<P>Live-cell-based biosensors have emerged as a useful tool for biotechnology and chemical biology. Genetically encoded sensor cells often use bimolecular fluorescence complementation or fluorescence resonance energy transfer to build a reporter unit that suffers from nonspecific signal activation at high concentrations. Here, we designed genetically encoded sensor cells that can report the presence of biologically active molecules via fluorescence-translocation based on split intein-mediated conditional protein trans-splicing (PTS) and conditional protein trans-cleavage (PTC) reactions. In this work, the target molecules or the external stimuli activated intein-mediated reactions, which resulted in activation of the fluorophore-conjugated signal peptide. This approach fully valued the bond-making and bond-breaking features of intein-mediated reactions in sensor construction and thus eliminated the interference of false-positive signals resulting from the mere binding of fragmented reporters. We could also avoid the necessity of designing split reporters to refold into active structures upon reconstitution. These live-cell-based sensors were able to detect biologically active signaling molecules, such as Ca<SUP>2+</SUP> and cortisol, as well as relevant biological stimuli, such as histamine-induced Ca<SUP>2+</SUP> stimuli and the glucocorticoid receptor agonist, dexamethasone. These live-cell-based sensing systems hold large potential for applications such as drug screening and toxicology studies, which require functional information about targets.</P> [FIG OMISSION]</BR>
Characteristics of Korean Trauma Patients: A Single-center Analysis Using the Korea Trauma Database

Purpose: Two years have passed since a level I trauma center was officially opened in the Gacheon Gil Hospital, South Korea. We analyzed 2014 and 2015 registered patient data from the Korean Trauma Data Base (KTDB) to identify trends in trauma patient care and factors that influence the quality of trauma care at the Gacheon Gil trauma center. Methods: Data was extracted from the KTDB included patient age, sex, systolic blood pressure at emergency room arrival, revised trauma score, injury severity score, trauma injury severity score, transfusion amount, and the cause of death was analyzed. Results: A total of 3269 trauma patients were admitted to our trauma center in 2014 and 3225 in 2015. Demographics and mechanism of injury were not significantly different between years. The severity of trauma injury was decreased in 2015 although the mortality rate was slightly increased. This requires further analysis. Conclusion: The aim of this study was to determine the general status and trends in trauma incidence and management outcomes for the Incheon area. We noted no significant changes in trauma status from 2014 to 2015. We need to collect and review trauma patient data over a long period in order to elucidate trauma incidence and management trends in the trauma field. Finally, studies using trauma patient data will indicate appropriate quality control factors for trauma care and help to improve the quality of trauma management. [ J Trauma Inj 2016; 29: 155-160 ]
Major Causes of Preventable Death in Trauma Patients

Purpose: Trauma is the top cause of death in people under 45 years of age. Deaths from severe trauma can have a negative economic impact due to the loss of people belonging to socio-economically active age groups. Therefore, efforts to reduce the mortality rate of trauma patients are essential. The purpose of this study was to investigate preventable mortality in trauma patients and to identify factors and healthcare-related challenges affecting mortality. Ultimately, these findings will help to improve the quality of trauma care. Methods: We analyzed the deaths of 411 severe trauma patients who presented to Gachon University Gil Hospital regional trauma center in South Korea from January 2015 to December 2017, using an expert panel review. Results: The preventable death rate of trauma patients treated at the Gachon University Gil Hospital regional trauma center was 8.0%. Of these, definitely preventable deaths comprised 0.5% and potentially preventable deaths 7.5%. The leading cause of death in trauma patients was traumatic brain injury. Treatment errors most commonly occurred in the intensive care unit (ICU). The most frequent management error was delayed treatment of bleeding. Conclusions: Most errors in the treatment of trauma patients occurred in early stages of the treatment process and in the ICU. By identifying the main causes of preventable death and errors during the course of treatment, our research will help to reduce the preventable death rate. Appropriate trauma care systems and ongoing education are also needed to reduce preventable deaths from trauma.
교육과정 재구성을 통한 문화예술교육 가능성 탐색: “사회로 예술하기” 사례를 중심으로

본 연구는 서울문화재단의 지원사업인 초등학교 3학년 사회와 예술의 통합수업 ‘사회로 예술하기’ 수업을 관찰, 분석하고, 사회과와 예술의 통합 의미와 가능성을 탐색하였다. 총 8차시로 구성되어 있는 프로그램에서 A학교에서 수업을 진행한 TA와 담임 교사의 허락을 얻은 2차시와 7차시를 대상으로 직접 관찰과 촬영한 비디오 분석을 한 결과는 다음과 같다. 교수·학습의 비율(수업몰두도), 인지주의 학습이론의 적용, 수업의 네 가지 요소, 수업의 형식 등 수업의 기초적 분야에서 요구하는 것을 적절히 수행하였는데, 사회와 예술이 통합되면서 그 만큼 교사와 학생이 사용할 수 있는 학습방법과 학습 자료의 선택기회가 더욱 넓어졌기 때문인 것으로 보인다. 활용할 소재와 도구가 많은 만큼 학습자의 참여가 매우 높았으며, 높은 학습동기가 유지되었다. 특히 학습자의 동기를 불러일으키는 많은 사진자료의 활용과 신체의 활용이 주효한 것으로 보였다. 특히 TA는 자신들의 전공을 최대한 살려 전문적인 모습들을 보여줌으로써 높은 수준의 시범으로 학습자의 주의와 신뢰를 얻었다. 그러한 결과는 90%가 넘는 높은 수업몰두도로 나타났다. 수업이 끝나는 단계에서 학생들은 학습정리를 하는데, 글쓰기 외에도 문화 예술적 기능을 활용해서 다양한 방법을 사용해도 되도록 허용했다. 쓰기, 그리기, 붙이기 등 학생 개인마다 자신의 취향대로 독창적인 노트를 만드는데 집중하는 모습을 보였다. 또한 통합예술교육 매트릭스로 분석했을 때 5가지의 행동목표와 7가지의 내용요소에 해당되는 많은 활동들이 실현되었다. 향후 이 매트릭스로 정교한 수업계획을 세워서 실천하면 사회과를 비롯하여 어떤 교과에서든지 더욱 통합예술교육의 효과가 클 것으로 생각된다. The purpose of this case study was to examine the significance of the integrated instruction: Social Studies and Arts and Culture by Curriculum Reconstruction, focused on the Art Plus(Arts-integrated curriculum for children: Social Studies) Project (APP). APP was designed to teach Social Studies using Arts and Culture materials at the third grade social studies class in elementary school by Seoul Foundation for Arts and Culture. The method of this study was class observation using video camera. One elementary social studies class in Seoul, participated in the Art Plus(Arts-integrated curriculum for children: Social Studies) Project, was collected. The second and seventh phase classes of the program were observed and analysed. The findings of the study were as the following: First, there was high task engagement of students over 90% in a class according to Seating Chart Observation Records(SCORE). Lecturers, experts in Arts and Culture, used many Arts and Culture materials and gave a good demonstration to students. These drew students` attention and lead them to engage in the work actively. Second, there were so many various and active group works especially using full body because of using Art and Culture and Social Studies integrated curriculum. It gave so many choices, alternative ideas, materials, activities using in class activities, to lecturers and students. Third, students were interested in making notes of the instruction at the closing time in class by themselves, because they were allowed to write, draw, stick and so on. Therefore they could make various unique note-books as they please.

This study aims to explore some aspects of perception about grammar inquiry and scientific inquiry in educational discussions. In grammar inquiry, generating an explicit hypothesis is not required because the inquiry based on the intuition or experience of language is the main process of the grammar inquiry. On the other hand, a hypothesis in the science inquiry is considered very important in that it provides a clue for resolving the science inquiry's problem. In addition, the key part of the inquiry process in grammar is referred to as “analysis”. Further, it is considered important to understand analytically some of language forms, meanings, and functions in the actual inquiry process. On the other hand, “prediction” and “inference” are regarded as the essential processes of science inquiry, because it is important to plan for the establishment and observation of variables based on predictions and inferences about causes and results in the science inquiry.
