<P> Transcription factor AP-2γ (TFAP2C) is a member of the transcription factor activating enhancer binding protein (AP) family. In the present study we determined the temporal and spatial expression patterns of TFAP2C in porcine parthenotes during preimplantation development. Porcine TFAP2C transcripts were expressed at all stages of preimplantation development, with highest expression at the 8-cell stage. In contrast with the mouse, TFAP2C protein was not restricted to the trophectoderm and was also detected in the ICM in blastocyst stage porcine parthenotes. In knockdown (KD) experiments, most TFAP2C-depleted embryos were arrested before the compacted 8-cell stage. This developmental failure is attributed to abnormal expression of genes involved in cell adhesion, tight junction biogenesis and cell proliferation. Interestingly, although the conserved region 4 (CR4) of the porcine OCT4 5′ upstream regionlacked the AP2C-binding motif, OCT4 transcript levels were elevated in porcine TFAP2C-KD 8-cell embryos, suggesting TFAP2C may be involved in the regulation of OCT4 in porcine embryos through other mechanisms. In summary, the results suggest that TFAP2C is necessary for the transition from de novo transcript synthesis by activation to compaction and further development, and the different expression patterns of TFAP2C in porcine embryos may reflect species-specific functions during preimplantation embryo development. </P>
Regulation of interleukin-11 expression in ovulatory follicles of the rat ovary

<P> The aim of the present study was to examine the regulation of interleukin (IL)-11 expression, as well as the role of IL-11, during ovulation in gonadotropin-primed immature rats. Injection of equine chorionic gonadotropin (eCG), followed by human CG (hCG) to induce superovulation stimulated expression of the Il11 gene in theca cells within 6 h, as revealed by northern blot and in situ hybridisation analyses. Real-time reverse transcription-polymerase chain reaction analysis showed that the IL-11 receptor, α subunit gene was expressed in granulosa and theca cells and that injection of hCG had no effect on its expression. IL-11 protein expression was stimulated in theca cells by hCG. LH-stimulated increases in Il11 mRNA levels in cultured preovulatory follicles were inhibited by protein kinase A and mitogen-activated protein kinase kinase inhibitors. Toll-like receptor (TLR) 2 and TLR4 were detected in preovulatory follicles, and the TLR4 ligand lipopolysaccharide, but not the TLR2 ligand Pam3Cys, increased Il11 mRNA levels in theca cells, but not in granulosa cells. Treatment of preovulatory follicles with IL-11 stimulated progesterone production and steroidogenic acute regulatory protein (Star) gene expression. Together, these results indicate that IL-11 in theca cells is stimulated by mitogen-activated protein kinase signalling and TLR4 activation, and increases progesterone production during ovulation. </P>

<P> Single crystal to single crystal transformations have been the subject of growing interest in recent years. In this article, several examples of single crystal to single crystal transformations that occur in metal-organic open frameworks (MOFs) upon guest removal and rebinding, guest exchange, and chemical oxidation are introduced. Depending on the structures of the MOFs, host framework structures can be retained or altered upon guest removal. When host framework structures are altered, significant rearrangements of the molecular components, which involve sponge-like shrinkage/swelling, sliding, swing, bending, and/or rotational motions, occurs throughout the entire crystal to retain the single crystallinity. </P>

<P> Sunlight-induced bromination of benzylic compounds was conducted in a capillary microreactor, resulting in mono-brominated compounds with yields of up to 94 %. These reactions can be considered to be eco-friendly since they were carried out without an artificial light source or additional temperature control. In addition, up to 257.9 mmol could be produced daily using cost-effective molecular bromine, which leads to potential improvement of industrial processes. </P>

<P>Despite the positive roles of histone deacetylase inhibitors in somatic cell nuclear transfer (SCNT), few studies have evaluated valproic acid (VPA) and its associated developmental events. Thus, the present study was conducted to elucidate the effect of VPA on the early development of bovine SCNT embryos and the underlying mechanisms of action. The histone acetylation level of SCNT embryos was successfully restored by VPA, with optimal results obtained by treatment with 3mM VPA for 24h. Importantly, the increases in blastocyst formation rate and inner cell mass and trophectoderm cell numbers were not different between the VPA and trichostatin A treatment groups, whereas cell survival was notably improved by VPA, indicating the improvement of developmental competence of SCNT embryos by VPA. Interestingly, VPA markedly reduced the transcript levels of endoplasmic reticulum (ER) stress markers, including sXBP-1 and CHOP. In contrast, the levels of GRP78/BiP, an ER stress-alleviating transcript, were significantly increased by VPA. Furthermore, VPA greatly reduced cell apoptosis in SCNT blastocysts, which was further evidenced by the increased levels of the anti-apoptotic transcript Bcl-xL and decreased level of the pro-apoptotic transcript Bax. Collectively, these results suggest that VPA enhances the developmental competence of bovine SCNT embryos by alleviating ER stress and its associated developmental damage.</P>
Interpenetrated Double Pillared-Layer CoII MOFs with pcu Topology

<P> Three double pillared-layer CoII metal-organic frameworks (MOFs) with a pcu topology of a long, conformationally flexible, dipyridyl spacer ligand, 1,4-bis[2-(4-pyridyl)ethenyl]benzene (bpeb), and aromatic dicarboxylates (1,4-benzenedicarboxylate (bdc), 2,6-naphthalenedicarboxylate (ndc), and biphenyl-4,4′-dicarboxylate (bpdc)) have been synthesised and structurally characterised by X-ray crystallography. The MOFs are denoted as [Co2(bpeb)2(bdc)2]·DMF·3H2O (1), [Co2(bpeb)2(ndc)2]·1.75DMF·3.75H2O (2), and [Co2(bpeb)2(bpdc)2]·3.5DMF·4H2O (3). In the dinuclear repeating unit, four carboxylates are bonded to two CoII atoms forming a (4,4) layer structure. The axial positions are occupied by bpeb ligands. Of these, 1 and 2 have 2-fold interpenetration, whereas 3 displays 3-fold interpenetration. The two bpeb space ligands in 1 have trans,trans,trans and trans,cis,trans conformations. In contrast, the bpeb ligands in 2 and 3 have a trans,cis,trans conformation. Although the olefin groups in two adjacent bpeb ligands, as the double pillars in 2 and 3, satisfy the conditions for photo-dimerisation to occur, they are photo-inactive. The conformational changes of bpeb, bonding modes of the dicarboxylates, and pore sizes in these double pillared-layer compounds have been discussed. </P>

<P> Aquaporin (AQP) water channels play an important role in fluid homeostasis and the control of epithelial cell volume. To understand the oviductal fluid homeostasis, the expression of aqp5 was examined in mouse oviduct. In the oviduct of cycling females, aqp1, aqp3, aqp4, aqp5, aqp6, aqp7, aqp8, and aqp11 mRNA were detected. Of these, expression of aqp5 mRNA increased significantly from the early prepubertal period to puberty. Epithelial AQP5 immunoreactivity was markedly increased during the same period and was most notable in the infundibulum. In immature female mice (3 weeks old), gonadotropin (pregnant mare’s serum gonadotropin (5 IU/head) and human chorionic gonadotropin (5 IU/head), single intraperitoneal injection) significantly increased oviductal aqp5 mRNA and AQP5 immunoreactivity in oviduct epithelia. In adult mouse oviduct epithelia, AQP5 was primarily found in the apical membrane, subapical cytoplasm and basolateral membrane of secretory non-ciliated cells, whereas weak to negligible immunoreactivity was found in β-tubulin-positive ciliated cells. Taking into account the fact that non-ciliated cells are well developed with subapical secretory vesicles as well as endosomes, AQP5 may also participate in the secretion and endocytosis in addition to water movement through non-ciliated secretory cells. AQP5 immunoreactivity was also found in the isthmic muscle and lamina propria beneath the epithelia. In cycling females, oviductal aqp5 mRNA levels were the highest at oestrus and the lowest at di-oestrus. AQP5 immunoreactivity in non-ciliated cells was notable in the infundibulum, where AQP5 immunoreactivity was relatively high at oestrus but low at dioestrus and pro-oestrus, indicating synchrony between aqp5 gene activation and the ovarian cycle. Together, the findings of the present study indicate that aqp5 specific to non-ciliated cells is activated during sexual maturation, supporting fluid homeostasis in mouse oviduct. </P>

<P> Crystal structure determination on the mixed ligand complex [Ru(tpy)(L1)](ClO4)2 (tpy = 2,2′:6′,2″-terpyridine; L1 = 4′-(ferrocenylmethylaminomethylphenyl)terpyridine) shows the ferrocenyl group to be located as remotely as possible from the RuII centre. This may explain the fact that emission from the RuII centre is detectable even at room temperature in solution. </P>
A Facile and Efficient Method for the Formation of Unsymmetrical Ureas Using DABAL-Me3

<P> A practical synthetic method for the formation of unsymmetrical-substituted ureas is described. The synthesis of the unsymmetrical ureas was readily performed from 2,2,2-trichloroethyl carbamate compounds by treatment of amines with bis(trimethylaluminum)-1,4-diazabicyclo[2.2.2]octane (DABAL-Me3). Using this reaction protocol, various trisubstituted and tetrasubstituted ureas were synthesized in high yields. This study offers a promising approach for the facile synthesis of a variety of unsymmetrical ureas from 2,2,2-trichloroethyl carbamates. </P>
