        Phosphorylation of DYNLT1 at Serine 82 Regulates Microtubule Stability and Mitochondrial Permeabilization in Hypoxia

        Hypoxia-induced microtubule disruption and mitochondrial permeability transition (mPT) are crucial events leading to fatal cell damage and recent studies showed that microtubules (MTs) are involved in the modulation of mitochondrial function. Dynein light chain Tctex-type 1 (DYNLT1) is thought to be associated with MTs and mitochondria. Previously we demonstrated that DYNLT1 knockdown aggravates hypoxia-induced mitochondrial permeabilization, which indicates a role of DYNLT1 in hypoxic cytoprotection. But the underlying regulatory mechanism of DYNLT1 remains illusive. Here we aimed to investigate the phosphorylation alteration of DYNLT1 at serine 82 (S82) in hypoxia (1% O2). We therefore constructed recombinant adenoviruses to generate S82E and S82A mutants, used to transfect H9c2 and HeLa cell lines. Development of hypoxia-induced mPT (MMP examining, Cyt c release and mPT pore opening assay), hypoxic energy metabolism (cellular viability and ATP quantification), and stability of MTs were examined. Our results showed that phosph-S82 (S82-P) expression was increased in early hypoxia; S82E mutation (phosphomimic) aggravated mitochondrial damage, ele-vated the free tubulin in cytoplasm and decreased the cellular viability; S82A mutation (dephosphomimic) seemed to diminish the hypoxia-induced injury. These data suggest that DYNLT1 phosphorylation at S82 is involved in MTs and mitochondria regulation, and their interaction and cooperation contribute to the cellular hypoxic tolerance. Thus, we provide new insights into a DYNLT1 mechanism in stabilizing MTs and mitochondria, and propose a potential therapeutic target for hypoxia cytoprotective studies.

        Analysis of the Supervision of Ecological Subsidies

        In view of the problem of the invalidity of the incentive mechanism of the ecological subsidies, which is due to the information asymmetry between the government``s supervision and the enterprise endeavor to fulfill their ecological responsibilities, this paper attempts to analyze the supervision of ecological subsidies based on the Principal-agent Model. Two conclusions are drawn: firstly, the government``s supervision regarding the effect of the enterprises`` fulfilling the ecological responsibilities can significantly reduce the information asymmetry; secondly, the government``s incentive strength and the enterprises`` endeavor level of fulfilling the ecological responsibilities are both improving the surveillance dynamics. Here is the suggestion: with the increasing of the surveillance dynamics of the government and the transparency of the enterprises`` fulfilling the ecological responsibilities, the government should meanwhile increase the subsidies incentive strength, therefore, to promote the effort level of the enterprises`` fulfilling the ecological responsibilities to approach to the Pareto optimal value.

        Simultaneous Determination of Hydroquinone and Catechol by N-doped Porous Biochar-modified Electrode

        N-doped porous biochar (NPB) with large conjugated systems could not only be used as enrichment carriers but also be in favor of electron transport in the electrochemical detection. The NPB-modified electrode was fabricated for the simultaneous detection of catechol (CA) and hydroquinone (HQ) to enhance the signal-to-noise ratio and further improve the sensitivity. A detection limit as low as 37 and 47?nM was achieved for CA and HQ, respectively. The proposed strategy with the merits of high sensitivity, selectivity, and reproducibility exhibited a great potential for the detection.

      • Relationships of uPA and VEGF Expression in Esophageal Cancer and Microvascular Density with Tumorous Invasion and Metastasis

        Objective: To investigate uPA and VEGF expression in esophageal cancer and relations with tumorous invasion and metastasis. Methods: Immunohistochemistry was used to detect uPA and VEGF expression in the normal epithelial tissue of esophageal mucosa and cancer tissue and detect CD34 labeled micrangium and analyze the relationships with clinical pathological features and tumor angiogenesis. Results: Positive rates for uPA and VEGF protein expression were significantly greater in esophageal cancer than normal epithelial tissue (P < 0.05), the two being linked (P <0.05). In addition, uPA and VEGF protein expression of the high microvessel density (MVD) group was significantly lower than in the low MVD group (P < 0.05), with relation to clinical pathological staging, differentiation and lymph node metastasis (P < 0.05). Conclusion: In esophageal cancer tissue, uPA and VEGF proteins are overexpressed and promote tumor angiogenesis, indicative of a poor prognosis.

        Growth and replication of ordered ZnO nanowire arrays on general flexible substrates

        <P>Vertically aligned and site controllable ZnO nanowire arrays have been synthesized and replicated <I>via</I> hydrothermal method on general flexible substrates. The replication was demonstrated for three generations. The morphology and density of the nanowire arrays could be optimized in the original generation by adjusting the chemical reaction parameters. The pattern of the original generation was inherited by the succeeding generations by a new transferring method. The growth mechanism of the replicated nanowire arrays was investigated with the help of inductively coupled plasma (ICP) etching and AFM tip scanning. The robustness of the ZnO nanowire arrays was obviously improved compared with the seedless ZnO nanowires grown on Au (111) surface, according to the excellent morphology preservation after ultrasonic wave treatment.</P> <P>Graphic Abstract</P><P>Vertically aligned and site controllable ZnO nanowire arrays have been synthesized and replicated <I>via</I> hydrothermal method on general flexible substrates.         Synthesis and Cytotoxicity of Novel 3-Amino-4-indolylmaleimide Derivatives

        In an attempt to develop potent and selective antitumor agents, a series of novel 3-amino-4-indolylmaleimides were designed and synthesized. The reaction showed high regioselectivity. The structure of compound 7a was determined by an X-ray single crystal diffraction method. The cytotoxicities of the title compounds were evaluated against HeLa, SMMC 7721 and HL 60 cancer cell lines by a standard MTT assay in vitro. The pharmacological results showed that some of the title compounds displayed moderate or high cytotoxic activity against the tested cell lines. Compound 7d was the most promising compound against the tested cancer cell lines. Structure-activity relationships are discussed based on the experimental data obtained. A hydroxyethylamino group at the 3-position in the side chain of indolylmaleimide is associated with an increase in cytotoxicity.

      • Risk Factors for Nodal Metastasis in cN0 Papillary Thyroid Microcarcinoma

        Background: Despite the majority of papillary thyroid microcarcinoma (PTMC) patients having an excellent prognosis, cervical lymph node metastases are common. The purpose of this study was to investigate the incidence and the predictive risk factors for occult central compartment lymph node metastasis (CLNM) in PTMC patients. Materials and Methods: 178 patients with clinically node-negative (cN0) PTMC undergoing prophylactic central compartment neck dissection in our hospital from January 2008 to Jun 2010 were enrolled. The relationship between CLNM and the clinical and pathological factors such as gender, age, tumor size, tumor number, tumor location, extracapsular spread (ECS), and coexistance of chronic lymphocytic thyroiditis was analyzed. Results: Occult CLNM was observed in 41% (73/178) of PTMC patients. Multivariate analysis showed that male gender, tumor size (${\geq}6mm$) and ECS were independent variables predictive of CLNM in PTMC patients. Conclusions: Male gender, tumor size (${\geq}6mm$) and ECS were risk factors of CLNM. We recommend a prophylactic central lymph node dissection (CLND) should be considered in PTMC patients with such risk factors.

        Synthesis of hexagonal boron nitride thin film on Pt substrates for resistive switching memory applications

        Hexagonal boron nitride (hBN), due to its high reliability as a two-dimensional (2D) dielectric material, has attracted much attention for its potential applications in nanoelectronic devices. Here, the use of radio frequency (RF) magnetron sputtering-grown hBN films to construct hBN-based resistive switching (RS) memory device is reported, and the RS mechanism is deduced. The hBN-based RS memory shows low operating voltage, reproducible write cycles, and long retention time. First-principles simulations further confirm the resistive switching. This work provides an important case to facilitate the future applications of 2D materials in the RS memory.

        Evaluation of the susceptibility of coal to spontaneous combustion by a TG profile subtraction method

        −It is imperative to have an in-depth understanding of the intrinsic reaction between coal and oxygen during low-temperature oxidation, as the reaction is the main source responsible for the self-heating and spontaneous combustion of coal. As low-temperature oxidation of coal involves a series of physical and chemical process and many parallel reactions, it is difficult to directly investigate the intrinsic reaction between coal and oxygen by conventional analytical method. Thermogravimetric analysis (TGA) was used to investigate the intrinsic reaction between coal and oxygen based on the mass change. By means of the subtraction analysis method of TGA, the TG-subtraction curves were obtained by subtracting the TG-N2 curves from the TG-air curves. The results indicate that a TG-subtraction curve can better reflect the intrinsic reaction of coal oxidation than a TG-air curve by eliminating the influence of evaporation of water and thermal decomposition of inherent oxygen-containing groups. In terms of the rate of mass increase, the intrinsic reactions can be divided into three stages: slow oxidation stage, advanced oxidation stage and rapid oxidation stage. The activation energy at each of the stages, obtained by Coats and Redfern’s model, can be used to as a technical parameter to evaluate the proneness of coal spontaneous combustion. The optimum experiment conditions were also developed to study low-temperature coal oxidation with the subtraction method of TGA.
