N/A Characteristics of population genetic structure of the Jindo dog breed inhabiting Korean mainland and the offshore islet, Jindo, where the name of the dog originated, were compared using four microsatellite DNA loci. Considerably higher genetic diversity estimates and allele numbers were observed in the Jindo islet population, indicating genetic heterogeneity in this population. This is quite different from what one can expect because currently much less numbers of Jindo dogs are raised in Jindo islet (∼10,000 in Jindo islet vs. ∼100,000 in mainland). We discussed the observed result in terms of non-random breeding in the mainland population and possible release of originally pure dogs to mainland. Although the mainland population showed a significant deviation from Hardy-Weinberg equilibrium, higher homozygosity, and highly positive FIS in two loci, the Jindo islet population revealed these phenomena only in one locus, suggesting that Jindo dogs in mainland propagate more severely through small numbers of dogs. This non-random breeding also appears to be responsible for the differentiated gene pool between the two populations.

ABSTRACT

The brown alga Undaria pinnatifida, which is called Mi-Yoek in Korea, has been traditionally consumed as a health food in East Asian countries. Recent studies have reported that U. pinnatifida has beneficial effects on arteriosclerosis, inflammation, fat metabolism, and tumors. In this study, we examined the anti-senescence effects of ethanol extracts of U. pinnatifida (UP-Ex) in human bone marrow mesenchymal stem cells (hBM-MSCs). UP-Ex protected hBM-MSCs against oxidative injury, as determined by MTT assays. This effect was confirmed by immunoblot analysis of the oxidation-sensitive protein p53 and the apoptotic protein cleaved caspase-3. Excessive intracellular reactive oxygen species (ROS) accumulation induced by oxidative stress was moderated in UP-Ex-treated hBM-MSCs (UP-Ex-MSCs). Similarly, expression of the ROS-scavenging enzymes superoxide dismutase 1 (SOD1), SOD2, and catalase was recovered in UP-Ex-MSCs. Excessive ROS induced by long-term cell expansion (passage 17) was significantly decreased along with restoration of the senescence proteins p53, p21, and p16 in UP-Ex-MSCs. UP-Ex treatment also improved the ability of these replicative, senescent hBM-MSCs (passage 17) to differentiate into osteocytes or adipocytes, suggesting that UP-Ex ameliorates the functional decline of senescent stem cells and may provide better therapeutic efficacy in stem cell therapy.

Abbreviations: hBM-MSCs: human bone marrow mesenchymal stem cells; DCF: 2′,7′-dichlorodihydrofluorescein; DCFH-DA: 2′,7′-dichlorofluorescein diacetate; MTT: 3-(4,5-dimethylthiazol-2-yl-)2,5-diphenyltetrazolium bromide; PBS: phosphate-buffered saline; PFA: paraformaldehyde; RIPA: radioimmunoprecipitation assay; ROS: reactive oxygen species; SOD1: superoxide dismutase 1; SOD2: superoxide dismutase 2.

Five compounds were isolated from the chloroform-soluble fraction of the methanolic extract of the dried rhizomes of Zingiber officinale (Zingiberaceae) through repeated column chromatography. Their chemical structures were elucidated as 4-, 6-, 8-, and 10-gingerols, and 6-shogaol using spectroscopic analysis. Among the five isolated compounds, 6-shogaol exhibited the most potent cytotoxicity against human A549, SK-OV-3, SK-MEL-2, and HCT15 tumor cells. 6-shogaol inhibited proliferation of the transgenic mouse ovarian cancer cell lines, C1 (genotype: $p53^{-/-}$, c-myc, K-ras) and C2 (genotype: $p53^{-/-}$, c-myc, Akt), with $ED_{50}$ values of 0.58 ${\mu}M$ (C1) and 10.7 ${\mu}M$ (C2).

This study was performed to determine the etiological agents in concurrent disorders in gastrointestinal and respiratory tract. Most of dogs had clinical signs including nasal and ocular discharge, coughing, vomiting, and diarrhea. Of the 22 dogs, seropositive rates of each virus were 54.5% (12/22) against canine distemper virus, 90.9% (20/22) against canine adenovirus 1, 36.4% (8/22) against canine adenovirus 2, 18.2% (4/22) against canine parvovirus, 81.8% (18/22) against canine hepatitis virus and 59.1% (13/22) against canine coronavirus. Canine distemper virus and canine parvovirus infection were 54.6% (12/22) in histopathological examination. In addition, mixed infections of canine distemper virus and adenovirus 2 were 9.1% (2/22). While simple infection of canine adenovirus 2 were 9.1% (2/22). E coli and Staphylococcus spp were isolated in facts as a rate of 72.7% (16/22) and 40.9% (9/22), respectively. Conclusionally, it is also estimated that environmental stress might be one of the causative factors.

Suspected infections of Campylobacter spp and Clostridium spp were observed in three dogs. The diagnosis was based on fecal cytology, Gram's stain, clinical signs and serum chemistry. The rectal swabs of diarrheic dogs were performed to confirm the enteropathogens. Suspected Campylobacter spp were a sea-gull shape and Clostridium spp had a large, clear endospore in rectal cytology. Treatment with appropriate antibiotics resulted in a complete resolution of all clinical abnormalities in three cases. The source of Campylobacter spp and Clostridium spp could not be found clearly in three cases, but gastrointestinal origin was most likely. When detecting the enteropathogens in feces, fecal smear with Wright's and Gram's stain should be made at first and also, if the patients have canine parvoviral enteritis, attention should be paid to confirm the Campylobacter spp and Clostridium spp. In addition, since Campylobacter spp and Clostridium spp as normal bacterial flora exists in canine intestines, it is thought that microbiological isolation should be performed to confirm the suspected Campylobacter spp and Clostridium spp as primary enteropathogens in subsequent study.
