        Recent Developments in High-performance Liquid Chromatography of Lipids

        The possibilities for HPLC analysis of lipids have been revolutionised by the availability of evaporative light-scattering detectors, with which the response is independent of the nature of the mobile phase and does not depend On the presence of specific chromophores in the lipids. It was thus possible to develop an HPLC procedure, involving ternary gradient elution, for separating all the lipid classes in animal tissues in a single step. Although reversed-phase HPLC has been widely used for the analysis of molecular species of lipids, sliver ion chromatography can be a valuable alternative. For example, a stable silver ion column for HPLC was developed which permitted resolution of molecular species of triacylglycerols, even from such complex samples as fish oils, again With light-scattering detection and gradient elution. The capacity for HPLC resolution of diastereomeric diacyl-sn-glycerol derivatives, prepared from triacylglycerols. has lead to a new simple method for stereospecific analysis of the latter.

        UAH Version 6 Global Satellite Temperature Products: Methodology and Results

        Version 6 of the UAH MSU/AMSU global satellite temperature dataset represents an extensive revision of the procedures employed in previous versions of the UAH datasets. The two most significant results from an end-user perspective are (1) a decrease in the global-average lower tropospheric temperature (LT) trend from +0.14oC decade−1 to +0.11oC decade−1 (Jan. 1979 through Dec. 2015); and (2) the geographic distribution of the LT trends, including higher spatial resolution, owing to a new method for computing LT. We describe the major changes in processing strategy, including a new method for monthly gridpoint averaging which uses all of the footprint data yet eliminates the need for limb correction; a new multi-channel (rather than multi-angle) method for computing the lower tropospheric (LT) temperature product which requires an additional tropopause (TP) channel to be used; and a new empirical method for diurnal drift correction. We show results for LT, the midtroposphere (MT, from MSU2/AMSU5), and lower stratosphere (LS, from MSU4/AMSU9). A 0.03oC decade−1 reduction in the global LT trend from the Version 5.6 product is partly due to lesser sensitivity of the new LT to land surface skin temperature (est. 0.01oC decade−1), with the remainder of the reduction (0.02oC decade−1) due to the new diurnal drift adjustment, the more robust method of LT calculation, and other changes in processing procedures.

      • Novel Di-, Tri- and Teraenoic Fatty Acids with bis-Methylene-Interrupted Double-Bond Systems from the Sponge Haliclona cinerea

        Unusual fatty acids, with up to 34 carbon atoms and containing one or two bis-methylene-interrupted double-bond systems, have been identified in the sponge Haliclona cinerea from the Black Sea. These include the dienes-5, 9-16:2, 7, 11-18:1, 9, 13-20:2, 13, 17-24:2, 15, 19-26:2, 17, 21-28:2, 19, 23-30:2 and 21, 25-32:2; trienes- 5, 9, 23-30:3, 5, 9, 24-31:3, 5, 9, 23, 27-34:4. In addition, 5, 9, 13-eicosatrienoic acid was present. Many of these do not appear to have been described before, and only 5, 9-16:2 and 5, 9, 23-30:3 are found often in sponges. They were identified by using silver-ion high-performance liquid chromatography to simplify the complex mixture of fatty acids of subsequent analysis by gas chromatography-mass spectrometry as pico-linyl ester derivatives. Deuteration with Wilkinson's catalyst in homogeneous solution confirmed the structures. We speculate that the di- and tetraenoic fatty acids arise by chain elongation of 5, 9-hexadecadienoic acid, also a major component of the lipids, followed by further insertion of double bonds in the 5 and 9 positions. The trienes may be formed from 9-hexadecenoic acid by similar mechanisms.
