Effect of Storage Condition on Yield and Quality of Angelica acutiloba Radix
Choi,Seong-kyu,Yun,Kyeong-Won,Chon,Sang-Uk,Seo,Young-nam 한국자원식물학회 2002 Plant Resources Vol.5 No.1
To develop an optimal storage method of root of Angelica acutiloba Radix, which has been grown as major herbal medicine material to be cultivated, root of Angelica acutiloba Radix, was stored for 10 months with different packing materials and sealing methods. The loss in dry weight as influenced by packing materials and sealing methods was the lowest at vacuum packing and followed by complete sealing methods with transparent polyethylene film. The ratio of root rot during the storage period was not significantly different between packing materials but was significantly different between sealing conditions. In conclusion, vacuum packing and complete sealing with polyethylene film appears to be most optimal for storage of Angelica acutiloba Radix.

Choi,Seong-Kyu,Yun,Kyeong-Won,Chon,Sang-Uk,Seo,Young-Nam,Seo,Kyoung-sun,Kwon,Byung-Sun 한국자원식물학회 2003 Plant Resources Vol.6 No.3
This study was conducted to produce effectively more leaves of Angelica acutiloba through year-round production system by using various mulching materials and polyethylene(PE) tunnel. Soil temperature by covering treatment was higher in PE mulching and PE tunnel than in rice straw mulching, especially showing the highest temperature in PE tunnel from April through May. Water content of soil was higher in PE mulching than in non-mulching. Leaf production of Angelica acutiloba through year-round production showed the highest in PE tunnel and root production in PE mulching. These results suggest that leaf production of Angelica acutiloba can be maximized through PE tunnel cultivation system.

Choi,Seong-Kyu,Yun,Kyeong-won,Chon,Sang-Uk,Lee,Jong-Ill,Seo,Young-Nam,Seo,Kyoung-Sun,Choi,Kyeong-Ju 한국자원식물학회 2004 Plant Resources Vol.7 No.3
This study was conducted to develop effective production system in greenhouse for leaves and stems of Angelica acutiloba by using connect pots in 2003 and 2004. Seed germination rate and plant biomass of Angelica acutiloba collected in 2004 were higher than those harvested in 2003. Germination rate of Angelica acutiloba seeds collected in 2003 was 10%, while germination rate of seeds collected in 2004 was above 90%. Especially, plant growth and yield of Angelica acutiloba grown in connect pots sized with 4×4×5cm(length×width×height) were the highest. These results indicate that leaf and stem production of Angelica acutiloba can be improved by using connect pots and optimizing seed collecting time in greenhouse.

Choi,Seong-Kyu,Yun,Kyeong-Won,Chon,Sang-Uk,Seo,Young-Nam,Seo,Kyoung-Sun,Kwon,Byung-Sun 한국자원식물학회 2004 Plant Resources Vol.7 No.2
Activate charcoal (AC) can be utilized as a soil conditioner in agricultural crop areas. This study was conducted to investigate the effect of AC on growth and yield of Zingiber mioga ROSC as affected by different amounts of AC. The results obtained are summarized as follows. Growth characteristics including plant height and leaf length were the highest when activated charcoal added with 10-30%, suggesting that optimum amount of activated charcoal was ranged from 10 to 30%. Growth and enlargement of the root were improved by 10% AC with higher rhizome length and weight.

This paper is a study on how critical terms has been discussed and converged in the early modern Korean literary circles. For this study, I focused on the concept of ‘description’ in the debate between Yum, Sang-sub and Kim, Dong-in. In order to re-examine this debate, productive intent is necessary. The countless signs of misunderstanding which was found in those texts is related to the confusion about the concept of terms. It is the general situation of early modern literary circles rather than a matter of Yum and Kim. They had trouble applying western literary theory in Korean language. This paper noted the ‘description’ among several critical terms, because it was the very hot issue of the early modern novel criticism. Yum and Kim emphasized description concept associated with essential attributes of ‘New’ literature like other critics. But the concept of description they thought is different from each other. Kim preferred to describe concisely by grasping the essence of the target, compared to Yum preferred detailed and rich description. It should be noted that they tried to create novel texts in order to show their own concept of description than to explain a theory. Because this debate may be interpreted as the main motivation producing major legacy of the history of early modern novels.

To construct transfurmed Bm5 cells, Autographa californica nuclear polyhedrosis virus(AcNPV) IE1 gene, an immediate early viral gene was firstly used in this study. AcNPV IE1 gene, which shares on 95.3% nucleotide sequence homology with Bombyx mori nuclear polyhedrosis virus (BmNPV) IE1 gene, was isolated and cloned into pBluescript. Neomycin gene from pKO-neo was inserted under the control of the IE1 promoter to yield pAcIE1-neo. The plasmid pAcIE1-neo was transfected into Bm5 or Sf9 cells, and neomycin-resistant cells were selected in TC100 medium containing 10% fetal bovine serum (FBS) and 1 mg/ml G4l8 for two weeks. Individual clones were picked and each was amplified for further characterization. The genomic DNA from neomycin-resistant cells was isolated and characterized by PCR using AcNPV IEI gene-specific primers and by Southern blot analysis using neomycin gene probe. We concluded that AcNPV IE1 gene was functional in B. mori-derived Bm5 cells as well as Spodoptera fugjprrda-derived Sf9 cells to produce stably-transformed insect cells

Choi, Chi-Won,Park, Edmond Changkyun,Yun, Sung Ho,Lee, Sang-Yeop,Lee, Yeol Gyun,Hong, Yeonhee,Park, Kyeong Ryang,Kim, Sang-Hyun,Kim, Gun-Hwa,Kim, Seung Il American Chemical Society 2014 JOURNAL OF PROTEOME RESEARCH Vol.13 No.10

Outer membrane vesicles (OMVs) are produced by various pathogenic Gram-negative bacteria such as Escherichia coli, Pseudomonas aeruginosa, and Acinetobacter baumannii. In this study, we isolated OMVs from a representative soil bacterium, Pseudomonas putida KT2440, which has a biodegradative activity toward various aromatic compounds. Proteomic analysis identified the outer membrane proteins (OMPs) OprC, OprD, OprE, OprF, OprH, OprG, and OprW as major components of the OMV of P. putida KT2440. The production of OMVs was dependent on the nutrient availability in the culture media, and the up- or down-regulation of specific OMPs was observed according to the culture conditions. In particular, porins (e.g., benzoate-specific porin, BenF-like porin) and enzymes (e.g., catechol 1,2-dioxygenase, benzoate dioxygenase) for benzoate degradation were uniquely found in OMVs prepared from P. putida KT2440 that were cultured in media containing benzoate as the energy source. OMVs of P. putida KT2440 showed low pathological activity toward cultured cells that originated from human lung cells, which suggests their potential as adjuvants or OMV vaccine carriers. Our results suggest that the protein composition of the OMVs of P. putida KT2440 reflects the characteristics of the total proteome of P. putida KT2440.

Graphic Abstract

ACS Electronic Supporting Info

Effects of Polyethyleneimine on the Sonochemical Synthesis of Gadolinium Ion-Modified ZnO Nanorods.
Choi, Seok Cheol,Yun, Won Suk,Sohn, Sang Ho American Scientific Publishers 2015 Journal of nanoscience and nanotechnology Vol.15 No.1

We prepared gadolinium (Gd) ion-modified ZnO nanorods by a sonochemical decomposition of zinc acetate dehydrate and gadolinium acetate hydrate precursor solutions with and without polyethyleneimine (PEI). We investigated the effects of PEI on the sonochemical synthesis of ZnO nanorods with and without Gd ion modifications. In the case of nascent ZnO nanorods, PEI in the precursor solutions can prohibit radial growth but allow axial growth, resulting in changes in the degree of preferred crystal orientations, and in the PL properties of the resulting nanorods. In the case of Gd ion-modified ZnO nanorods, we observed that the ZnO nanorods, fabricated sonochemically in the precursor solutions with PEI, exhibited a peak broadening of the ZnO(002) crystal plane and decreasing crystal orientation with respect to the c plane. We note that PEI can negatively affect the crystal orientation and crystallinity of Gd ion-modified ZnO nanorods, even though it cannot affect the lattice constant.
