        MiR-103a-3p Contributes to the Progression of Colorectal Cancer by Regulating GREM2 Expression

        Purpose: Our research aimed to investigate the influence of miR-103a-3p on the growth and apoptosis of colorectal cancer (CRC)cells. Materials and Methods: Bioinformatics was employed to analyze differentially expressed microRNAs and predict target genes. qRT-PCR was applied to detect the expression of miR-103a-3p in CRC and normal cells. HCT116 and Caco-2 were chosen, andmiR-103a-3p mimics, miR-103a-3p inhibitor, as well as specific siRNAs targeting GREM2, were constructed. We subsequentlyevaluated alternations in cell proliferation, cell cycle and cell cycle regulators, apoptosis, and related proteins (Bcl-2 and Bax) byCCK-8 testing, Western blotting, luciferase reporter, colony formation, and Annexin V-FITC/PI. Possible binding sites for miR-103a-3p on the 3'UTR of GREM2 were checked with luciferase assay, and the impact of GREM2 on miR-103a-3p activity was also validatedwith above biological function testing. Additionally, the effect of miR-103a-3p knockdown in CRC cells and the molecularmechanism of miR-103a-3p targeting GREM2 were also studied. Results: Bioinformatics analysis revealed that miR-103a-3p expression increased remarkably in CRC, and targeted regulatory correlationexisted between miR-103a-3p and GREM2. MiR-103a-3p inhibitor significantly impeded proliferative capacity and caused cellcycle arrest, as well as apoptosis, in HCT116 and Caco-2 cells. Consistent with this finding, overexpression of GREM2 showed similareffects to miR-103a-3p inhibition. Moreover, we demonstrated that miR-103a-3p connected target GREM2 and GREM2 knockdownreversed the effects of miR-103a-3p inhibitor on HCT116 and Caco-2 cell proliferation, cell cycle, and apoptosis. Further study showedthat miR-103a-3p targeting GREM2 appeared to affect CRC progression via the transforming growth factor-β pathway. Conclusion: MiR-103a-3p could augment CRC progression by targeting GREM2 and that miR-103a-3p/GREM2 could be potentialnovel targets for CRC therapy.

        Three-dimensional dynamics simulation analysis of high clearance sprayer with road excitation

        The stability of a high clearance sprayer (HCS) can be considered in developing modern agriculture. We built a three-dimensional dynamics model of HCS with road excitation considering the influence of complex terrain on the whole vehicle vibration. The numerical simulation method was proposed to solve differential equations based on the Newmark-β method. The response curves of whole vehicle in the time and frequency domain response were analyzed. The simulation results show that the vibration amplitude difference of left and right wheel is bigger than amplitude difference of right and rear wheel. The average amplitude of roll motion is bigger than pitching motion. The reason is that since the man-machine seat is set on the left of whole vehicle, an additional roll torque is brought. Besides, the displacement amplitude of whole vehicle is bigger than man-machine seat. The reason is that the vibration absorption system of seat can reduce vibration effectively. In amplitude-frequency response curves, the multiple resonance points occur with the road excitation given in this paper. The maximum response amplitude of angle velocity and acceleration of pitching motion is bigger than roll motion. This study has an important theoretical and practical value for structure dynamic optimization of HCS.

        Application of Biophysical Properties of Meridians in the Visualization of Pericardium Meridian

        Background: The biophysical properties of the meridian system, an important concept of traditional Chinese medicine, include low impedance, resounding voice, and high acoustic conductance, all of which are helpful for elucidating the essence of meridians. Objectives: To visualize the human pericardium meridian (PC) based on the resounding voice property of meridians. Methods: Visualization of the PC was performed by injection of fluorescein sodium at the PC6 acupoint (Neiguan) on the PC. Before injection, percussion active points (PAPs) were identified by the virtue of their resounding voice properties. After injection, the trajectories of fluorescein migration throughout the body surface were recorded and analyzed. The distribution of fluorescein in the tissue was further studied using cross-sections of hind limbs of mini-pigs, in which fluorescein was injected into low impedance points. Results: The identified PAP lines were colocalized with PC. Following intradermal fluorescein injection, 1-3 fluorescent lines, which were unrelated to the arm veins, were observed in 7 of 10 participants; 85.4% of fluorescent signals were coincident with PAPs and their intensity had a negative correlation with the body mass index (r = –0.56, p = 0.045). Cross-sections showed a Y-shaped fluorescence pattern where the two migration lines on the surface were the two vertices of the “Y.” Conclusion: The trajectories of fluorescein in the body are suggestive of the anatomical structure of meridians. The PC is related to the deep horizontal interstitial channels that connect to the body surface through vertical interstitial spaces. These biophysical properties and techniques for meridian visualization are valuable for revealing the anatomical structure of meridians.
