        Studies on the Intestinal Microflora of Chicken Under Tropical Condition

        Three media, i. e., MOD-SD, M98-5 and M98-5 supplemented with chicken fecal extract were tested as isolation media for anaerobic bacteria present in the duodenum, jeju-ileum and cecum of chicken. The results showed that the mean colony counts of medium M98-5 were similar with those of MOD-SD medium in all intestinal samples at the incubation periods of 2, 6 and 10 days. Supplementation with chicken fecal extract of M98-5 medium significantly increased (p < 0.05) the colony counts of bacteria from the duodenum, jeju-ileum and cecum. The colony counts at 6-day incubation were similar with those at 10-day incubation, but were much higher than the counts at 2-day incubation. The major types of bacteria found in the duodenum and jeju-ileum of chicken were tentatively identified as Lactobacillus, Streptococcus and E. coli. In the cecum, ten tentatively identified groups of bacteria, namely, Streptococcus, Staphylococcus, Lactobacillus, E. coli, anaerobic coccus, Eubacterium, Propionibacterium, Clostridium, Fusobacterium and Bacteroides were isolated. Anaerobes were found to comprise nearly the entire microbial population of the cecum. Predominating in all sections of the intestine were homofermentative lactobacilli. The main Lactotacillus species in chicken intestine were L. acidophilus, L. fermentum and L. brevis.

        Magnetic and Magneto-Optical Properties of Co-Sputtering FePt Films

        FePt thin .lms have been prepared at dierent substrate temperatures by DC magnetron co-sputtering. The .lms change from a disordered fcc structure to a L10 fct structure with temperature. The magnetic and the magneto-optical properties of those FePt .lms have been studied. The dependence of spectral changes on the formation of a chemically-ordered L10structure is observed in the polar magneto-optical Kerr rotation. In the spectra, the ultraviolet peak located in the high-energy range (4.0 { 4.5 eV), which is related to electronic transitions on Pt atomic sites, is observed for all samples. On the contrary, the peak related to transitions on Fe atomic sites, which is located in the low-energy range (1.5 { 2.0 eV), is only observed for highly chemically-ordered samples. Compared with the spectra for the L10 structure of FePt .lms pre- pared by using molecular beam epitaxy, the infrared peak related to Fe sites is shifted to low energy.

        Potato (Solanum tuberosum L. cv. Gogu valley) protein as an antimicrobial agent in the diets of broilers.

        <P>Two experiments were conducted to evaluate potato protein (PP, experiment 1) and refined PP (RPP, experiment 2) obtained from Gogu valley tubers as an antimicrobial agent in broiler diets. In both the experiments, 1-d-old male Ross 308 chicks were allotted to 5 treatments and performance, nutrient retention, and microbial populations in excreta and cecum were studied. Dietary treatments were as follows: basal diet (negative control, NC), basal diet with antibiotic (positive control, PC, 10 mg/kg of avilamycin), and low, medium, or high levels of PP (0.25, 0.50, and 0.75%, respectively, in experiment 1) or RPP (200, 400, and 600 mg/kg, respectively, in experiment 2). The overall gain and retention of DM (d 20 to 21) and CP (d 20 to 21 and d 41 to 42) were greater in birds fed PC and high PP diets than birds fed the NC diet. Population of total aerobic bacteria and coliforms was lowest in the cecum and excreta of birds fed the PC diet and highest in birds fed the NC diet. An increase in dietary PP linearly improved BW gain, feed intake, and feed conversion ratio during starter phase and overall BW gain. Also, there was linear improvement in retention of DM (d 20 to 21) and CP (d 20 to 21 and d 41 to 42) and reduced populations of total aerobic bacteria and coliforms in the cecum (d 42) and excreta (d 28 and 42) due to an increase in dietary PP. In the second experiment, the PC diet and diets with increasing levels of RPP had no effect on performance and nutrient retention. Birds fed the PC diet had the lowest microbial population in excreta and cecum, whereas the population of total aerobic bacteria and coliforms in excreta and cecum decreased (linear, P < 0.05) as the level of RPP was increased in the diet. These results suggest that both PP and RPP obtained from Gogu valley potato tubers have in vivo antimicrobial activity.</P>

        Species diversity and relative abundance of vaginal lactic acid bacteria from women in Uganda and Korea

        <P>Abstract</P><P>Aims: </P><P>Lactobacilli play an important role in maintaining vaginal health of women. The aim of this study was to compare the species richness and relative abundance of <I>Lactobacillus</I> and other lactic acid bacteria in women of two geographically distant countries, Uganda and Korea.</P><P>Methods and Results: </P><P>Vaginal samples were obtained from two women populations in Uganda and Korea. The <I>Lactobacillus</I> Rogosa SL agar was used for initial isolation of lactic acid bacteria. After phenotypic analyses, the 16S rRNA gene was amplified by polymerase-chain reaction and analysed by the BLAST program and phylogenetic tree construction. A total of 338 (128 Korean and 210 Ugandan) vaginal lactic acid bacterial strains were isolated, including five genera: <I>Lactobacillus</I>, <I>Leuconostoc</I>, <I>Pediococcus</I>, <I>Streptococcus</I> and <I>Weissella</I>. While <I>Lactobacillus crispatus</I> was common in both populations, <I>Lactobacillus fermentum</I> was common only in Korean women, and <I>Lactobacillus gasseri</I>, <I>Lactobacillus reuteri</I> and <I>Lactobacillus vaginalis</I> only in Ugandan women. Among other lactic acid bacteria, <I>Weissella</I> was more common in Ugandan, and <I>Pediococcus</I> in Korean women. All <I>Weissella</I> strains produced hydrogen peroxide, and all <I>Pediococcus</I> strains inhibited <I>Candida</I> species.</P><P>Conclusion: </P><P>Although many lactic acid bacteria colonize women, their species distributions may be different in women of geographically separated communities.</P><P>Significance and Impact of the Study: </P><P>The knowledge of species richness and relative abundance of vaginal lactic acid bacteria, including <I>Lactobacillus</I>, <I>Pediococcus</I> and <I>Weissella</I>, may lead to the design of better probiotic products as bacterial replacement therapy.</P>

        INFLUENCE OF DRIED Sacillus subtilis AND LACTOBACILLI CULTURES ON INTESTINAL MICROFLORA AND PERFORMANCE IN BROILERS

        Two hundred 10-day-lid, male Arbor Acres broiler chicks divided randomly into 4 groups of 50 chicks each were used. Different feeding treatment was carried out for each group. Chicks in treatment 1 were fed a basal diet(Starter feed)(control); treatment 2, a basal diet + 0.1% B. subtilis culture; treatment 3, a basal diet + 0.2% lactobacilli culture in the feed; and treatment 4, a basal diet + 5 g lactobacilli in the drinking water. The viable bacterial counts for each treatment were approximately $10^9cells/kg$ feed. The weight gain in chickens given feeds incorporated with B. subtilis and lactobacilli was significantly(p < 0.05) higher than those of the control. With regard to feed efficiency, there was a definite tendency towards a higher feed : gain lower(p < 0.05) feed : gain ratio. A significantly(p < 0.05) larger population of Lactobacillus was found in the small intestine of chickens fed with feed incorporated with B. subtilis at 21 and 28 days and with lactobacilli at 14, 21 and 28 days. Populations of intestinal E. coli in broilers given feed added with B. subtilis were not significantly(p < 0.05) different from those of the control, but in chickens fed lactobacilli-added feed, their populations wee significantly lower(p < 0.05) at 14 and 21 days. No significant differences were found among the treatments and the control in the occurrence of Salmonella and Campylobacter during the whole experimental period.

        Xylanase Supplementation Improved Digestibility and Performance of Growing Pigs Fed Chinese Double-low Rapeseed Meal Inclusion Diets: In vitro and In vivo Studies

        An in vitro and a feeding trial were conducted to investigate the effect of xylanase supplementation on the feeding value of growing pig diets containing high proportions of Chinese double-low rapeseed meals (DLRM). Seven diets were formulated to meet NRC (1998) nutrient requirements. Diet 1 based on corn-soybean meal was used as positive control 1, and diet 2, a practical diet which incorporated a conventional level of Chinese DLRM (60 g/kg diet), as positive control 2. Diet 3 contained a higher level of DLRM (100 g/kg diet) as the negative control. Diet 3 plus xylanase at 0.10, 0.25, 0.50 and 0.70 g/kg diet created diets 4, 5, 6 and 7, respectively. The seven diets were incubated in triplicate with the in vitro two-stage enzyme incubation method to predict responses of diets to xylanase in terms of digestibility of dry matter (DM), crude protein (CP) and neutral detergent fibre (NDF). In vitro, the negative control had the lowest CP and NDF digestibility. Both DM and CP digestibility were increased (p<0.05) owing to xylanase supplementation either at 0.50 or 0.70 g/kg diet, and NDF digestibility was improved following xylanase addition at all of the test levels. There was a high linear correlation ($r^2>90$, p<0.05) between the activity concentration of the enzyme when transformed into its logarithmic value and in vitro digestibility coefficients of DM, CP or NDF. In the feeding trial, 112 crossbred pigs were randomly assigned to seven dietary treatments with 16 replicate pens of one pig each. An obvious dose effect on growth rate was observed ($r^2=0.79$, p<0.05) within the inclusion levels of xylanase. Compared with the negative control, xylanase addition at 0.70 g/kg diet resulted in significantly increased ADG (878 g/d vs. 828 g/d, p<0.05), and a tendency towards improved growth rate (868 g/d vs. 828 g/d, p = 0.10) was also observed following the inclusion of xylanase at 0.50 g/kg diet. It would appear that the nutrient utilization of corn and Chinese DLRM diets by pigs could be enhanced by an appropriate amount of xylanase addition. The in vitro and in vivo results suggested that the in vitro incubation method is feasible for predicting responses of pigs to exogenous enzymes and identifying those preparations that possess potential for improvement of the nutritive values of feedstuffs.

        Synthesis and evaluation of a novel chiral derivatization reagent for resolution of carboxylic acid enantiomers by RP-HPLC

        <P>A novel derivatization reagent, N-[1-Oxo-5-(triphenylphosphonium)pentyl]-(S)-3-aminopyrrolidine (OTPA), with triphenylphosphine (TPP) as a basic structure carrying a permanent positive charge was developed for the enantiomeric separation of chiral carboxylic acids by high-performance liquid chromatography (HPLC). OTPA reacted with the carboxylic acids at 40 degrees C within 90 min in the presence of 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) and 1-hydroxybenzotriazole (HOBt). The degree of epimerization (racemization) during the derivatization reaction was negligible. The separability of the diastereomers was evaluated in terms of their resolution value (Rs). The Rs values of the derivatives of non-steroidal anti-inflammatory drugs (NSAIDs), which were selected as the representative carboxylic acids, were completely separated by reversed-phase chromatography using an ODS (4.6 mm x 150 mm I.D., 5.0 mu m) column. The resolution Rs values were 1.54-2.23 for the evaluated carboxylic acids and the OTPA-derivatization was also effective for the enantiomeric separation of chiral carboxylic acids. The calibration curves (r(2) > 0.9971) were linear over the concentration range of 0.0125-1.25 mM for each enantiomer of ketoprofen (KET), and naproxen (NAP), 0.05-1.0 mM for each enantiomer of ibuprofen (IBU), 2-phenylpropionic acid (PPA), and loxoprofen (LOX), and 0.05-1.25 mM for each enantiomer of PBA. The limit of detection (S/N = 3) for each of the enantiomers of the NSAIDs and chiral carboxylic acid enantiomers was 1.4-7.6 mu mol/L. The inter-day and intra-day assay precisions were all <6.77% and the mean recoveries (%) of the NSAIDs and chiral carboxylic acids from the spiked human plasma were 95.27-101.12%.         Acanthoic acid, a diterpene in Acanthopanax koreanum, protects acetaminophen-induced hepatic toxicity in mice

        The protective effect of a diterpenoid acanthoic acid (AA) isolated from Acanthopanax koreanum Nakai was investigated in acetaminophen (APAP)-induced hepatic toxicity. Drug-induced hepatotoxicity induced by an intraperitoneal (i.p.) injection of 300mg/kg (sub-lethal dose) of APAP. Pretreatment with AA (50 and 100mg/kg) orally 2h before the APAP administration attenuated the APAP-induced acute increase in serum aspartate aminotransferase (AST), and alanine aminotransferase (ALT) activites, replenished the depleted hepatic glutathione (GSH), superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-Px) activities, decreased malondialdehyde (MDA) level and considerably reduced the histopathological alterations in a manner similar to silymarin (Sily). Immunohistochemical analyses also demonstrated that AA could reduce the appearance of necrosis regions as well as caspase-3 and hypoxia inducible factor-1α (HIF-1α) expression in liver tissue. Our results indicated that AA protected liver tissue from the oxidative stress elicites by APAP-induced liver damage and suggestes that the hepatic protection mechanism of AA would relate to antioxidation and hypoxia factor on APAP-induced hepatotoxicity.

        China has a long history as an agricultural country, geographical indications of agricultural resources are very rich. If it has the protection of geographical indication legal system, will improve the quality and market competitiveness of agricultural products, thereby increasing farmers`` income. But China``s current status is in charge of the department set up to protect geographical indications on repeat, there is no independent legal system. These question not only for the protection of geographical indications Chinese impact on the efficiency, but also geographical indications of goods may have legitimate rights and interests conflict. In order to successfully solve these problems, it is necessary for the protection of geographical indications of goods related to the legal system and make changes, but there is a need for "Law on the Protection of geographical indications of goods." According to "Law on the Protection of geographical indications of goods" to build a management system and implementation of protection of geographical indications of goods.
