author is planning to suggest a new point of view on him, thinking that an anonymous essay named “On political parties” (1890) is his work.

This study shows that we can see three kinds of people in the comparative history of the UK and Japan, through the research as follows: First, both Eden and Cedar PAUL who criticised the liberal arts education for the working class, especially by the WEA in the UK, and TSUCHIDA Kyôson who was the top of the same kind of education for adults in Japan, had problems on respecting the general-public. Second, as Tsuchida wondered if the WEA might have been an organisation for conciliating the working class, he would not get to know about the WEA. So, he missed the autodidact culture of it. Third, the young people at Chiyo Village in Nagano Prefecture rose up for their self-education, close to the autodidact culture of the UK, after Tsuchida"s failing of continuing the movement of Free Colleges, mainly because of his lack of respecting the general-public.
