      • Insulin can block apoptosis by decreasing oxidative stress via phosphatidylinositol 3-kinase- and extracellular signal-regulated protein kinase-dependent signaling pathways in HepG2 cells

        Objective : Insulin has well-known activities in controlling energy metabolism, cellular proliferation and biosynthesis of functional molecules to maintain a biological homeostasis. Recently, several studies have suggested that insulin may protect cells from apoptosis in different cell lines: however, little is known about the nature of its anti-apoptotic actigity. In many clinical disorders, including type 2 diabetes mellitus, oxidative stress and the production of reactive oxygen species (ROS) is increased. With these facts as a background, we examined here whether insulin protects HepG2 cells from apoptosis by decreasing oxidative stress and, if so, which signaling steps are involved in this process. Methos : Intracellular DNA content, the degree of nuclear condensation or poly(ADP-ribose) polymerase hydrolysis was measured to verify the occurrence of apoptotic events. Caspase-3 activity and ROS accumulation within cells were also measured. Western blot analysis was performed to identify fignaling molecules activated in response to insulin. Results : Serum starvation resulted in a marked accumulation of ROS, activation of caspase-3, and subsequent apoptotic cell death which were, in turn, markedly blocked by the addition of insulin. The anti-apoptotic activity of insulin was sensitive to blockade of two different signaling steps, activations of phosphatidylinosltol 3-kinase (PI3 kinase) and extracellular signal-regulated protein kinase (ERK). Conclusion : Insulin exerts and anti-apoptotic activity by suppressing the excessive accumulation of ROS within cells though signaling pathways including stimulation of PI3 kinase and ERK in HepG2 cells.

      • 기업의 ESG 경영과 인권―일본의 인권규범 정비동향을 참고로

        기업이 환경 및 인권과 관련된 사회적 책임을 완수하고 있는지, 또한 적절한 지배 구조를 구축하였는지를 고려하고 투자행동을 하는 투자자의 시선을 의식하지 않을 수 없는 ESG 경영의 시대가 되었다. 특히 ESG 투자에 대한 관심의 증대와 함께 Social의 핵심이라 할 수 있는 인권 및 노동에 대한 인식이 예전보다 중시되고 기업이 인권의 존중을 위해 노력해야 한다는 인식이 국제적으로도 확산되고 있다. 본고에서는 기업과 인권 관련 국제적 표준 수립 동향을 시작으로 일본의 인권존중 관련 제도정비의 과정 및 최근 입법동향, 일본기업들의 인권규범 도입관련 인식, 최근 일본의 인권규범 관련 추진노력이 주는 의미와 우리나라의 인권규범 도입 상황과 향후 대응방향 등에 대하여 살펴본다. 본래 근대적 인권의 개념은 국가에 의한 인권침해를 전제로 한 것이었지만, 차츰 사적 영역의 주체들이 인권을 침해하는 상황이 생겨나면서 인권존중책임의 주체는 기업을 포함한 사적 영역으로 확대되었다. 특히 1990년대 이후, 다국적 기업에 의한 인권침해의 문제가 대두되면서 기업의 사회적 책임에 관한 국제기준이 논의되고, 국제기구차원의 인권 관련 규범화 노력이 진행되었다. 과거에는 인권존중 및 보호가 국가의 책무로만 인식되던 것이 기업과 같은 사적 주체의 책무로 파악할 필요성이 생겨 났기 때문이다. 기업들의 국경을 초월한 인권침해와 관련하여 가장 중요한 규범이 유엔이 제정한 「비즈니스와 인권에 관한 이행원칙」이고 이를 수용한 국가들이 행동계획(NAP)을 수립한다. 2013년에 영국이 세계 최초로 행동계획(NAP)을 수립하였고, 이후 서방국가들에 이어 2020년 10월에는 일본이 「비즈니스와 인권에 관한 행동계획(2020∼2025)」을 공표하였다. 이러한 모습은 인권문제에 적극적으로 대응하는 것이 기업의 중장기적 이익의 확보에도 도움이 된다는 인식이 국제사회에서도 공감대를 형성하고 있다는 것을 보여준다. 그런데 인권의 보장을 통해 지속 가능한 사회를 실현하는 것은 정부의 역할만으로는 부족하고 기업들이 경영에 있어서 인권의 중요성에 대해 관심을 가져야 한다. 사회에서 기업의 영향력이 커지면 커질수록 기업의 책임에 대한 사회적 기대가 높아지게 되고 기업과 인권의 문제는 회피할 수 없는 기업경영의 중요한 과제가 되기 때문이다. 더욱이 종래에는 기업의 경영에서 인권의 중요성이 간과되어 왔지만, ESG 투자가 확대되는 상황에서는 기업의 인권존중책임이 강조될 수밖에 없으므로 기업들이 인권경영을 적극적으로 추구하는 것이 바람직하다. The era of so-called ESG management has arrived, where ESG investment, which considers whether a company is fulfilling its social responsibilities related to the environment and human rights, and whether it has established an appropriate governance structure, has become common. With the growing interest in ESG investment, the awareness of human rights and labor, which can be said to be the core of social part, is more important than ever before, and the idea that companies should strive to respect human rights is spreading and common internationally. In this paper, starting with the trend of establishment of international standards related to business and human rights, the process of institutional reform related to human rights respect in Japan and recent legislative trends, recognition of the introduction of human rights standards by Japanese companies, and the significance of Japan's recent efforts to promote human rights standards are examined. Next, we will look at the situation in Korea related to the revision of human rights norms and the direction of future response. Originally, the concept of modern human rights was based on the premise of human rights violations by the state, but gradually, as a situation emerged where subjects in the private sector were violating human rights, the subject of responsibility for respecting human rights expanded to the private sector including corporations. In particular, since the 1990s, as the problem of human rights violations by multinational corporations emerged, international standards for corporate social responsibility were discussed, and efforts were made to standardize human rights at the international level. This is because, in the past, the issue of respect for and protection of human rights was recognized only as the responsibility of the state, but the need to understand it as the responsibility of private entities such as corporations has increased. The most important norm in relation to transboundary human rights violations by companies is the 「Guiding Principles on Business and Human Rights」 established by the United Nations, and countries that have accepted it establish action plans (NAPs). In 2013, the UK established the world's first Action Plan (NAP), followed by Western countries, and in October 2020, Japan announced the 「Action Plan on Business and Human Rights (2020-2025)」. This figure shows that the international community is also forming a consensus on the perception that actively responding to human rights issues is helpful in securing mid- to long-term profits for companies. However, the role of the government alone is not enough to realize a sustainable society through the guarantee of human rights, and companies must pay attention to the importance of human rights in management. This is because the greater the influence of a company in society, the higher the social expectations for corporate responsibility, and the issue of business and human rights becomes an unavoidable issue of corporate management. Moreover, although the importance of human rights in business management has been overlooked in the past, it is desirable for companies to actively pursue human rights management because the corporate responsibility to respect human rights is inevitably emphasized in a situation where ESG investment expands.

      • Effects of high occupational physical activity, aging, and exercise on heart rate variability among male workers

        <P><B>Objectives</B></P><P>Effects of aging and leisure time physical activity (LPA) might influence the effect of occupational physical activity (OPA) on risk for cardiovascular disease (CVD). This study was conducted to determine whether OPA affects CVD after controlling the effects of LPA and other risk factors for CVD such as job stress.</P><P><B>Methods</B></P><P>Participants were 131 male Korean manual workers. Tests for heart rate variability (HRV) were conducted for five minutes in the morning at work. We defined OPA as the combined concept of relative heart rate ratio (RHR), evaluated using a heart rate monitor.</P><P><B>Results</B></P><P>Whereas high OPA was not related to any HRV items in the younger age group, high OPA was associated with an increased number of low-value cases among all HRV items in older workers. Exercise had beneficial effects only in the younger group. After controlling for exercise and other risk factors, the odds ratios of the root-mean square of the difference of successive normal R-R intervals (rMSSD) and high frequency band power (HF) among the older age and high OPA group compared with the younger age and low OPA group were 64.0 and 18.5, respectively. Social support and shift work were independent risk factors in HRV.</P><P><B>Conclusions</B></P><P>OPA in aging workers increases CVD risks. This study provides support for the need for protection of aging workers from physical work overload, and indicates the need for further study of optimal limits of OPA.</P>

      • Eckol isolated from <i>Ecklonia cava</i> attenuates oxidative stress induced cell damage in lung fibroblast cells

        <P><B>Abstract</B></P><P>We have investigated the cytoprotective effect of eckol, which was isolated from <I>Ecklonia cava</I>, against oxidative stress induced cell damage in Chinese hamster lung fibroblast (V79-4) cells. Eckol was found to scavenge 1,1-diphenyl-2-picrylhydrazyl radical, hydrogen peroxide (H<SUB>2</SUB>O<SUB>2</SUB>), hydroxy radical, intracellular reactive oxygen species (ROS), and thus prevented lipid peroxidation. As a result, eckol reduced H<SUB>2</SUB>O<SUB>2</SUB> induced cell death in V79-4 cells. In addition, eckol inhibited cell damage induced by serum starvation and radiation by scavenging ROS. Eckol was found to increase the activity of catalase and its protein expression. Further, molecular mechanistic study revealed that eckol increased phosphorylation of extracellular signal-regulated kinase and activity of nuclear factor κ B. Taken together, the results suggest that eckol protects V79-4 cells against oxidative damage by enhancing the cellular antioxidant activity and modulating cellular signal pathway.</P>

        Effect of Workload on Musculoskeletal Degeneration

        Objective: The aim of this study is to investigate the effect of workload on musculoskeletal degeneration. Background: It is important that workers maintain a health body for a long time as a measure of aging workers. In order do so, it is necessary to find out if workload can effect on musculoskeletal degeneration. Method: Epidemiological studies on workload or occupation and disc degeneration, osteoarthritis of knee were identified through database and bibliography searches. Results: Epidemiological studies showed that workload certainly effect on musculoskeletal degeneration. But we couldn"t know which of individual factors and occupational factors further contribute to the musculoskeletal degeneration. And we could find that studies on workload and genetic factors were very few. In addition, there was also very few studies that it was possible interaction between individual factors as obesity and occupational factors as heavy manual lifting. Conclusion: Our reviews suggest that it need to conduct study between workload and genetic factors for musculoskeletal degeneration. This further study can identify attributable risk of workload for musculoskeletal degeneration, and contribute to the measure of aging workers. Application: This paper can help to establish research plan for the measure of aging workers.

        Reconstruction of the Korean Asbestos Job Exposure Matrix

        Background: A job-exposure matrix (JEM) is an important surrogate indicator to evaluate past exposure levels. Although a Korean asbestos JEM has been constructed previously, this JEM includes only a few industrial and occupational groups. This study aimed to reconstruct the JEM by integrating the latest organized data to improve its utility. Methods: We used recent Korean standard industry and occupation codes and extracted 36 articles from a systematic literature review to initiate the reconstruction of the previous Korean asbestos JEM. The resulting data consisted of 141 combinations of industrial and occupational groups. Data from the Netherlands's JEM were also reviewed and categorized into 70 industrial and 117 occupational groups by matching with the Korean data. We also utilized Germany's data, which consisted of 10 industrial and 14 occupational groups. Results: The reconstructed Korean asbestos JEM had 141 combinations of industries and occupations. The time periods are from the 1980s to the 2000s in 10-year intervals. Most of the data were distributed between the 1990s and the 2000s. Occupations with high exposure to asbestos included knitting and weaving machine operators, automobile mechanics or assemblers, ship mechanics or assemblers, mineral ore and stone products processing mechanics, and metal casting machine operators or mold makers. Conclusions: The reconstructed Korean asbestos JEM has expanded the type and duration of the occupational groups of the previous JEM and can serve as an important reference tool for evaluating asbestos exposure and designing compensation and prevention policies in Korea.
