Background: Myocardial ischemia and reperfusion injury (MI/RI) is a complex pathophysiological process, which can lead to severe myocardial injury. The long noncoding RNA alpha-2-macroglobulin antisense RNA 1 (A2M-AS1) has been revealed to be abnormally expressed in MI, However, its function in MI and the potential mechanism are still unclear. Objective: To evaluate the functional role of A2M-AS1 in hypoxia/reoxygenation (H/R)-induced neonatal cardiomyocytes and its potential molecular mechanism. Methods: Dataset GSE66360 was obtained from GEO database for analyzing the RNA expression of A2M-AS1 and interleukin 1 receptor type 2 (IL1R2). KEGG pathway enrichment analysis of the genes that co-expressed with A2M-AS1 was performed. Human neonatal cardiomyocytes were subjected to H/R to construct in vitro models. QRT-PCR and Western blot were adopted to test the levels of mRNA and protein. The viability and apoptosis of cardiomyocytes were tested by CCK-8 and flow cytometry assays, respectively. Results: The expression of A2M-AS1 was notably downregulated in H/R-treated cardiomyocytes. Overexpression of A2M-AS1 can notably enhance the cell viability of H/R-damaged cardiomyocytes, whereas knockdown of A2M-AS1 showed the opposite outcomes. Besides, a negative correlation was showed between A2M-AS1 and IL1R2 expression. In H/R-treated cardiomyocytes, overexpression of IL1R2 weakened the promoting proliferation and anti-apoptosis effects caused by overexpressing A2M-AS1, however, IL1R2-knockdown abolished the anti-proliferation and pro-apoptosis effects caused by silencing A2M-AS1. Conclusion: This study demonstrates the potential regulatory role of A2M-AS1/ IL1R2 axis in cardiomyocytes suffered from H/R, and provides insight into the protection of MI/RI.
Anomalous Dependence of Population Growth on the Birth Rate in the Plant-herbivore System

We performed a simulation of the two-species plant-herbivore system by using the agent-based NetLogo program and constructed a dynamic model of populations consistent with the simulation results. The dynamic model is a three-dimensional system including the mean energy of the herbivore in addition to two variables denoting the populations of plants and herbivores. A steady-state analysis of the dynamic model shows that the dependence of the herbivore population on the birth and the death rates observed from the agent model is consistent with the prediction of the dynamic model. Especially, the anomalous dependence of the herbivore population on the birth rate, where the population decreases with the birth rate for small death rate, is consistently explained by a phase plane analysis of the dynamic model.
A re-examination of the current design rule for staggered bolted connections

This paper summarised and re-examined the theoretical basis of the commonly used design rule developed by Cochrane in the 1920s to consider staggered bolt holes in tension members, i.e., the s 2 /4g rule. The rule was derived assuming that the term two times the bolt hole diameter (2d0) in Cochrane’s original equation could be neglected, and assuming a value of 0.5 for the fractional deduction of a staggered hole in assessing the net section area. Although the s 2 /4g rule generally provides good predictions of the staggered net section area, the above-mentioned assumptions used in developing the rule are doubtful, in particular for a connection with a small gauge-to-bolt-hole diameter (g/d0) ratio. It was found that the omission of 2d0 in Cochrane’s original equation appreciably overestimates the net section area of a staggered bolted connection with a small g/d0 ratio. However, the assumed value of 0.5 for the fractional deduction of a staggered hole underestimates the staggered net section area for small g/d0 ratios. To improve the applicability of the above two assumptions, a modified design equation, which covers a full range of g/d0 ratio, was proposed to accurately predict the staggered net section area and was validated by the existing test data from the literature and numerical data derived from this study. Finally, a reliability analysis of the test and numerical data was conducted, and the results showed that the reliability of the modified design equation for evaluating the net section resistance of staggered bolted connections can be achieved with the partial factor of 1.25.
Stimulation of keratinocyte differentiation by extract of combined medicinal plant

Skin is the outermost organ and acts as a barrier between the organism and environment. Skin protects the organism from environmental insults, such as chemicals, pathogens, and UV light. Much of the protective function of skin is dependent on the epidermis, a multi-layered epithelium that is composed of various cell types such as keratinocytes and melanocytes. Keratinocytes produce protective components through a sophisticated differentiation process. Disturbance of keratinocyte differentiation is related to several skin diseases such as psoriasis and atopic dermatitis. In this study, we prepared extract of combined medicinal plants (ECMP) consisting of Taraxacum platycarpum H. Dahlstedt, Heartleaf Houttuynia, Glycyrrhiza uralensis Fischer, and root bark of Ulmus davidiana. We demonstrated that ECMP enhanced keratinocyte differentiation and barrier functionality using an in vitro cell culture system and in vivo animal test. Treatment of cultured keratinocytes with ECMP resulted in induction of keratinocyte differentiation, as evidenced by increased differentiation markers such as involucrin, loricrin, and filaggrin. In line with these results, ECMP decreased proliferation of keratinocytes cultured in vitro. ECMP applied topically to tape-stripped mouse skins accelerated reduction of transepidermal water loss (TEWL), indicating fast recovery of barrier function. Immunohistochemistry showed that ECMP increased the filaggrin level in tape-stripped mouse skins. These results suggest that ECMP may be applicable for keratinocyte differentiation-related skin diseases.
