For successful clinical tumor immunotherapy outcomes, strong immune responses against tumor antigens must be generated. Cell-based vaccines compromise one strategy with which to induce appropriate strong immune responses. Previously, we established a natural killer T-cell (NKT) ligand-loaded, adenoviral vector-transduced B-cell-based anticancer cellular vaccine. To enhance tumor antigen delivery to B cells, we established a modified adenoviral vector (Ad-k35) that encoded a truncated form of the breast cancer antigen Her2/neu (Ad-k35HM) in which fiber structure was substituted with adenovirus serotype 35. We observed increased tumor antigen expression with Ad-k35HM in both human and murine B cells. In addition, an Ad-k35HM-transduced B-cell vaccine elicited strong antigen-specific cellular and humoral immune responses that were further enhanced with the additional loading of soluble NKT ligand KBC009. An Ad-k35HM-transduced, KBC009-loaded B-cell vaccine efficiently suppressed the in vivo growth of established tumors in a mouse model. Moreover, the vaccine elicited human leukocyte antigen (HLA)-A2 epitope-specific cytotoxic T-cell responses in B6.Cg (CB)-Tg (HLA-A/H2-D) 2Enge/Jat mice. These findings indicated that the Ad-k35 could be appropriate for the preclinical and clinical development of B-cell-based anticancer immunotherapies.

The components of Magnolia obovata are known to have many pharmacological activities. In this study, we investigated the effects of obovatol, a neolignan compound isolated from the leaves of M. obovata, on nitric oxide (NO) production and NF-κB activity in lipopolysaccharide (LPS)-activated RAW 264.7 cells. The results show that obovatol (1-5 μM) significantly inhibited LPS-induced NO production in a concentration-dependent manner (IC<SUB>50</SUB>: 0.91 μM). Consistent with the inhibitory effect on NO production, obovatol inhibits the expression of inducible nitric oxide synthase and cyclooxygenase-2 expression. Furthermore, obovatol suppressed NF-κB (p50 and p65) translocation to the nucleus as well as IκB release resulting in the inhibition of the DNA binding activity of the NF-κB. Obovatol also inhibited c-Jun N-terminal kinase (JNK) and extracellular signal-regulated kinase (ERK) signal, which are the most significantly involved signal in NO production and NF-κB activation. When the cells were treated with the combination of obovatol with U0126 (an ERK inhibitor) or SP600125 (a JNK inhibitor) as well as with SC-514 (an IKK2 inhibitor), much more inhibition of NO production was observed than that by obovatol alone. The present results suggest that obovatol has an inhibitory effect on NO production through the inhibition of NF-κB/MAPK activity, and thus can be used as an anti-inflammatory agent.

We have investigated the effect of oxygen vacancy on the structural and electrical properties of NaNbO<SUB>3</SUB> sub-micron cubes. Through the Rietveld analysis and Raman scattering, we have confirmed that both NaNbO<SUB>3</SUB> and oxygen deficient NaNbO<SUB>3-δ</SUB> have Pmc2<SUB>1</SUB> symmetry, which has no space-inversion. The oxygen vacancy seems to induce the change from the ferroelectricity of NaNbO<SUB>3</SUB> to the piezoelectricity of NaNbO<SUB>3-δ</SUB> and the increase of electric conduction. This work may provide the plausible way to realize sub-micron-size lead-free piezoelectric semiconductors with perovskite structure, which should be quite useful for piezoelectric energy harvesting, switchable diode, and photovoltaic applications.

도축 후 돈육의 육질평가에 있어 중요한 요인인 사후 24시간 pH를 측정하여 pH가 5.31∼5.50, 5.51∼5.70, ≥5.71에 따른 육질을 조사한 결과, 사후 pH가 더 낮을수록 단백질 함량이 높고, 지방 함량이 낮았다(P<0.05). 사후 24시간 pH가 높을수록 보수력은 증가하였으며, 가열감량과 육즙감량은 유의적으로(P<0.05) 감소하였다. 육의 경도, 탄력성, 씹힘성과 지방경도 역시 사후 24시간 pH가 높을수록 유의적인 증가를 보였다(P<0.05). 육색특성 중 CIE L*값은 사후 24시간 pH가 낮을수록 유의적으로 높았으며, a*와 b*값, chroma 값은 최종 pH가 5.71이상의 돈육에서 유의적으로 낮게 나타났다(P<0.05). Hue 값은 pH 5.31∼5.70 범위에서 가장 높았으며, ΔE 값도 pH 5.31∼5.50에서 가장 높았다. 관능적 특성의 분석결과, 다즙성과 연도는 최종 pH가 높을수록 향상되었으나, 향미는 최종 pH 범위에 따른 차이가 인정되지 않았다. 따라서 본 연구결과, 도축 후 24시간 돈육의 pH 범위에 따라 육질의 차이를 보이기 때문에 사후 적정 pH를 유지할 수 있도록 도축전 관리, 도축방법, 도축 후 냉각 온도관리 등을 개선하는 것이 고품질 돈육생산에 있어 중요한 것으로 사료된다. Meat quality of the domestic pork loins(n=537) classified by 3 groups(5.31∼5.50, 5.51∼5.70 and ≥5.71) according to pH at 24hr post-mortem(pH_24) was investigated. In proximate chemical compositions, protein was highest and fat was lowest in the pork loins of pH_24 5.31∼5.50 group. Water holding capacity increased as pH_24 increased, whereas purge loss and cooking loss decreased as pH_24 increased. Meat color values(CIE L*, a*, b*, Chroma, Hue and ΔE) decreased as pH_24 increased. In texture traits, hardness and chewiness were lowest and fat hardness was highest in the pork loins of pH_24≥5.71 group when compared to the other pH_24 groups. However, Warner-Bratzler Shear force, springiness and cohesiveness were not significantly different among the pH_24 groups(P>0.05). In sensory properties, juiciness and tenderness were highest in pH_24≥5.71 group. From the results of this study, pork quality was highly related to pH_24. Therefore, the factors affecting the post-mortem pH, such as stress before slaughter, slaughtering methods, and cooling condition slaughter must be properly controlled and improved to produce high quality pork.

<P>Abstract</P><P>The insect digestive system is the first line of defence protecting cells and tissues of the body from a broad spectrum of toxins and antinutritional factors in its food. To gain insight into the nature and breadth of genes involved in adaptation to dietary challenge, a collection of 20 352 cDNAs was prepared from the midgut tissue of cowpea bruchid larvae (<I>Callosobruchus maculatus</I>) fed on regular diet and diets containing antinutritional compounds. Transcript responses of the larvae to dietary soybean cystatin (scN) were analysed using cDNA microarrays, followed by quantitative real-time PCR (RT-PCR) confirmation with selected genes. The midgut transcript profile of insects fed a sustained sublethal scN dose over the larval life was compared with that of insects treated with an acute high dose of scN for 24 h. A total of 1756 scN-responsive cDNAs was sequenced; these clustered into 967 contigs, of which 653 were singletons. Many contigs (451) did not show homology with known genes, or had homology only with genes of unknown function in a B<SMALL>LAST</SMALL> search. The identified differentially regulated sequences encoded proteins presumptively involved in metabolism, structure, development, signalling, defence and stress response. Expression patterns of some scN-responsive genes were consistent in each larval stage, whereas others exhibited developmental stage-specificity. Acute (24 h), high level exposure to dietary scN caused altered expression of a set of genes partially overlapping with the transcript profile seen under chronic lower level exposure. Protein and carbohydrate hydrolases were generally up-regulated by scN whereas structural, defence and stress-related genes were largely down-regulated. These results show that insects actively mobilize genomic resources in the alimentary tract to mitigate the impact of a digestive protease inhibitor. The enhanced or restored digestibility that may result is possibly crucial for insect survival, yet may be bought at the cost of weakened response to other stresses.</P>

<P>We report the pyroelectric power generation of a relaxor ferroelectric 0.7Pb(Mg1/3Nb2/3)O-3-0.3PbTiO(3) (PMN-PT) single crystal. For a temperature variation rate of 0.5 K/s, the PMN-PT generated a closed-circuit current of 10 nA and an open-circuit voltage of 1.1 V at room temperature. The pyroelectric current and voltage of the PMN-PT single crystal were proportional to the temperature variation rate and were sharply enhanced near the structural phase-transition temperature. Additionally, the PMN-PT pyroelectric generator produced stable power for excessive thermal cycling and was highly sensitive to random thermal fluctuations. <P>The absorption and fluorescence spectroscopic properties of all series of lanthanide elements (La, Ce, (...), Yb, Lu) were investigated in high temperature molten LiCl-KCl eutectic. Based on absorption spectra, the molar absorptivities for major absorption bands of lanthanides (Ln) were determined by analyzing the calibration curves at various concentrations. Moreover, the nephelauxetic parameters, beta, for lanthanides in molten LiCl-KCl salt were evaluated by comparing those for aqueous lanthanide ions. In addition, fluorescence spectra and lifetimes of lanthanide elements were obtained at various Ln(III) concentrations, and this is the first to systematically present fluorescence spectroscopic properties of the whole lanthanide elements in high temperature molten LiCl-KCl salt. Such absorption and fluorescence spectra can provide a fundamental basis for the understanding and development of various pyrochemical processes in molten LiCl-KCl eutectic. (C) 2016 Elsevier B.V.
