      • Scalable Solvothermal Synthesis of Superparamagnetic Fe<sub>3</sub>O<sub>4</sub> Nanoclusters for Bioseparation and Theragnostic Probes

        <P>Magnetic nanoparticles have had a significant impact on a wide range of advanced applications in the academic and industrial fields. In particular, in nanomedicine, the nanoparticles require specific properties, including hydrophilic behavior, uniform and tunable dimensions, and good magnetic properties, which are still challenging to achieve by industrial-scale synthesis. Here, we report a gram-scale synthesis of hydrophilic magnetic nanoclusters based on a one-pot solvothermal system. Using this approach, we achieved the nanoclusters with controlled size composed of magnetite nanocrystals in close-packed superstructures that exhibited hydrophilicity, superparamagnetism, high magnetization, and colloidal stability. The proposed solvothermal method is found to be highly suitable for synthesizing industrial quantities (gram-per-batch level) of magnetic spheres with unchanged structural and magnetic properties. Furthermore, coating the magnetic spheres with an additional silica layer provided further stability and specific functionalities favorable for biological applications. Using in vitro and in vivo studies, we successfully demonstrated both positive and negative separation and the use of the magnetic nanoclusters as a theragnostic nanoprobe. This scalable synthetic procedure is expected to be highly suitable for widespread use in biomedical, energy storage, photonics, and catalysis fields, among others.</P> [FIG OMISSION]</BR>

        골든 타임을 고려한 신속한 혈액 공급사슬 설계

        각 지역의 혈액원이 인근에 위치한 병원에 혈액을 공급하는 현재의 혈액 공급 체계에서는 긴급하게 혈액이 필요한 응급 상황 발생 시, 필요한 혈액을 신속하게 배송하기 어렵다. 특히 혈액원에서 한 시간 이상의 거리에 위치한 병원들은 수혈이 늦어지는 경우 환자의 치료 결과에 부정적인 영향을 미칠 수 있다. 본 연구에서는 기 존 병원 중 일부를 혈액 분배 병원으로 지정하고, 주변에 위치한 병원들에게 빠르게 혈액을 배송할 수 있는 새로운 혈액 공급 사슬 체계를 제안한다. 기존 병원들 중 혈액 분배 병원을 선정하고, 혈액 분배 병원에 인근 병 원을 할당하기 위해 Minimax 최적화 모델을 개발하였다. 경상북도에 위치한 병원 중 응급 의료기관으로 지정 된 병원들을 대상으로 실제 데이터를 수집하여 수치실험을 진행하였다. 응급상황에 대응하기 위해서는 최소 5 개 이상의 병원들이 혈액 분배 병원으로 지정되어야 함을 알 수 있었으며, 민감도 분석을 통하여 의사결정의 기준을 제공하였다. Under emergency circumstances, a hospital requires a large amount of blood and a regional blood services center is not able to supply the blood to hospitals urgently. In particular, hospitals taking more than one hour from the regional blood services center cannot give a transfusion into patients, which will result in critical impact on the treatment. This paper aims to supply blood quickly to hospitals by developing a new agile blood supply chain system. We develop a minimax mathematical model for assigning regional hospitals to blood distribution hospitals and grouping hospitals based on the assigned blood distribution hospitals. Numerical experiments are conducted with the data about hospitals in Gyeongsangbuk-do, Korea. An in-depth analysis is carried out by changing the number of blood distribution hospitals and comparing the minimax model with a minisum model. Our results show that the number of the blood distribution hospitals should be greater than five and the minimax model is suitable for emergency response.

        최근 전자상거래의 활성화와 더불어 기존의 유선인터넷에서 사용하고 있는 전자지갑 (S/W)등 전자상거래 방식에서 Smart Cαrd 나 이동통신 단말기와 같은 독립적인 유/ 무선 단일을 사용하는 전자지불 시스템에 대한 관심이 고조되고 있다. 본 연구에서는 전자지불 시스템에 Bluetooth를 이용하여 안전한 사용자 인증 및 종단간의 보안을 제공 전자화폐의 발행/지불이 가능한 전자지불 시스템의 모델을 제시하고자 한다. 각 단일의 서브셋(subset)으로 Bluetooth 컨트롤 시스템과 이를 이용하는 전자지불 시스템으로서 전자지갑 및 전자화폐 발행기/자동판매기를 구현하여 각각의 보안 통신과 전자화폐의 발행/지불이 가능함을 나타냈다. 구현 결과물은 실제 전자지불 시스템에 적용이 가능할 것으로 보인다.

        Design and Fabrication of 1.25-Gb/s Full-Triplex Transceiver Module With PLC on a Lossy Si Substrate for G/E-PONs

        <P> 1.25 Gb/s optoelectronic full-triplex transceiver module with planar-lightwave-circuits was designed and fabricated for the fiber-to-the-home services according to G/E-PONs standards. A low electrical crosstalk of the critical characteristics for the reliable operation of the 1.25 Gb/s full-triplex transceiver module is intensively investigated because the electrical crosstalk on a resistive silicon substrate is more serious than that on a dielectric substrate. It is observed that the performances of the transmitter and receiver satisfy the transmitter and receiver specifications defined in the standards. From this proposed module layout, a design convenience as well as a great reduction of the silicon substrate size by about 50 % was completely achieved. Consequently, the 1.25 Gb/s full-triplex transceiver module was fabricated with electrical and mechanical packaging technologies such of a low crosstalk design and a passive alignment method. </P>

      • Numerical analysis of convective drying of a moving moist object

        <P><B>Abstract</B></P> <P>Convective drying of a moving moist object is investigated numerically solving the conservation equations of mass, momentum, energy and air moisture concentration based on a <I>k</I>– ∊ turbulence model in the external region of the object as well as the conservation equations of energy and object moisture concentration in the internal region. The external and internal conservation equations are coupled to the heat and mass transfer conditions on the object surface including the effects of evaporation and water activity. The numerical results show that the analogy between heat and mass transfer for the convective drying is valid only when the object temperature is close to the equilibrium temperature. The simple analytical formulations developed for the total drying rate are found to compare well with the numerical results. The effects of object velocity and thickness and the wall heat flux on the drying rate are quantified.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Complete numerical analysis is conducted for convective drying of a moving moist object. </LI> <LI> Analytical formulations are presented for evaluation of the total drying rate. </LI> <LI> The object flow rate is an important parameter to determine the drying rate. </LI> <LI> The optimum condition of object flow rate increases with the wall heat flux. </LI> </UL> </P>

        Validation of fetus aneuploidy in 221 Korean clinical samples using noninvasive chromosome examination: Clinical laboratory improvement amendments-certified noninvasive prenatal test

        Purpose: We developed and validated a fetal trisomy detection method for use as a noninvasive prenatal test (NIPT) including a Clinical Laboratory Improvement Amendments (CLIA)-certified bioinformatics pipeline on a cloud-based computing system using both Illumina and Life Technology sequencing platforms for 221 Korean clinical samples. We determined the necessary proportions of the fetal fraction in the cell-free DNA (cfDNA) sample for NIPT of trisomies 13, 18, and 21 through a limit of quantification (LOQ) test. Materials and Methods: Next-generation sequencing libraries from 221 clinical samples and three positive controls were generated using Illumina and Life Technology chemistries. Sequencing results were uploaded to a cloud and mapped on the human reference genome (GRCh37/hg19) using bioinformatics tools. Based on Z-scores calculated by normalization of the mapped read counts, final aneuploidy reports were automatically generated for fetal aneuploidy determination. Results: We identified in total 29 aneuploid samples, and additional analytical methods performed to confirm the results showed that one of these was a false-positive. The LOQ test showed that the proportion of fetal fraction in the cfDNA sample would affect the interpretation of the aneuploidy results. Conclusion: Noninvasive chromosome examination (NICE), a CLIA-certified NIPT with a cloud-based bioinformatics platform, showed unambiguous success in fetus aneuploidy detection.

        Identification of a Transferrin Receptor-binding Peptide from a Phage-displayed Peptide Library

        펩타이드 문고 기술을 이용하여 흑색종 세포주인 B16F10에 결합하는 펩타이드 리간드를 검색하였다. 먼저 세포 내부로 들어간 파지들을 선택하는 방법으로 두 번 검색한 후 표면에 결합한 파지들 가운데 트랜스페린 단백질을 이용하여 트랜스페린 수용체에 결합한 파지들만을 선별적으로 용출시키는 방법으로 세 번 검색하였다. 다음으로 이 두 가지 방법을 통해 선별된 파지들에 표현된 펩타이드들을 Pseudomonas exotoxin의 전이 영역과 촉매 영역에 융합시킨 재조합 독소들을 만들었다. B16F10 세포에 대한 각 재조합 독소의 활성을 측정하여 일곱 개의 클론을 선택한 후 염기서열을 분석하였다. 그 결과 그 가운데 한 클론에서 표현하는 펩타이드의 아미노산 서열이 사람의 트랜스페린과 유사한 서열을 갖는 것으로 확인되었다. 그 펩타이드를 화학적으로 합성한 후 항암제를 포함하는 리포솜에 붙여 실험한 결과 트랜스페린 수용체를 통해 치료물질을 전달할 수 있는 가능성을 지닌 것으로 평가되었다. Using a phage peptide library approach, we have isolated a peptide ligand that binds to transferrin receptor on the surface of human melanoma cell, B16F10. The library was first screened twice by recovering internalized phages and was further screened three times by competitively eluting transferrin receptor-specific phages with human transferrin among the phages bound to the cell surface. The peptides displayed by the selected phages were fused to translocation and catalytic domain of Pseudomonas exotoxin to prepare recombinant toxins. After estimating cytotoxicity of each recombinant toxin toward B16F10 cell, seven clones were selected. Sequence analysis revealed that one of the clones displayed a peptide which had a significant sequence homology with human transferrin. The peptide was chemically synthesized and was shown to be functional in delivering cytotoxic agents into B16F10 cell via interaction with transferrin receptor.
