        Ionic Liquids as Selectors for Controlling the Crystallization Nucleation of Hen Egg White Lysozyme

        Ionic liquids (ILs) have biomaterial applications and are used for protein crystallization. The effect of two imidazolium-based ILs, 1-butyl-3-methylimidazolium chloride ([C4mim]Cl) and 1,3-dimethylimidazolium iodide ([dmim]I),on the nucleation kinetics of lysozyme was investigated by determining the nucleation induction time, and nucleation parameters were evaluated. The values of interfacial tension calculated for solutions with added 30 g/L ILs [C4mim]Cl and [dmim]I, and without added ILs were 99.03, 109.7, and 107.3 mJ/m2, respectively. Compared with solutions without IL addition, the critical free energy change, size, and molecular number of critical nuclei decreased and the nucleation rate increased after the addition of [C4mim]Cl. In contrast, the critical free energy change, size, and molecular number of critical nuclei increased and the nucleation rate decreased after the addition of [dmim]I. These new findings provide insights into controlling lysozyme crystallization separation, and present ILs as potentially useful additives for controlling the crystallization of macromolecules.

        A new strategy for protein crystallization : Effect of ionic liquids on lysozyme crystallization and morphology

        Protein crystallization is a complex physical and chemical process. The high-quality protein crystal is still apersistent bottleneck to the application of X-ray crystallography in structural biology. The additives may promote formationof crystal nucleus and subsequent growth in protein crystallization. As a distinct material, ionic liquids (ILs)have aroused great attention and interest for protein crystallization due to their unique properties. We reviewed the progressof protein crystallization and reported research about protein crystal morphology control by ILs, as crystal growthtemplate, in aqueous solutions. ILs encourage changes in some cases in terms of growth morphology and crystal size. The effect of ILs on lysozyme growth morphology can be attributed to changing interaction among lysozyme moleculesin aqueous solutions. This work can provide some initial insight into the preparation of high quality crystal and thedevelopment of new crystal form.

        Solvated behavior and crystal growth mechanism of erythromycin in aqueous acetone solution

        The solubility of erythromycin acetone solvate and dihydrate was experimentally determined in aqueous acetone mixtures at different temperature. It has been demonstrated that solubility curves of the two solvates intersected at given solvent composition at various temperature, suggesting a transition behavior between two solvates. The induction period of acetone solvate at different supersaturation was measured by the laser monitoring observation technique. Based on classical homogeneous nucleation theory, the solid-liquid interfacial tension and surface entropy factor were calculated from the induction period data. From the surface entropy factor values calculated, together with surface morphology observation by the atomic force microscopy (AFM), the growth mechanism of erythromycin acetone solvate is consistent with continuous growth mode.

        Experimental study of beam-column connections with web opening in a low-rise steel frame

        Steel frame structures have been widely used in multi-storey and high-rise buildings and the connections in these structures are critical. In the Northridge and Kobe Earthquake, beam-column connections suffered damage due to brittle fracture. According to seismic design codes, ductility of the beam to column connection is also necessary. A study on the behavior of a beam to column connection with the aim of improving ductility as well as preventing brittle failure was carried out. In order to control the position of a plastic hinge on the beam, a connection with a hole in the beam web was developed. Five specimens with different parameters under cyclic load were assessed. The results are presented in terms of the stress distribution of the beam, hysteretic behavior, and ultimate capacity. Furthermore, the finite element method was also used to analyze the model, and the results were compared with those obtained from the experiment. It is shown from the analysis and experimental results that this type of connection is effective in terms of improving ductility for a beam to column connection in low-rise buildings.

        Measurement and correlation of solubility of xylitol in binary water+ethanol solvent mixtures between 278.00 K and 323.00K

        The solubility of xylitol in ethanol+water solvent mixtures was measured at temperatures ranging from 278.00 K to 323.00 K at atmospheric pressure by using a laser technique. The results of these measurements were correlated by the combined nearly ideal binary solvent CNIBS/Redlich-Kister equation. The experimental solubility and correlation equation in this work can be used as essential data and models in the purification process of xylitol. The variant 2 in the CNIBS/R-K models was confirmed to be more adaptable to predict solubility of xylitol in binary ethanol +water system. Using the experimentally measured solubilities, the thermodynamic properties of dissolution of xylitol,such as Gibbs energy, molar enthalpy of dissolution, and molar entropy of dissolution, were calculated.
