Mining of Serum Glycoproteins by an Indirect Approach Using Cell Line Secretome

Glycosylation is the most important and abundant post-translational modification in serum proteome. Several specific types of glycan epitopes have been shown to be associated with various types of disease. Direct analysis of serum glycoproteins is challenging due to its wide dy-namic range. Alternatively, glycoproteins can be disco-vered in the secretome of model cell lines and then con-firmed in blood. However, there has been little experi-men-tal evidence showing cell line secretome as a tractable target for the study of serum glycoproteins. We used a hydrazine-based glycocapture method to selectively enrich glycoproteins from the secretome of the breast cancer cell line Hs578T. A total of 132 glycoproteins were identified by nanoLC-MS/MS analysis. Among the identified proteins, we selected 13 proteins that had one or more N-gly-cosylation motifs in the matched peptides, which were included in the Secreted Protein Database but not yet in the Plasma Proteome Database (PPD), and whose antibod-ies were commercially available. Nine out of the 13 se-lected proteins were detected from human blood plasma by western analysis. Furthermore, eight proteins were also detected from the plasma by targeted LC-MS/MS, which had never been previously identified by data-dependent LC-MS/MS. Our results provide novel proteins that should be enrolled in PPD and suggest that analysis of cell line secretome with subfractionation is an efficient strategy for discovering disease-relevant serum proteins.

<P><B>Abstract</B></P> <P><B>Background</B></P> <P>Atopic dermatitis (AD) is a chronic relapsing eczematous skin disease with an increasing prevalence. In Korea, Jaungo is a traditional medicinal ointment, which is commonly used for treating skin wounds.</P> <P><B>Objective</B></P> <P>Thus, we aim to explore the basic clinical efficacy and safety data for Jaungo in patients with AD.</P> <P><B>Methods</B></P> <P>This study was a pilot randomized, double-blind, placebo-controlled, single-center trial with three groups. The participants in treatment group 1 applied Jaungo to the lesion twice a day for 3 weeks. Those in treatment group 2 applied Jaungo and placebo ointments to the lesion once a day for 3 weeks. Those in the placebo group applied placebo ointments to the lesion twice a day for 3 weeks. The Eczema Area and Severity Index and SCORing Atopic Dermatitis scores, transepidermal water loss value, and Dermatology Life Quality Index score were assessed. The outcomes used to evaluate safety were the Draize score, blood test results, and expert opinion.</P> <P><B>Results</B></P> <P>In total, 28 patients (82.4%) completed the study. During the study, significant decline of EASI scores in treatment group 2 and placebo group was observed (p < 0.05). And there was significant decline of SCORAD scores in treatment group1 and placebo group (p < 0.05). Patients in all groups showed decreased TEWL and DLQI scores with no significant difference. There was significant decline of IL-17 in all groups (p < 0.05). No serious adverse event was observed.</P> <P><B>Conclusion</B></P> <P>This is the first study that has explored the potential therapeutic effect of Jaungo as a complementary therapy for AD. However, further large study with adjusting placebo ingredients is needed to confirm the effectiveness of Jaungo in patients with chronic-phase AD.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Jaungo is topical herbal agents most widely used in tradition Korean Medicine, especially used for skin dryness and allergic skin diseases. </LI> <LI> Jaungo and its three carrier oils may have the potential therapeutic effect in patients with chronic-phase AD, especially on their dry skin. </LI> <LI> This is the first study that has explored the potential therapeutic effect of Jaungo as a complementary therapy for AD. </LI> <LI> However, further large study with adjusting placebo ingredients is needed to confirm the effectiveness of Jaungo in patients with AD. </LI> </UL> </P>

<P>Many hydroxyflavone derivatives have been found in nature and shown to have many biological functions. Because their function is changed by the position and number of hydroxyl group, their structural identification is a fundamental and necessary step for understanding their functions. In the present study, the complete <SUP>1</SUP>H and <SUP>13</SUP>C NMR spectral assignments were presented for 6 hydroxyflavones, and NMR data of additional 14 hydroxyflavone derivatives were compared with those of the 6 hydroxyflavones. In addition, the partially incorrect NMR data of two of the dihydroxyflavones whose NMR data were previously reported were corrected. Copyright © 2007 John Wiley & Sons, Ltd.</P>
유아교사의 행복감에 대한 직무스트레스와 사회적 지지의 영향

본 연구는 유아교사의 행복감에 직무스트레스와 사회적 지지가 미치는 영향을 조사하였다. 이를 위해 경기도와 인천시 지역의 유아교사 265명을 대상으로 설문지를 통해 자료 수집을 하였다. 자료 수집은 행복감, 직무스트레스, 사회적 지지 검사 도구를 사용하였으며, 자료처리는 SPSS 18.0 프로그램을 이용하여 Pearson의 적률상관분석과 단계적 중다회귀분석을 실시하였다. 그 결과는 다음과 같다. 첫째, 유아 교사의 행복감과 직무스트레스는 부적 관계가 있는 것으로 나타났으며, 행복감과 사회적 지지는 정적 관계가 있는 것으로 나타났다. 둘째, 유아 교사들의 행복감에 직무스트레스와 사회적 지지가 미치는 상대적 영향력을 살펴보면, 직무스트레스의 하위 요인인 원아들과의 활동과 사회적지지의 하위 요인인 평가적지지가 행복감에 유의미한 영향력을 미치고 있었다. 즉, 유아교사의 직무스트레스와 사회적지지는 유아교사의 행복감을 설명해주는 중요한 변인들이라는 것을 알 수 있다. 이러한 결과는 유아교사의 직무스트레스를 감소시키고 행복감을 높일 수 있도록 하기 위한 사회적 지지 방안을 모색할 필요가 있다는 것을 시사해 준다. This study examined the impact teacher`s of job stress and social supporst on early cuildhood educationteachers` happiness. To that end, data was collected through questionnaires from 265 early childhood teachers in Gyeonggido and Incheon. Data collection used happiness, job stress, social support test tools and data processing was carried out the Pearson`s product-moment correlation analysis and stepwise multiple regression analysis using SPSS 18.0 program. The results are as follows : First, it appeared that happiness of early cuildhood education teachers was a negative correlation with their job stress and a positive correlation with social support. Second, the importance of the job stress and social support of job stress, which is a subordinate factor of job stress and social support, has had significant implications for the happiness of job stress and social support. It turns out That job stress and social support are critical variables in explaining the happiness of the early cuildhood education teachers. These results suggest that it is necessary to explore ways to reduce the job stress of early cuildhood education teachers and encourage them to increase their sense of happiness.
