The USP21/YY1/SNHG16 axis contributes to tumor proliferation, migration, and invasion of non-small-cell lung cancer

        Pei Xu,Haibo Xiao,Qi Yang,Rui Hu,Lianyong Jiang,Rui Bi,Xueyan Jiang,Lei Wang,Ju Mei,Fangbao Ding,Jianbing Huang 생화학분자생물학회 2020 Experimental and molecular medicine Vol.52 No.-

Deubiquitinases (DUBs) and noncoding RNAs have been the subjects of recent extensive studies regarding their roles in lung cancer, but the mechanisms involved are largely unknown. In our study, we used The Cancer Genome Atlas data set and bioinformatics analyses and identified USP21, a DUB, as a potential contributor to oncogenesis in nonsmall-cell lung cancer (NSCLC). We further demonstrated that USP21 was highly expressed in NSCLCs. We then conducted a series of in vitro and in vivo assays to explore the effect of USP21 on NSCLC progression and the underlying mechanism involved. USP21 promoted NSCLC cell proliferation, migration, and invasion and in vivo tumor growth by stabilizing a well-known oncogene, Yin Yang-1 (YY1), via mediating its deubiquitination. Furthermore, YY1 transcriptionally regulates the expression of SNHG16. Moreover, StarBase bioinformatics analyses predicted that miR4500 targets SNHG16 and USP21. A series of in vitro experiments indicated that SNHG16 increased the expression of USP21 through miR-4500. In summary, the USP21/YY1/SNHG16 axis plays a role in promoting the progression of NSCLC. Therefore, the USP21/YY1/SNHG16/miR-4500 axis may be a potential therapeutic target in NSCLC treatment.

Use of software-defined radio receivers in two-way satellite time and frequency transfers for UTC computation

        Jiang, Zhiheng,Zhang, Victor,Huang, Yi-Jiun,Achkar, Joseph,Piester, Dirk,Lin, Shinn-Yan,Wu, Wenjun,Naumov, Andrey,Yang, Sung-hoon,Nawrocki, Jerzy,Sesia, Ilaria,Schlunegger, Christian,Yang, Zhiqiang,Fu BUREAU INTERNATIONAL DES POIDS ET MESURES 2018 METROLOGIA -BERLIN- Vol.55 No.5

<P>Two-way satellite time and frequency transfer (TWSTFT) is a primary technique for the generation of coordinated universal time (UTC). About 20 timing laboratories around the world continuously operate TWSTFT using satellite time and ranging equipment (SATRE19) modems for remote time and frequency comparisons in this context. The precision of the SATRE TWSTFT as observed today is limited by an apparent daily variation pattern (diurnal) in the TWSTFT results. The observed peak-to-peak variation have been found as high as 2 ns in some cases. Investigations into the origins of the diurnals have so far provided no complete understanding about the cause of the diurnals. One major contributor to the diurnals, however, could be related to properties of the receive part in the modem. In 2014 and 2015, it was demonstrated that bypassing the receive part and the use of software-defined radio (SDR) receivers in TWSTFT ground stations (SDR TWSTFT) instead could considerably reduce both the diurnals and the measurement noise.</P> <P>In 2016, the International Bureau of Weights and Measures (BIPM) and the Consultative Committee for Time and Frequency (CCTF) working group (WG) on TWSTFT launched a pilot study on the application of SDR receivers in the TWSTFT network for UTC computation.</P> <P>The first results of the pilot study were reported to the CCTF WG on TWSTFT annual meeting in May 2017, demonstrating that SDR TWSTFT shows superior performance compared to that of SATRE TWSTFT for practically all links between participating stations. In particular, for continental TWSTFT links, in which the strongest diurnals appear, the use of SDR TWSTFT results in a significant suppression of the diurnals by a factor of between two and three. For the very long inter-continental links, e.g. the Europe-to-USA links where the diurnals are less pronounced, SDR TWSTFT achieved a smaller but still significant gain of 30%. These findings are supported by an evaluation of some of the links with an alternate technique based on GPS signals (GPS IPPP) as reported in this paper.</P> <P>Stimulated by these results, the WG on TWSTFT prepared a recommendation for the 21st CCTF meeting, which proposed the introduction of SDR TWSTFT in UTC generation. With CCTF approval of the recommendation, a roadmap was developed for the implementation of SDR TWSTFT in UTC generation. In accordance with the roadmap, most of the stations that participated in the pilot study have updated the SDR TWSTFT settings to facilitate the use of SDR TWSTFT data in UTC generation. In addition, the BIPM conducted a final evaluation to validate the long-term stability of SDR TWSTFT links, made test runs using the BIPM standard software for the calculation of UTC, now including SDR TWSTFT data, and started to calculate SDR TWSTFT time links as backup from October 2017. The use of SDR TWSTFT in UTC generation will begin in 2018.</P>

Association between expression levels and growth trait-related SNPs located in promoters of the MC4R and MSTN genes in Spinibarbus hollandi

        Yang Yang,Zhaojun Lan,Hu Shu,Huiqiang Zhou,Xiaolu Jiang,Liping Hou,Pinghua Gu 한국유전학회 2018 Genes & Genomics Vol.40 No.11

Melanocortin 4 receptor: (MC4R) and Myostatin (MSTN) are two important growth trait-related genes in animals. In this study, we showed that two SNPs, MC4R-719A>G and MSTN-519C>T, found in the promoters of the MC4R and MSTN genes, respectively, are both associated with growth traits in Spinibarbus hollandi. Furthermore, we observed that there were significant associations between the expression levels of the MC4R and MSTN genes and these two growth trait-related SNPs. The expression level of MC4R gene in brain was lower in GG genotype fish with extremely high growth performance than that in AA genotype fish with extremely low growth performance. Expression level of the MSTN gene in muscle was lower in TT genotype fish with extremely high growth performance than that in CC and CT genotype fish with lower growth performance. The results indicated that these SNPs located in the promoters of MC4R and MSTN are associated with growthrelated traits through modification of gene expression levels. The MSTN and MC4R SNPs may have useful application in effective marker-assisted selection aimed to increase output in S. hollandi.

BK Channel Deficiency in Osteoblasts Reduces Bone Formation via the Wnt/β-Catenin Pathway

        Jiang, Lan,Yang, Qianhong,Gao, Jianjun,Yang, Jiahong,He, Jiaqi,Xin, Hong,Zhang, Xuemei Korean Society for Molecular and Cellular Biology 2021 Molecules and cells Vol.44 No.8

Global knockout of the BK channel has been proven to affect bone formation; however, whether it directly affects osteoblast differentiation and the mechanism are elusive. In the current study, we further investigated the role of BK channels in bone development and explored whether BK channels impacted the differentiation and proliferation of osteoblasts via the canonical Wnt signaling pathway. Our findings demonstrated that knockout of Kcnma1 disrupted the osteogenesis of osteoblasts and inhibited the stabilization of β-catenin. Western blot analysis showed that the protein levels of Axin1 and USP7 increased when Kcnma1 was deficient. Together, this study confirmed that BK ablation decreased bone mass via the Wnt/β-catenin signaling pathway. Our findings also showed that USP7 might have the ability to stabilize the activity of Axin1, which would increase the degradation of β-catenin in osteoblasts.

Sun-induced chlorophyll fluorescence is more strongly related to absorbed light than to photosynthesis at half-hourly resolution in a rice paddy

        Yang, Kaige,Ryu, Youngryel,Dechant, Benjamin,Berry, Joseph A.,Hwang, Yorum,Jiang, Chongya,Kang, Minseok,Kim, Jongmin,Kimm, Hyungsuk,Kornfeld, Ari,Yang, Xi Elsevier 2018 Remote sensing of environment Vol.216 No.-

<P><B>Abstract</B></P> <P>Sun-induced chlorophyll fluorescence (SiF) is increasingly used as a proxy for vegetation canopy photosynthesis. While ground-based, airborne, and satellite observations have demonstrated a strong linear relationship between SiF and gross primary production (GPP) at seasonal scales, their relationships at high temporal resolution across diurnal to seasonal scales remain unclear. In this study, far-red canopy SiF, GPP, and absorbed photosynthetically active radiation (APAR) were continuously monitored using automated spectral systems and an eddy flux tower over an entire growing season in a rice paddy. At half-hourly resolution, strong linear relationships between SiF and GPP (R2 = 0.76) and APAR and GPP (R2 = 0.76) for the whole growing season were observed. We found that relative humidity, diffuse PAR fraction, and growth stage influenced the relationships between SiF and GPP, and APAR and GPP, and incorporating those factors into multiple regression analysis led to improvements up to R2 = 0.83 and R2 = 0.88, respectively. Relationships between LUEp (=GPP/APAR) and LUEf (=SiF/APAR) were inconsistent at half-hourly and weak at daily resolutions (R2 = 0.24). Both at diurnal and seasonal time scales with half-hourly resolution, we found considerably stronger linear relationships between SiF and APAR than between either SiF and GPP or APAR and GPP. Overall, our results indicate that for subdiurnal temporal resolution, canopy SiF in the rice paddy is above all a very good proxy for APAR at diurnal and seasonal time scales and that therefore SiF-based GPP estimation needs to take into account relevant environmental information to model LUEp. These findings can help develop mechanistic links between canopy SiF and GPP across multiple temporal scales.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Observed SiF is a better proxy of APAR than GPP. </LI> <LI> Found considerable environmental effects on the SiF-GPP relationships </LI> <LI> Improved GPP estimation by combining SiF or APAR with environmental variables </LI> <LI> Contribute to canopy SiF-GPP mechanistic links across multiple temporal scales </LI> </UL> </P>

      • Synthesis and Selective Sorption Behavior of Pt(IV) Ion-Imprinted Polymer Particles

Jiang, Yang,Kim, Dukjoon American Chemical Society 2014 INDUSTRIAL & ENGINEERING CHEMISTRY RESEARCH - Vol.53 No.34

<P>Pt(IV) ion-imprinted polymer (Pt(IV)-IIP) was synthesized in the form of porous particles with dimensions of 100–400 μm by the copolymerization of divinylbenzene and styrene in the presence of self-assembled chelates of 4-vinylpyridine and dimethylglyoxime functional monomers along with Pt(IV) ions. The chemical structure of Pt(IV)-IIP was identified by Fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy, and energy dispersive X-ray spectroscopy. The pore size and structure of Pt(IV)-IIP was characterized by Brunauer–Emmett–Teller analysis and scanning electron microscope. The effects of pH and the initial concentration of Pt(IV) on the selective adsorption behavior for the template ion was investigated using flame atomic absorption spectroscopy. Compared to nonimprinted polymer particles, the Pt(IV)-IIPs illustrated excellent selectivity toward the template ion, Pt(IV), over other competitive ions such as Ni(II), Cu(II), and Pd(II). The selectivity coefficients (α) of Pt(IV)/Ni(II), Pt(IV)/Cu(II), Pt(IV)/Zn(II), and Pt(IV)/Pd(II) were 330.27, 359.89, 463.91, and 4.85, respectively. The study on the selective separation of Pt(IV) from the spent catalyst solution revealed the potential application of Pt(IV)-IIP particles in the practical industry.</P>

Multiscale Spatial Position Coding under Locality Constraint for Action Recognition

Yang, Jiang-feng,Ma, Zheng,Xie, Mei The Korean Institute of Electrical Engineers 2015 Journal of Electrical Engineering & Technology Vol.10 No.4

        – In the paper, to handle the problem of traditional bag-of-features model ignoring the spatial relationship of local features in human action recognition, we proposed a Multiscale Spatial Position Coding under Locality Constraint method. Specifically, to describe this spatial relationship, we proposed a mixed feature combining motion feature and multi-spatial-scale configuration. To utilize temporal information between features, sub spatial-temporal-volumes are built. Next, the pooled features of sub-STVs are obtained via max-pooling method. In classification stage, the Locality-Constrained Group Sparse Representation is adopted to utilize the intrinsic group information of the sub-STV features. The experimental results on the KTH, Weizmann, and UCF sports datasets show that our action recognition system outperforms the classical local ST feature-based recognition systems published recently.

      • Who Uses Travel Websites? A Comparison of Demand Across Websites

Yang, Yang,Jiang, Lan Smart Tourism Research Center 2022 Journal of smart tourism Vol.2 No.3

Understanding demand for travel websites can help hoteliers better evaluate their own multi-channel distribution and reputation management strategies. In this study, we estimated demand for eight major travel websites based on the user percentage in 3,120 U.S. counties. Results highlighted the importance of four types of factors: sociodemographic, economic, Internet use-related, and travel-related. Differences between websites were also compared based on estimated coefficients. For example, the demand for Expedia.com appeared to be driven by age, education background, income, and foreign travel history, whereas the demand for Hotels.com was driven by the proportion of the African American population and domestic travel history. Lastly, implications are provided.

Effects of Artemisia argyi flavonoids on growth performance and immune function in broilers challenged with lipopolysaccharide

        Yang, Shuo,Zhang, Jing,Jiang, Yang,Xu, Yuan Qing,Jin, Xiao,Yan, Su Mei,Shi, Bin Lin Asian Australasian Association of Animal Productio 2021 Animal Bioscience Vol.34 No.7

Objective: This research aimed to study the effects of Artemisia argyi flavonoids (AAF) supplemented in diets on the growth performance and immune function of broiler chickens challenged with lipopolysaccharide (LPS). Methods: A total of one hundred and ninety-two 1-d-old broiler chicks were assigned into 4 treatment groups, which were, respectively, fed a basal diet (control), fed a diet with 750 mg/kg AAF, fed a basal diet, and challenged with LPS, fed a diet with 750 mg/kg AAF, and challenged with LPS. Each treatment had six pens with 8 chicks per pen. On days 14, 16, 18, 20 (stress phase I) and 28, 30, 32, 34 (stress phase II), broilers were injected with LPS (500 ㎍/kg body weight) or an equivalent amount of saline. Results: The results demonstrated that dietary AAF significantly improved the body weight (d 21) and alleviated the decrease of average daily gain in broilers challenged with LPS on d 21 and d 35 (p<0.05). Dietary AAF increased bursa fabricius index, and dramatically attenuated the elevation of spleen index caused by LPS on d 35 (p<0.05). Furthermore, serum interleukin-6 (IL-6) concentration decreased with AAF supplementation on d 21 (p<0.05). Diet treatment and LPS challenge exhibited a significant interaction for the concentration of IL-1β (d 21) and IL-6 (d 35) in serum (p<0.05). Additionally, AAF supplementation mitigated the increase of IL-1β, IL-6 in liver and spleen induced by LPS on d 21 and 35 (p<0.05). This study also showed that AAF supplementation significantly reduced the expression of IL-1β (d 21) and nuclear transcription factor kappa-B p65 (d 21 and 35) in liver (p<0.05), and dietary AAF and LPS treatment exhibited significant interaction for the gene expression of IL-6 (d 21), toll like receptor 4 (d 35) and myeloid differentiation factor 88 (d 35) in spleen (p<0.05). Conclusion: In conclusion, AAF could be used as a potential natural immunomodulator to improve growth performance and alleviate immune stress in broilers challenged with LPS.
