<P>A chiral, fluoride-catalyzed asymmetric cascade sulfaMichael/aldol condensation reaction of 1,4-dithiane-2,5-diol and a series of alpha,beta-unsaturated ketones is described to access chiral trisubstituted tetrahydrothiophene derivatives. The target products, including the spiro tetrahydrothiophene derivatives bearing a five-, six-, and seven-membered ring, were highly functionalized and showed high ee value. This established protocol realized a highly enantioselective reaction with a catalytic amount of KF and Song's chiral oligoEG via in situ generated chiral fluoride to construct useful heterocyclic skeletons with great complexity.</P>

<P>Structural motifs containing contiguous halide bearing stereocenters are common in natural products as well as bioactive molecules. A few successful examples have been reported in the area of asymmetric vicinal dihalogenation of alkenes for accessing dihalogenated products; in this report, an alternative generation method of contiguous halide-bearing stereocenters alpha,beta,gamma,delta relative to carbonyl group in excellent enantioselectivity is proposed by utilizing a Song's oligoEG catalyst-catalyzed asymmetric beta-elimination. According to this methodology, a wide range of anti-syn-trihalides and anti-syn-anti-tetrahalides with high levels of enantioselectivity were synthesized. The synthetic utility of the contiguous halide-bearing stereocenters was demonstrated by several transformations. The results of high-resolution mass spectrometry indicated that the favorable interaction between catalyst and one of the enantiomers of racemic contiguously multihalogenated ketone contributed to the original enantioselectivity of dehydrohalogenation. A deuterium kinetic isotope effect experiment revealed that this beta-elimination reaction proceeds by the E2 mechanism. This strategy opens a new pathway for the asymmetric synthesis of contiguous halide bearing stereocenters of great complexity.</P>
Improved Insecticidal Activities of Novel Bacillus thuringiensis cry1-type proteins

Proteinaceous insecticidal proteins, Cry proteins, from Bacillus thuringiensis (Bt) are insecticidal proteins that are highly active against several species of Lepidoptera. Thus, cry genes encoding these Cry proteins have been widely applied for construction of transgenic crops resistant to pest insects. In this study, through the 3D structure prediction and accompanying mutagenesis study for the Mod-Cry1Ac, 7 and 16 amino acid residues from domain I and II, respectively, responsible for its insecticidal activity against larvae of Spodoptera exigua and Ostrinia furnacalis were identified. We used site-directed mutagenesis to improve the insecticidal activity of Mod-Cry1Ac, resulted 31 mutant cry genes. These mutant cry genes encodes potent insecticidal proteins in the form of crystalline protoxins of 95 kDa. SDS-PAGE analysis of the recombinant polyhedra revealed that expressed Cry proteins was occluded into polyhedra and activated stably to 65 kDa by trypsin. When the insecticidal activities of these mutant Cry proteins against to larvae of P. xylostella, S. exigua and O. furnacalis were assayed, they showed higher or similar insecticidal activity compared to those of Cry1Ac and Cry1C. Especially, Mutant-N16 is considered to have the potential for the efficacious biological insecticide since it showed the highest insecticidal activity.
Construction of Potent Insecticide Leads, Novel Bacillus thuringiensis cry1-type Genes

Crystals of proteinaceous insecticidal proteins, Cry proteins, produced by Bacillus thuringiensis (Bt) have been generally used to control insect pests. In this study, through the 3D structure prediction and accompanying mutagenesis study for the Mod-Cry1Ac, 7 and 16 amino acid residues from domain I and II, respectively, responsible for its insecticidal activity against larvae of Spodoptera exigua and Ostrinia furnacalis were identified. To construct novel cry genes with enhanced insecticidal activity, we randomly mutated these 23 amino acid sequences by in vitro muti site-directed mutagenesis, resulting in totally 24 mutant cry genes. For further characterization, these mutant cry genes were expressed as a fusion protein with polyhedrin using baculovirus expression system. SDS-PAGE analysis of the recombinant polyhedra revealed that expressed Cry proteins was occluded into polyhedra and activated stably to 65 kDa by trypsin. When the insecticidal activities of these mutant Cry proteins against to larvae of P. xylostella and S. exigua were assayed, they showed higher or similar insecticidal activity compared to those of Cry1Ac and Cry1C. Especially, among them Mutant-N16 showed the highest insecticidal activity against to both of P. xylostella and S. exigua. Therefore, Mutant-N16 is considered to have the potential for the efficacious biological insecticide.
Complete mesocolic excision in right hemicolectomy

Purpose: To demonstrate the feasibility, safety, and technical strategies of hand-assisted laparoscopic complete mesocolic excision (HAL-CME) and to compare oncological outcomes between HAL-CME and the open approach (O-CME) for right colon cancers. Methods: Patients who were scheduled to undergo a right hemicolectomy were divided into HAL-CME and O-CME groups. Measured outcomes included demographic variables, perioperative parameters, and follow-up data. Demographic variables included age, sex distribution, body mass index (BMI), American Society of Anesthesiologists (ASA) physical status classification, previous abdominal surgery, tumor localization, and potential comorbidities. Perioperative parameters included incision length, operative time, blood loss, conversion rate, postoperative pain score, postoperative first passage of flatus, duration of hospital stay, total cost, number of lymph nodes retrieved, TNM classification, and postoperative complications. Follow-up data included follow-up time, use of chemotherapy, local recurrence rate, distant metastasis rate, and short-term survival rate. Results: In total, 150 patients (HAL-CME, 78; O-CME, 72) were included. The groups were similar in age, sex distribution, BMI, ASA classification, history of previous abdominal surgeries, tumor localization, and potential comorbidities. Patients in the HAL-CME group had shorter incision lengths, longer operative times, less operative blood loss, lower pain scores, earlier first passage of flatus, shorter hospital stay, higher total costs, similar numbers of lymph nodes retrieved, similar TNM classifications, and a comparable incidence of postoperative complications. The 2 groups were also similar in local recurrence rate, distant metastasis rate, and short-term survival rate. Conclusion: The results demonstrate that the HAL-CME procedure is a safe, valid, and feasible surgical method for right hemicolon cancers.

<P><B>Abstract</B></P> <P>Biochar effect on the potential mobility of dibutyl phthalate (DBP), cadmium (Cd), and lead (Pb) in co-contaminated soils is not well investigated. A laboratory leaching study was conducted to evaluate the effect of biochars derived from bamboo (BB) and pig (PB) on the leachability of DBP, Cd, and Pb through soil columns packed with two soils with low or high organic carbon content (LOC; 0.35% C: HOC; 2.24% C) and spiked with DBP, Cd, and Pb. Application of PB to the LOC soil significantly (<I>P</I> < 0.05) reduced the leaching loss by up to 88% for DBP, 38% for Cd, and 71% for Pb, whereas its impact was insignificant in the HOC soil. The higher efficacy of PB in reducing the leaching of DBP, Cd, and Pb in the LOC soil than that of BB might be related to PB's higher specific surface area, surface alkalinity, pH, and mineral contents compared to those of BB. Co-contamination of Cd and Pb enhanced leaching of DBP in the LOC soil treated with PB, possibly by competition for the sorption sites. Leaching of DBP, Cd, and Pb were significantly (<I>P</I> < 0.05) higher in the LOC soil than in the HOC soil. This study revealed that the effectiveness of biochars was dependent on the soil organic carbon content. Application of PB to the LOC soil was effective in reducing the leaching risk of DBP, Cd, and Pb.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Pig biochar reduced leaching of DBP, Cd, and Pb in the low organic carbon (LOC) soil. </LI> <LI> Contaminant leaching was higher in the LOC soil than in the high organic carbon soil. </LI> <LI> Existence of Cd and Pb enhanced mobility of DBP in the pig biochar-treated LOC soil. </LI> <LI> Alkalinity and phosphate in biochar controlled the leaching loss of Cd and Pb. </LI> <LI> Impact of pig biochar on leaching of DBP, Cd, and Pb is stronger than bamboo biochar. </LI> </UL> </P>
A study on the effect of subsurface crack propagation on rolling contact fatigue in a bearing ring

To study the subsurface fatigue crack propagation under rolling contact fatigue (RCF), a subsurface fatigue crack is firstly observed ina ball bearing race. Then, a three-dimensional model of a bearing ring containing a subsurface crack is used to evaluate fatigue crackpropagation based on stress intensity factor (SIF) calculations. Several parameters, such as the crack shape, depth and size, are varied toinvestigate their effects on RCF in the bearing ring.

Polymorphisms in miRNA binding sites have been shown to affect miRNA binding to target genes, resulting in differential mRNA and protein expression and susceptibility to common diseases. Our purpose was to predict SNPs (single nucleotide polymorphisms) within miRNA binding sites of inflammatory genes in relation to gastric cancer. A complete list of SNPs in the 3'UTR regions of all inflammatory genes associated with gastric cancer was obtained from Pubmed. miRNA target prediction databases (MirSNP, Targetscan Human 6.2, PolymiRTS 3.0, miRNASNP 2.0, and Patrocles) were used to predict miRNA target sites. There were 99 SNPs with MAF>0.05 within the miRNA binding sites of 41 genes among 72 inflammation-related genes associated with gastric cancer. NF-${\kappa}B$ and JAK-STAT are the two most important signaling pathways. 47 SNPs of 25 genes with 95 miRNAs were predicted. CCL2 and IL1F5 were found to be the shared target genes of hsa-miRNA-624-3p. Bioinformatic methods could identify a set of SNPs within miRNA binding sites of inflammatory genes, and provide data and direction for subsequent functional verification research.
