        동창의 임상 및 병리조직학적 소견과 Ki-67 발현 양상

        N/A Background : Pernio or chilblain is an abnormal reaction to damp cold air, resulting in pruritic erythematous swellings on acral skin. The histologic features are subepidermal edema, diffuse fluffy edema of endothelial cell, and superficial or superficial and deep perivascular lymphocytic infiltrates. Many studies with a large sample size have been conducted in western countries, but there were no collective studies of pernio that have been reported in Korea. Objective : The purpose of this study was to clarify the clinical and histopathologic features of pernio in Korea. Methods : We reviewed the clinical and histopathologic findings in ten patients with pernio who visited the Department of Dermatology, Kosin Medical College Gaspel Hospital during the 10 year period from 1988 to 1997. Results : The results were as follows. 1. The age distribution was in the range of 6∼50 year old. The prevalance was heighest in the third and fourth decade. Sex ratio was 1 : 4 (2 males : 8 females). The duration of symptoms ranged from 4 days to 30 years. All patients developed skin lesions on the acral area. Two male patients developed skin lesions on the hand associated with cold exposure and outdoor works. All patient had itching and burning papules and/or plaques, and one had a painful hard nodule. Laboratory studies were all negative and within normal ranges. 2. Histopathologic features revealed subepidermal edema, diffuse fluffy edema of endothelial cell, and superficial or superficial and deep perivascular lymphocytic infiltrates in all specimens. 3. Treatment includes prophylatic measures against cold and nifedipine(calcium channel inhibiting drug). In more severe cases, pentoxifylline might be helpful. Conclusion : Our study suggests that pernio commonly occurs in twenty to forty year old females during fall and winter. Upon exposure to cold temperatures, the acral skin develops pruritic erythematous papules and plaques. If these lesions persisit for a long time or recur, they may progress to vesicle and ulceration and possibly develop vasculitis which does not respond to therapy well. Thus, a thorough understanding of clinical and histopathologic features of pernio is vital in making a diagnosis, a treatment plan and determining the prognosis.       • 음식물찌꺼기 퇴비로부터 호염성 미생물의 분리 및 동정

        An halophilic bacterium was isolated from food-waste. The isolated strain was Gram-negative, aerobic bacterium. The range of temperature for growth of the isolate were 30-40℃. NaCl concentration for growth of it was 5-15%. The isolate was catalase and oxidase positive and sensitive to bacterium. It showed starch hydrolyzing and acid forming characteristics. The morphological, physiological and biochemical characteristics of the isolate resembled those of the Halococcus sp., therefore it was identified as Haloarcula sp. CH-667.

        대구지역 야생조류로부터 분리한 Salmonella속 균의 생물화학적 특성과 Plasmid Profile

        The present study was conducted to investigate the prevalence, biochemical properties and plasmid profiles of Salmonella organisms isolated from wild birds in Daegu area during the period from May, 1999 to August, 2000. Twenty strains of Salmonella were identified from 192 samples of heron feces and 86 samples of pigeon feces. Among Salmonella isolates, S. montevideo were the most frequent serotype (16 strains), 3 strains of them were S. typhimurium and the other one was untypable. The biochemical properties of 20 strains of Salmonella isolated were in accordance with the traditional characteristics of Salmonella spp. in the majority of biochemical test such as IMViC, carbohydrate fermentation and amino acid decarboxylation. The majority of isolates were highly susceptible to amikacin, ampicillin, amoxicillin, carbenicillin, cephalothin, ciprofloxacine, chloramphenicol, enrofloxacin, furazolidone, gentamicin, kanamycin, nalidixic acid, polymixin B and sulfamethoxazole, while they were moderately susceptible to streptomycin and tetracyclin. However, the majority of them were highly resistant to nitrofurantoin. In plasmid profile, 21.1 kilobase (Kb), 20 Kb and 13.2 Kb fractions were common to three S. tvphimurium and one untypable strains, and no fraction was shown in all strains of S. montevideo.

        효소에 의한 꿩고기 가수분해물의 제조

        This studies was carried out to investigate the processing possibility of pheasant meat extracts treated with proteases. The crude protein, aminonitrogen, degree of hydrolysis, yield and amino acid composition of pheasant meat extracts when it was treated with proteases at various temperature and reaction time were analyzed. The crude protein contents of pheasant meat extracts processed in 130℃ were more than when it was done in 100℃, but the contents of aminonitrogen were not quite different between two processing temperature. The content of crude protein and aminonitrogen when pheasant meat was hydrolyzed with protease NP and prozyme A. The yields of pheasant meat extracts, when pheasant meat were treated at 100℃ and 130℃, were from 2.24 to 7.10% and from 5.51 to 10.45%, respectively. And the yield of extraction depended on extraction temperature, kinds of enzyme, amount of enzyme, extraction time. The content of aminonitrogen in pheasant meat extracts treated with enzyme was much higher than any other treatments. And it depended on amount of enzyme, extraction time and temperature. The amount of the amino acids in pheasant meat extracts treated by protease NP were eminently higher than by heat at 100℃ or 130℃.

      • Inhibition of protein kinase CKII activity by euchrestaflavanone B purified from Cudrania tricuspidata.

        <P>The CKII (EC 2.7.1.37) inhibitory compound was purified from the root barks of Cudrania tricuspidata and identified as (2S)-2-[2,4-dihydroxy-5-(3-methyl-but-2-enyl)-phenyl]-5,7-dihyroxy-6-(3-methyl-but-2-enyl)chroman-4-one (euchrestaflavanone B). Euchrestaflavanone B was shown to inhibit the phosphotransferase activity of CKII with IC50 of about 78 microM. Steady-state studies revealed that euchrestaflavanone B acted as a competitive inhibitor with respect to the substrate ATP. A value of 16.4 microM was obtained for the apparent Ki. Concentration of 0.8 microM euchrestaflavanone B caused 50% growth inhibition of human cancer cells U937 and HeLa. Euchrestaflavanone B-induced cell death was characterized with the cleavage of poly(ADP-ribose) polymerase and procaspase-3, indicating that the inhibitor triggered apoptosis. Because protein kinase CKII is involved in cell proliferation and oncogenesis, these results suggest that euchrestaflavanone B may function by inhibiting oncogenic disease, at least in part, through the inhibition of CKII activity.</P>

      • SMV-G6H Determines DNA Fragment Size Containing Rsv4 Locus for MAS in Soybean Breeding

        We previously reported that reverse transcription-polymerase chain reaction/restriction fragment length polymorphism (RT- PCR/RFLP) was an effective method to identify SMV strains. Using this method a new SMV strain G6H was successfully identified. To introduce resistance locus Rsv4 of V94-5152, we made crosses between two parents, Hwanggumkong and V94-5152, and obtained 6 BC3 F3 progeny lines, which have different size of DNA fragment of Rsv4 locus region. To confirm the virus resistance of progeny lines, artificial inoculation were conducted with 10 SMV strains, G1-G7, G7A, G6H, and G7H. Genomic DNA of tested lines was extracted and used marker genotyping using 9 SSR marker, which covered about 20 cM genetic distance including Rsv4 locus. In the virulence test, only two progeny lines showed resistance to all the SMV strains like a V94-5152. However, the other lines showed necrotic symptoms to G6H strain. It is considered that a minor gene is located near the Rsv4 locus between Satt157 and Sat_254 marker which interacts with G6H. A new strain can be a clue to find a minor gene in the SMV resistance soybean breeding.

        Assessment of microbial safety and quality changes of brown and white cooked rice treated with atmospheric pressure plasma

        The microbial and physicochemical properties of brown and white cooked rice treated by atmospheric pressure plasma (APP). APP was produced (250 W, 15 kHz, ambient air) and applied to brown and white cooked rice for 5, 10, and 20 min. The 20-min plasma treatment reduced in bacterial counts by 2.01 log CFU/g when cooked rice were inoculated with Bacillus cereus or Escherichia coli O157:H7. The pH of the brown cooked rice was decreased by the 5-min plasma. The hardness values of APP-treated brown and white cooked rice were lower than untreated samples. The reducing sugar contents of brown and white cooked rice were significantly higher than those in untreated samples. Lipid oxidation of APP-treated brown and white cooked rice were higher compared to untreated samples. These results indicate that APP improves microbial quality, although further studies should be conducted to change the physicochemical qualities of brown and white cooked rice induced by APP.

        Thermal shock behaviors of TiN coatings on Inconel 617 and Silicon wafer substrates with finite element analysis method

        The degradation behaviors of TiN coating layers under thermo-mechanical stress were investigated in terms of comparison of finite element analysis (FEA) and experimental data. The coating specimen was designed to quarter cylinder model, and the pulsed laser ablation was assumed as heat flux condition. The FEA results showed that heat accumulation at the center of the laser-ablated spot occurred and principle stress was concentrated at the lower region of the coating layer. The microstructural observation revealed that surface melting and decrease of the coating thickness occurred in the TiN/Inconel 617 and the interfacial cracks formed in the TiN/Si. The delamination was caused by the mechanical stress from the center to the outside of the ablated spot as the FEA results expected. It was considered that the improvement of the thermal shock resistance was attributed to higher thermal conductivity of Si wafer than that of Inconel 617.
